Can three incongruence tests predict when data should be combined? 总被引:17,自引:14,他引:17
Advocates of conditional combination have argued that testing for
incongruence between data partitions is an important step in data
exploration. Unless the partitions have had distinct histories, as in
horizontal gene transfer, incongruence means that one or more data support
the wrong phylogeny. This study examines the relationship between
incongruence and phylogenetic accuracy using three tests of incongruence.
These tests were applied to pairs of mitochondrial DNA data partitions from
two well-corroborated vertebrate phylogenies. Of the three tests, the most
useful was the incongruence length difference test (ILD, also called the
partition homogeneity test). This test distinguished between cases in which
combining the data generally improved phylogenetic accuracy (P > 0.01)
and cases in which accuracy of the combined data suffered relative to the
individual partitions (P < 0.001). In contrast, in several cases, the
Templeton and Rodrigo tests detected highly significant incongruence (P
< 0.001) even though combining the incongruent partitions actually
increased phylogenetic accuracy. All three tests identified cases in which
improving the reconstruction model would improve the phylogenetic accuracy
of the individual partitions.

We introduce a new approach to detect individual microparticles that contain NIR fluorescent dye by multispectral optoacoustic tomography in the context of the hemoglobin-rich environment within murine liver. We encapsulated a near infrared (NIR) fluorescent dye within polystyrene microspheres, then injected them into the ileocolic vein, which drains to the liver. NIR absorption was determined using multispectral optoacoustic tomography. To quantitate the minimum diameter of microspheres, we used both colorimetric and spatial information to segment the regions in which the microspheres appear. Regional diameter was estimated by doubling the maximum regional distance. We found that the minimum microsphere size threshold for detection by multispectral optoacoustic tomography images is 78.9 µm. 相似文献

The identification of a novel metastasis suppressor function for the MAP Kinase Kinase 4 protein established a role for the stress-activated kinases in regulating the growth of disseminated cancer cells. In this review, we describe MKK4's biological mechanism of action and how this information is being used to guide the development of new models to study cancer cell dormancy and metastatic colonization. Specifically, we describe the novel application of microvolume structures, which can be modified to represent characteristics similar to those that cancer cells experience at metastatic sites. Although MKK4 is currently one of many known metastasis suppressors, this field of research started with a single daring hypothesis, which revolutionized our understanding of metastasis, and opened up new areas of exploration for basic research. The combination of our increasing knowledge of metastasis suppressors and such novel technologies provide hope for possible clinical interventions to prevent suffering from the burden of metastatic disease. 相似文献

Background
Relaxin is the endogenous ligand of the G-protein coupled receptor RXFP1, previously known as LGR7. In humans relaxin can also activate, but with lower affinity, the closely related receptor for the insulin-like peptide from Leydig cells, RXFP2, previously known as LGR8. The lack of relaxin impairs male fertility but the precise distribution and the function of relaxin receptors in the male reproductive tract is not known. HeLa cells were transformed to express antisense RNA against initiation factor eIF-4E mRNA from an inducible promoter. In the absence of inducer, these cells (AS cells) were morphologically similar to control cells but grew four- to sevenfold more slowly. Induction of antisense RNA production was lethal. Both eIF-4E mRNA and protein levels were reduced in proportion to the degree of antisense RNA expression, as were the rates of protein synthesis in vivo and in vitro. Polysomes were disaggregated with a concomitant increase in ribosomal subunits. Translation in vitro was restored by addition of the initiation factor complex eIF-4F but not by eIF-4E alone. Immunological analysis revealed that the p220 component of eIF-4F was decreased in extracts of AS cells and undetectable in AS cells treated with inducer, suggesting that p220 and eIF-4E levels are coordinately regulated. eIF-4A, another component of eIF-4F, was unaltered. 相似文献

