We explore the possible cellular cytotoxic activity of an amphiphilic silicon(IV) phthalocyanine with axially ligated rhodamine B under ambient light experimental environment as well as its in vivo antitumour potential using Hep3B hepatoma cell model. After loading into the Hep3B hepatoma cells, induction of cellular cytotoxicity and cell cycle arrest were detected. Strong growth inhibition of tumour xenograft together with significant tumour necrosis and limited toxicological effects exerted on the nude mice could be identified. 相似文献

Starting from a orexin 1 receptor selective antagonist 4,4-disubstituted piperidine series a novel potent 5-azaspiro[2.4]heptane dual orexin 1 and orexin 2 receptor antagonist class has been discovered. SAR and Pharmacokinetic optimization of this series is herein disclosed. In order to preserve environmental quality, alternative strategies to chemical-intensive agriculture are strongly needed. In this study, we characterized in vitro the potential plant growth promoting (PGP) properties of a gamma-proteobacterium, named MIMR1, originally isolated from apple shoots in micropropagation. The analysis of the 16S rRNA gene sequence allowed the taxonomic identification of MIMR1 as Luteibacter rhizovicinus. The PGP properties of MIMR1 were compared to Pseudomonas chlororaphis subsp. aurantiaca DSM 19603T, which was selected as a reference PGP bacterium. By means of in vitro experiments, we showed that L. rhizovicinus MIMR1 and P. chlororaphis DSM 19603T have the ability to produce molecules able to chelate ferric ions and solubilize monocalcium phosphate. On the contrary, both strains were apparently unable to solubilize tricalcium phosphate. Furthermore, the ability to produce 3-indol acetic acid by MIMR1 was approximately three times higher than that of DSM 19603T. By using fluorescent recombinants of strains MIMR1 and DSM 19603T, we also demonstrated that both bacteria are able to abundantly proliferate and colonize the barley rhizosphere, preferentially localizing on root tips and in the rhizoplane. Finally, we observed a negative effect of DSM 19603T on barley seed germination and plant growth, whereas MIMR1, compared to the control, determined a significant increase of the weight of aerial part (+22 %), and the weight and length of roots (+53 and +32 %, respectively). The results obtained in this work make L. rhizovicinus MIMR1 a good candidate for possible use in the formulation of bio-fertilizers. 相似文献

Trypanosoma cruzi infection leads to development of a chronic disease but the mechanisms that the parasite utilizes to establish a persistent infection despite activation of a potent immune response by the host are currently unknown. Unusual characteristics of T. cruzi are that it possesses cellular levels of pyrophosphate (PPi) at least 10 times higher than those of ATP and molar levels of inorganic polyphosphate (polyP) within acidocalcisomes. We characterized an inorganic soluble EF‐hand containing pyrophosphatase from T. cruzi (TcVSP) that, depending on the pH and cofactors, can hydrolyse either pyrophosphate (PPi) or polyphosphate (polyP). The enzyme is localized to both acidocalcisomes and cytosol. Overexpression of TcVSP (TcVSP‐OE) resulted in a significant decrease in cytosolic PPi, and short and long‐chain polyP levels. Additionally, the TcVSP‐OE parasites showed a significant growth defect in fibroblasts, less responsiveness to hyperosmotic stress, and reduced persistence in tissues of mice, suggesting that PPi and polyP are essential for the parasite to resist the stressful conditions in the host and to maintain a persistent infection. 相似文献

