Summary The morphological development ofClaviceps paspali immobilized in Ca-alginate gel was examined. During consecutive reincubations, the immobilized mycelia differentiated into swollen, arthrosporoid-like cells, which never appeared during fermentation of free mycelium. Summary Kinetic parameters of production of clavine alkaloids were evaluated in twoClaviceps purpurea strains. Mutagenesis brought about enhanced resistance of the biosynthetic system towards alkaloids. Addition of glucose into the fermentation medium altered the zero order kinetics of production to activation-inhibition kinetics. Summary The effect was studied of the method of drying chymotrypsin attached to bead cellulose on its chemical and physical characteristics. These characteristics are not deteriorated when replacing the commonly used lyophilisation by fluid drying in the air stream. The preparations saved essentially the same proteolytic activity as well as stability even when increasing the drying air temperature to 70°C.  相似文献

Our objective was to examine the developmental fate of sperm nuclei in oocytes fertilized under conditions of meiotic arrest. Therefore zona-free metaphase II oocytes and oocyte fragments (nucleate and anucleate) were fertilized in the presence of colcemid. In anucleate oocyte fragments, normal male pronuclei develop. In contrast, in intact oocytes and nucleate fragments sperm nuclei after initial decondensation undergo secondary condensation. This state is maintained as long as the oocytes are treated with colcemid. When the drug is removed 3 h after insemination, the meiotic spindle(s) is reconstructed, the second polar body(ies) is extruded, and a female pronucleus (or micronuclei) forms. At the same time the sperm nucleus decondenses again and transforms into a male pronucleus. In addition oocytes fertilized in the presence of colcemid could not be refertilized. These observations suggest that oocytes and oocyte fragments fertilized in the presence of colcemid undergo activation despite the failure of pronucleus formation. The inhibitory effect of colcemid on the formation of pronuclei is expressed only in the presence of oocyte chromosomes. We suggest that colcemid stabilizes factors responsible for chromosome condensation that are associated with oocyte chromosomes but not factors (whether the same or different) present in the cytoplasm.  相似文献

An analytical review of recent publications of home and foreign authors on the problem of laboratory diagnosis of campylobacteriosis is presented. The commercial nutrient media, methods of creation of the microaerophilic conditions for cultivation of campylobacter are presented. The filtration method is preferable for isolation of these agents from the studied material highly contaminated by accompanying microflora. A special attention is paid to immunodiagnosis of campylobacteriosis: agglutination reaction, coagglutination reaction, passive hemagglutination reaction, immunoenzyme and radioimmune analyses. Seroepidemiological examination of the staff at one of meat-packing factories in the Republic carried out by the method of indirect immunoenzyme analysis has revealed high levels of anticampylobacteriosis antibodies in 17.9% of examinees. The promising trends in perfection of the methods for laboratory diagnosis of campylobacteriosis are outlined.  相似文献

Three-dimensionally preserved and chemically prepared skulls and natural casts of representatives of the families Benthosuchidae, Melosauridae, and Capitosauridae yield data on the structure of the ethmoidal endocranium, i. e. of those nasal cranial structures that consisted originally of cartilage. This study demonstrates that the ethmoidal endocranium was principally a dorsoventrally compressed plate, pierced by a broad and oblique canal which communicated anteriorly with the outer dorsal surface by the fenestra endonarina and posteriorly with the mouth cavity by the fenestra endochoanalis(seu foris). The canal was very short, and housed the olfactory organ. The ethmoidal endocranium was connected with the palatoquadrate by the commissura quadratocranialis anterior; there was no lateral ethmoidal commissure, however, in older individuals the anterior section of the palatoquadrate might also contact the postchoanal part of the nasal endocranial skeleton.  相似文献

Conidia of the speciesFusarium culmorum /W.G.Sm./ Sacc. andFusarium graminearum Schwabe are characterized by variability in zearalenone production and dimensions depending on the substrate. The sporulation of isolates from some wheat eultivars have been deprived in vivo and in vitro in the first passage, but not their pathogenicity and toxic metabolites production. Nonsporulating strains produced lower quantités of zearalenone than sporulating ones. Liquid filtrates of such nonsporulating strains had a high phytotoxic effect on wheat caryopses. The crystalline toxin DAS /0,25 ug/ml/ had low phytotoxic effect on wheat caryopses.  相似文献

The neural network that efficiently and nearly optimally solves difficult optimization problems is defined. The convergence proof for the Markovian neural network that asynchronously updates its neurons' states is also presented. The comparison of the performance of the Markovian neural network with various combinatorial optimization methods in two domains is described. The Markovian neural network is shown to be an efficient tool for solving optimization problems.  相似文献

Streptomycin and N-methyl-N’-nitro-N-nitrosoguanidine (MNNG) cause a 100% hereditary bleaching ofEuglena gracilis. In spite of the outcomes being the same, the corresponding mechanisms differ. By investigating the incorporation of the14C-labelled precursors of macromolecule synthesis we showed streptomycin to inhibit the synthesis of nucleic acids and proteins to the same extent (56%), whereas in the case of MNNG a 66% inhibition of nucleic acid synthesis and a 24% one of protein synthesis were observed.  相似文献