A cDNA molecule encoding a major part of the human Norepinephrine transporter(hNET) was synthesized by means of Polymerase Chain Reaction(PCR) technique and used as a probe for selecting the human genomic NET gene.A positive clone harbouring the whole gene was obtained from a human lymphocyte genomic library through utilizing the “genomic walking” technique.The clone,designated as phNET,harbours a DNA fragment of about 59 kd in length inserted into BamH I site in cosmid pWE15.The genomic clone contains 14 exons encoding all amino acid residues in the protein.A single exon encodes a distinct transmembrane domain,except for transmembrane domain 10 and 11,which are encoded by part of two exons respectively,and exon 12,which encodes part of domain 11 and all of domain 12.These results imply that there is a close relationship between exon splicing of a gene and structureal domains of the protein,as is the case for the human γ-aminobutyric acid transporter(hGAT) and a number of other membrane proteins. 相似文献

The effect of relaxed functional constraints on the photosynthetic gene rbcL in photosynthetic and nonphotosynthetic parasitic plants 总被引:1,自引:0,他引:1
The photosynthetic gene rbcL has been lost or dramatically altered in some
lineages of nonphotosynthetic parasitic plants, but the dynamics of these
events following loss of photosynthesis and whether rbcL has sustained
functionally significant changes in photosynthetic parasitic plants are
unknown. To assess the changes to rbcL associated with the loss of
functional constraints for photosynthesis, nucleotide sequences from
nonparasitic and parasitic plants of Scrophulariales were used for
phylogeny reconstruction and character analysis. Plants in this group
display a broad range of parasitic abilities, from photosynthetic
("hemiparasites") to nonphotosynthetic ("holoparasites"). With the
exception of Conopholis (Orobanchaceae), the rbcL locus is present in all
parasitic plants of Scrophulariales examined. Several holoparasitic genera
included in this study, including Boschniakia, Epifagus, Orobanche, and
Hyobanche, have rbcL pseudogenes. However, the holoparasites Alectra
orobanchoides, Harveya capensis, Harveya purpurea, Lathraea clandestina,
Orobanche corymbosa, O. fasciculata, and Striga gesnerioides have intact
open reading frames (ORFs) for the rbcL gene. Phylogenetic hypotheses based
on rbcL are largely in agreement with those based on sequences of the
nonphotosynthetic genes rps2 and matK and show a single origin of
parasitism, and loss of photosynthesis and pseudogene formation have been
independently derived several times in Scrophulariales. The mutations in
rbcL in nonparasitic and hemiparasitic plants would result in largely
conservative amino acid substitutions, supporting the hypothesis that
functional proteins can experience only a limited range of changes, even in
minimally photosynthetic plants. In contrast, ORFs in some holoparasites
had many previously unobserved missense substitutions at functionally
important amino acid residues, suggesting that rbcL genes in these plants
have evolved under relaxed or altered functional constraints.

Background
The most common pesticide products for controlling malaria-transmitting mosquitoes combine two distinct modes of action: 1) conventional insecticidal activity which kills mosquitoes exposed to the pesticide and 2) deterrence of mosquitoes away from protected humans. While deterrence enhances personal or household protection of long-lasting insecticidal nets and indoor residual sprays, it may also attenuate or even reverse communal protection if it diverts mosquitoes to non-users rather than killing them outright.Methods
A process-explicit model of malaria transmission is described which captures the sequential interaction between deterrent and toxic actions of vector control pesticides and accounts for the distinctive impacts of toxic activities which kill mosquitoes before or after they have fed upon the occupant of a covered house or sleeping space.Results
Increasing deterrency increases personal protection but consistently reduces communal protection because deterrent sub-lethal exposure inevitably reduces the proportion subsequently exposed to higher lethal doses. If the high coverage targets of the World Health Organization are achieved, purely toxic products with no deterrence are predicted to generally provide superior protection to non-users and even users, especially where vectors feed exclusively on humans and a substantial amount of transmission occurs outdoors. Remarkably, this is even the case if that product confers no personal protection and only kills mosquitoes after they have fed.Conclusions
Products with purely mosquito-toxic profiles may, therefore, be preferable for programmes with universal coverage targets, rather than those with equivalent toxicity but which also have higher deterrence. However, if purely mosquito-toxic products confer little personal protection because they do not deter mosquitoes and only kill them after they have fed, then they will require aggressive "catch up" campaigns, with behaviour change communication strategies that emphasize the communal nature of protection, to achieve high coverage rapidly. 相似文献49.
