To determine whether or not the occurrence of sporadic amyotrophic lateral sclerosis (sALS) is associated with both excess nitric oxide (NO) metabolites and decreased protective superoxide dismutase (SOD) activity in the cerebrospinal fluid (CSF), we measured nitrate concentration and SOD activity in the CSF of sALS patients and in age- and gender-matched controls. We found stable NO metabolite levels to be significantly higher and SOD activity lower in the CSF of sALS patients. In addition, SOD showed a negative correlation with motor neuron axonal damage expressed as the amplitude of motor action potentials in upper limbs. Our results provide new evidence in vivo suggesting that NO products and SOD activity play a role in oxidant/antioxidant imbalance in sporadic ALS.  相似文献

Postsynaptic density-95/disks large/zonula occludens-1 (PDZ) domains are relatively small (80-120 residues) protein binding modules central in the organization of receptor clusters and in the association of cellular proteins. Their main function is to bind C-terminals of selected proteins that are recognized through specific amino acids in their carboxyl end. Binding is associated with a deformation of the PDZ native structure and is responsible for dynamical changes in regions not in direct contact with the target. We investigate how this deformation is related to the harmonic dynamics of the PDZ structure and show that one low-frequency collective normal mode, characterized by the concerted movements of different secondary structures, is involved in the binding process. Our results suggest that even minimal structural changes are responsible for communication between distant regions of the protein, in agreement with recent NMR experiments. Thus, PDZ domains are a very clear example of how collective normal modes are able to characterize the relation between function and dynamics of proteins, and to provide indications on the precursors of binding/unbinding events.  相似文献

Here we report on a lipid-signalling pathway in plants that is downstream of phosphatidic acid and involves the Arabidopsis protein kinase, AGC2-1, regulated by the 3'-phosphoinositide-dependent kinase-1 (AtPDK1). AGC2-1 specifically interacts with AtPDK1 through a conserved C-terminal hydrophobic motif that leads to its phosphorylation and activation, whereas inhibition of AtPDK1 expression by RNA interference abolishes AGC2-1 activity. Phosphatidic acid specifically binds to AtPDK1 and stimulates AGC2-1 in an AtPDK1-dependent manner. AtPDK1 is ubiquitously expressed in all plant tissues, whereas expression of AGC2-1 is abundant in fast-growing organs and dividing cells, and activated during re-entry of cells into the cell cycle after sugar starvation-induced G1-phase arrest. Plant hormones, auxin and cytokinin, synergistically activate the AtPDK1-regulated AGC2-1 kinase, indicative of a role in growth and cell division. Cellular localisation of GFP-AGC2-1 fusion protein is highly dynamic in root hairs and at some stages confined to root hair tips and to nuclei. The agc2-1 knockout mutation results in a reduction of root hair length, suggesting a role for AGC2-1 in root hair growth and development.  相似文献

The effect of a single blood meal on the host-seeking response of Anopheles gambiae was investigated in the laboratory using a behavioural bioassay, whereas possible changes at the chemosensory level were monitored using electroantennogram recording (EAG). To avoid the possible confounding effect of body size, mosquitoes of a large size class only were used. Five-day old female mosquitoes were given a blood meal on a human arm and exposed to the emanations of a human hand in an olfactometer at 3, 24, 40, 48 and 72 h following the meal and their behaviour and EAG response to host stimuli were compared with that of unfed mosquitoes (controls) of corresponding age. During egg development, mosquitoes had access to glucose and an oviposition tray. The ovarian development of blood-fed mosquitoes that responded to host odours was compared with that of blood-fed mosquitoes that had not been exposed to host odours. The EAG response of blood-fed and control mosquitoes to host odour was examined upon stimulation with air led over incubated human sweat, hexanoic acid, indole and geranyl acetone. EAGs were recorded at times after a blood meal corresponding with those used in the behavioural experiment. There was no host-seeking response at 3 and 24 h post blood meal (pbm). Seven percent of the mosquitoes responded to human emanations 40-h pbm, 27% at 48 h and 68% at 72 h following a blood meal. The average response of controls to host stimuli varied from 35 (at t=40 h) to 67%. There was no ovarian development in the unfed group of mosquitoes. Of the mosquitoes that responded to host odour 48 h pbm, 12.5% (n=5) had ovaries in Christophers' stage IV and the remainder in stage V. Of the mosquitoes that responded 72 h pbm, 66.7% (n=94) had ovaries in stage V and 31.2% (n=44) had recently oviposited. Maximum EAG amplitudes recorded from blood-fed and control mosquitoes were similar for mosquitoes in Christophers' stages I-III, whereas in stage IV EAG amplitudes recorded from the blood-fed group were significantly lower than those of the corresponding control group in response to headspace of incubated human sweat and to indole. The results show that there was a strong inhibition of host seeking in An. gambiae for a period of at least 40 h following a blood meal. Host-seeking returned to pre-blood meal levels 72-h post feeding and was associated with egg maturation. The inhibition of host-seeking behaviour was accompanied by an inhibition of olfactory sensitivity to headspace of incubated sweat and indole just before the resumption of the host-seeking response. The implications of these findings for mosquito surveillance with host odours are discussed.  相似文献

