The membrane glycoprotein enzyme, alkaline phosphatase was induced in cultured human fibroblasts by dibutyryl cyclic AMP, sodium butyrate, the serum glycoprotein fetuin, the Tamm-Horsfall urinary glycoprotein, and by a number of inhibitors of DNA synthesis. The uninduced basal enzyme activity increased at later stages of growth when the cells became confluent. Induction by dibutyryl cyclic AMP or fetuin was most effective when the agents were added after the cells had reached stationary phase and was maximal after at least two days of exposure. The levels of induction resulting from the addition of pairs of the agents, dibutyryl cyclic AMP, n-butyrate and fetuin were additive indicating that these have different modes of action. The inhibitors of DNA synthesis, cytosine arabinoside, hydroxyurea, and methothrexate were less effective inducers. Bromodeoxyuridine which also has non-DNA mediated effects induced to the same extent as dibutyryl cyclic AMP.Similar experiments with sex- and age-matched cell strains derived from patients with cystic fibrosis failed to detect differences in the levels of induction from those observed in normal cells. In addition, the combined inductive effects of Tamm-Horsfall glycoprotein, isoproterenol and theophylline, were similar with normal and cystic fibrosis cells. 相似文献

In solution, nitrocarboxypeptidase A, modified at tyrosyl-248, exhibits a nitrotyrosyl pK apparent of 6.3. In the crystalline state, the pK apparent is about 8.2. The separation of animals into age classes is often fraught with difficulties, but is essential for the interpretation of survival rates and population structure. This study investigated both live hedgehogs and road kill in order to develop techniques that could be utilised to separate age classes in the field. Body measurements were taken from 24 live hedgehogs caught at a site in County Cork, Ireland over a three year period. Additionally, carcasses from around Ireland were collected and measured. There was a significant correlation between the weight and hind foot length of both live and dead hedgehogs. The weight of individuals was also significantly correlated with the body length and jaw length of hedgehogs. In all of these correlations, the sample clustered clearly into two groups with ‘juveniles’ meeting all of the following criteria: a hind foot length of <3.6 cm, a body length of <?16 cm and a jaw length of below 4.5 cm. It is suggested that these parameters, combined with the presence of growing spines, could provide a way of separating age classes in the field. This would be of particular benefit to short term studies, allowing the population structure to be accurately assessed and the reproductive success and sustainability of a population to be investigated. Road kill were also aged using the dentary bone in order to investigate the age profiles of hedgehogs killed on the road. Amongst those killed, 54 % of hedgehogs were 1-year-old or less. The majority of hedgehogs (87 %) were between 0–3-years-old, the oldest females were nine (n?=?2), and the oldest males were eight years old (n?=?2), respectively. Long‐term population studies on large mammals are rare. Here, we have examined the threatened scimitar‐horned oryx, addax and dama gazelle's populations over the last 20 years in Bou Hedma National Park. Using monthly count data of the three studied species collected since 1995, we examined their population trends. Using autocorrelation analyses, we discovered endogenous natural cyclical fluctuations in the numbers of each species, with a periodicity of approximately 3 years. For all three studied species which seem to be opportunistic breeders, births and deaths occurred throughout the year, although with notable seasonality. By means of cross‐correlation, we discovered that during the first 7 years for which data were available, addax numbers were positively correlated with those of dama and inversely correlated with numbers of oryx. This pattern reversed during the following 4‐year period. The number of oryx was negatively correlated with dama during the first 4 years and then became positively correlated during the subsequent 7‐year period. Thus, we draw attention to difference in response to environmental and anthropogenic factors. Incorporating fundamental long‐term population data into developing management approaches, especially for potentially competitive species, is vital for their future long‐term survival and the success of conservation actions. 相似文献

Odonate populations and species numbers are declining globally. Successful conservation requires sound assessments of both
odonate distributions and habitat requirements. Odonates have aquatic (larval) and terrestrial (adult) stages, but most surveys
that are used to inform conservation managers are undertaken of the adult stage. This study investigates whether this bias
towards adult records in odonate recording is misinterpreting the environmental quality of sites. The habitat focus is farmland
ponds, a key feature of agricultural landscapes. We tested whether or not, adult, larval and exuvial surveys lead to similar
conclusions on species richness and hence on pond quality. Results showed that pond surveys based upon larvae and exuviae
are equally suitable for the reliable assessment of presence/absence of odonates, but that adult surveys are not interchangeable
with surveys of larvae/exuviae. Larvae were also found at ponds with no emerging individuals due to changes in habitat quality,
therefore presence of exuviae remains the only proof of life-cycle completion at a site. Ovipositing females were recorded
at all ponds where exuviae were totally absent hence adult surveys over-estimate pond quality and low-quality ponds are functioning
as ecological traps. Highly mobile and generalist species were recorded at more locations than other species. Adult surveys
also bias recording towards genera, species and populations with non-territorial mate-location strategies. Odonate biodiversity
monitoring would benefit from applying the best survey method (exuviae) to avoid wasting valuable financial resources while
providing unbiased data, necessary to achieve conservation objectives. 相似文献

To determine the incidence of secondary meningococcal infection in close family and household contacts of index patients and to review the efficacy of chemoprophylaxis the records of 3256 cases occurring from 1984 through 1987 were examined. Seventeen secondary cases (0.5%) of infection were identified among these groups. The median interval between index and secondary cases was seven weeks. Fourteen secondary cases occurred more than one week after the disease was diagnosed in the index case. Three secondary cases had not received chemoprophylaxis and in another case the infecting strain had acquired resistance to rifampicin. Prophylaxis for the close contacts of 10 out of 11 of the remaining index patients failed to fulfil all the criteria of an optimal regimen. Even after optimal chemoprophylaxis the medical practitioner and the family should be aware of the increased and prolonged risk of secondary meningococcal infection among close contacts of patients with the disease. 相似文献

Summary The functions of a number of amino acid residues in proteins have been studied by chemical modification techniques and much useful information has been obtained. Methods using dicarbonyl compounds for the modification of arginine residues are the most recent to have been developed. Since their introduction about 10 years ago, they have led to the identification of a large number of enzymes and other proteins that contain arginine residues critical to biological function. These reagents are discussed in terms of their chemical reactivity and mechanisms of action and in relation to the unique chemical properties of the guanidinium group. Butanedione, phenylglyoxal and cyclohexanedione are the most commonly employed arginyl reagents, and their relative advantages are examined. A survey of the functional role of arginine residues in enzymes and other proteins is presented in which nearly 100 examples are cited. The prediction that arginine residues would be found to serve a general role as anionic binding sites in protein has obviously been validated.