Speciation may be influenced by geographic variation in animal signals, particularly when those signals are important in reproductive decisions. Here, we describe patterns of geographic variation in the song of rufous‐naped wrens Campylorhynchus rufinucha. This species complex is a morphologically variable taxon confined to tropical dry forest areas from Mexico to northwestern Costa Rica. Morphological and genetic analyses suggest that there are at least three partially isolated groups within the complex, including a secondary‐contact zone in coastal western Chiapas between the subspecies C. r. humilus and C. r. nigricaudatus. Based on recordings throughout their geographic range, we investigate the effects of historical isolation on song structure and analyze whether genetic differences or climatic conditions explain observed patterns of variation. Our findings, based on a culturally‐transmitted and sexually‐selected trait, support the hypothesis that three evolutionary units exist within this taxon. Our results suggest that song differences between genetic groups were influenced by historical isolation. We report a strong relationship between vocal dissimilarity and genetic distance, suggesting that differences in vocal characteristics are probably affected by the same factors that drive genetic divergence. We argue that the evolution of song in this taxon is influenced by vicariant events, followed by accumulation of changes in song structure due to several possible factors: cultural drift in song structure; genetic drift in features related to song production; or natural selection acting on features that influence songs, such as body and beak size. 相似文献

Colonization of newly created habitats is a challenge for waterbird populations in a changing world. Knowing which habitat characteristics are required by waterbird populations is a research challenge for rational management of the new ecosystems and their aquatic bird populations. Since 1989 intertidal dredge islands have been built in the lagoon of Venice using sediments coming from regular dredging of lagoon channels and inlets. Kentish Plover, a species declining in Europe, readily uses these new sites as soon as they become available. Between 2005 and 2007, 75 dredge islands were surveyed each year and the number of breeding pairs of Kentish Plover estimated. Each year about one-third of available dredge islands was used by Kentish Plover. Between 34 (in 2005) and 131 (in 2007) breeding pairs were found, and possible differences in vegetation and morphological characteristics between occupied and unoccupied sites were investigated. Only age, mean elevation above sea level and extension of bare ground were statistically different; Kentish Plover preferred younger sites, with higher elevation and with larger areas of bare ground. The largest groups of breeding pairs, up to thirty pairs, were found on islands which also supported colonies of Little Terns. In the study period dredged islands supported about 60 % of the total breeding population of the lagoon of Venice and 4–6 % of the estimated Italian population. Along coastal sites where human pressure on beaches is particularly heavy, man made habitats such as dredge islands may become a valuable alternative breeding site for this and other species of conservation concern. Management works aimed at promoting the occurrence of this species at selected dredge islands have been made in the lagoon of Venice. 相似文献

Three isolates of Histoplasma capsulatum were identified from mice lung, liver, and spleen inoculated with soil samples of the X hotel’s ornamental potted plants that had been fertilized with organic material known as compost. The presence of H. capsulatum in the original compost was detected using the dot-enzyme-linked immunosorbent assay. Nested-PCR, using a specific protein Hcp100 coding gene sequence, confirmed the fungal identification associated with an unusual histoplasmosis outbreak in Acapulco. Although, diversity between the H. capsulatum isolate from the hotel and some clinical isolates from Guerrero (positive controls) was observed using random amplification of polymorphic DNA based-PCR, sequence analyses of H-anti and ole fragment genes revealed a high homology (92–99%) between them. 相似文献

The relationship between the circadian and homeostatic control of body temperature was studied in golden hamsters maintained under a 14:10 LD cycle. Telemetric records of body temperature showed that body temperature oscillates daily with a low phase during the light section of the LD cycle and a high phase during the dark section. The low phase of the temperature rhythm was found to start two hours after lights on and to last about 8 hours. The high phase was found to start immediately after lights off and to last about 8 hours also. Metabolic heat production was measured by indirect calorimetry during the high phase and the low phase of the body temperature rhythm. Heat production in a thermoneutral environment was higher during the high phase of the body temperature rhythm than during the low phase, but cold-induced thermogenesis was greater during the low phase than during the high phase. This finding suggests that the autonomic thermoregulatory system is more responsive to cold stress during the low phase than during the high phase. Consequently, the daily oscillation of body temperature cannot be explained by an elevation of the thermoregulatory set point during the high phase of the rhythm. The homeostatic and circadian control of body temperature seem to be exerted separately from each other. 相似文献