In this article we present a series of non-cytotoxic potent human choline kinase (CK) inhibitors that exhibit nanomolar antiplasmodial activity in vitro. The most active antiplasmodial compounds, 10ab, bearing a pyridinium cationic head were inactive against CK, while compounds 10g and 10j with a quinolinium moiety exhibit moderate inhibition of both the parasite and the enzyme. The results point towards an additional mechanism of action unrelated to CK inhibition that remains to be established.  相似文献

Novozyme 435, which is a commercial immobilized lipase B from Candida antarctica (CALB), has been proven to be inadequate for the kinetic resolution of rac‐indanyl acetate. As it has been previously described that different immobilization protocols may greatly alter lipase features, in this work, CALB was covalently immobilized on epoxy Immobead‐350 (IB‐350) and on glyoxyl‐agarose to ascertain if better kinetic resolution would result. Afterwards, all CALB biocatalysts were utilized in the hydrolytic resolution of rac‐indanyl acetate and rac‐(chloromethyl)‐2‐(o‐methoxyphenoxy) ethyl acetate. After optimization of the immobilization protocol on IB‐350, its loading capacity was 150 mg protein/g dried support. Furthermore, the CALB‐IB‐350 thermal and solvent stabilities were higher than that of the soluble enzyme (e.g., by a 14‐fold factor at pH 5–70°C and by a 11‐fold factor in dioxane 30%–65°C) and that of the glyoxyl‐agarose‐CALB (e.g., by a 12‐fold factor at pH 10–50°C and by a 21‐fold factor in dioxane 30%–65°C). The CALB‐IB‐350 preparation (with 98% immobilization yield and activity versus p‐nitrophenyl butyrate of 6.26 ± 0.2 U/g) was used in the hydrolysis of rac‐indanyl acetate using a biocatalyst/substrate ratio of 2:1 and a pH value of 7.0 at 30°C for 24 h. The conversion obtained was 48% and the enantiomeric excess of the product (e.e.p) was 97%. These values were much higher than the ones obtained with Novozyme 435, 13% and 26% of conversion and e.e.p, respectively. © 2018 American Institute of Chemical Engineers Biotechnol. In herbivorous insects, the interaction between adult preference and progeny performance on specific host plants is modified by maternal feeding experience and host plant quality. Ultimately, changes in the strength of this interaction can affect insect population dynamics. In this study, we hypothesized that adult host plant preference influences progeny performance through a maternal feeding experience × host plant interaction, that is, the effect of adult feeding experience on progeny performance will depend on the host plant. Second, that decoupling of the preference–performance relationship due to host switching results in different population vital rates changing population dynamics. An increase in development time and a decrease in body size of individuals in the alternate host should decrease population growth. We tested these hypotheses using two lines of the tortoise beetle Chelymorpha varians Blanchard fed with two hosts (Convolvulus arvensis and Calystegia sepium). Maternal feeding experience treatments were crossed with host plant species, and the offspring’s developing time and adult size were measured. The host plant influence on the beetle’s population vital rates was tested using stage-structured matrix population models and life table response experiments. Host plant preference affected offspring body size through a host plant effect that contributed to adaptive life history responses only in the better quality host. C. varians’ population growth was positive when fed with either host; comparatively, however, C. sepium had a negative effect on growth by reducing all transition probabilities of the life cycle stages of the beetle. Here, we show that individuals of C. varians prefer and perform differently on distinct hosts and that these patterns influence population vital rates in different ways. When beetles prefer the host plant where their progeny performs best, life history responses and life stage transitions lead to higher population growth; otherwise, growth rate decreases.  相似文献
