The production of pharmaceutical proteins in plants is creating a broad spectrum of new high-value traits in traditional crop species. As the production of these recombinant proteins moves from bench to field scale, containment and the presence of unwanted secondary metabolites are significant practical issues. We have developed a hybrid male-sterile low-alkaloid tobacco (MSLA) production platform. Recombinant protein is produced in leaves that are harvested prior to flowering. If considered for direct in vivo mammalian use the low-alkaloid background genotype addresses concerns about nicotine, and male sterility further reduces the risk of gene leakage. We have applied this system to the production of human interleukin-10 (phIL-10), a contra-inflammatory cytokine with potential application in the treatment of inflammatory bowel disease and autoimmune diseases. Transgenic low-alkaloid tobacco lines properly assembled a biologically active phIL-10 homodimer. Hybrids made by crossing a single homozygous high-expressing phIL-10 line with a MSLA female were field tested in a high density production system and harvested after 30 days. Recombinant phIL-10 yields were found to be similar in the hybrids and the homozygous control. MSLA tobacco is a practical, self-contained system for the production of plant recombinant proteins.  相似文献

The Neuregulin‐1 (Nrg1)/ErbB pathway plays multiple, critical roles in early cardiac and nervous system development and has been implicated in both heart and nerve repair processes. However, the early embryonic lethality of mouse Nrg1 mutants precludes an analysis of Nrg1's function in later cardiac development and homeostasis. In this study, we generated a novel nrg1 null allele targeting all known isoforms of nrg1 in zebrafish and examined cardiac structural and functional parameters throughout development. We found that zebrafish nrg1 mutants instead survived until young adult stages when they exhibited reduced survivorship. This coincided with structural and functional defects in the developing juvenile and young adult hearts, as demonstrated by reduced intracardiac myocardial density, cardiomyocyte cell number, swimming performance and dysregulated heartbeat. Interestingly, nrg1 mutant hearts were missing long axons on the ventricle surface by standard length (SL) 5 mm, which preceded juvenile and adult cardiac defects. Given that the autonomic nervous system normally exerts fine control of cardiac output through this nerve plexus, these data suggest that Nrg1 may play a critical role in establishing the cardiac nerve plexus such that inadequate innervation leads to deficits in cardiac maturation, function and survival.  相似文献

The Tipula abdominalis larval hindgut microbial community presumably facilitates digestion of the lignocellulosic diet. The microbial community was investigated through characterization of bacterial isolates and analysis of 16S rRNA gene clone libraries. ABSTRACT

Pseudosuccinea columella and Radix natalensis live in the same habitat in Egypt and are important intermediate hosts of Fasciola hepatica and F. gigantica. Our study aimed to characterise both snail species using molecular analysis and shell measurements. The ranges of morphometric parameters overlapped in the two lymnaeids, indicating that they do not clearly differentiate the two species. PCR-sequence analysis of the nuclear ribosomal small subunit rRNA and the polymorphic mitochondrial cytochrome oxidase subunit 1 (CO1) genes were used to determine the genetic identity and the potential diversity of the snails. Little intrasequence variations were detected in the sequences of both gene loci, indicating the potential homogeneity of lymnaeid populations in Egypt. Generated sequences of the mitochondrial CO1 gene locus for R. natalensis showed obvious heterogeneity compared to other sequences in GenBank. Molecular characterisation of these lymnaeids might help to understand the snails’ biodiversity in a bid to control these populations and their related diseases.  相似文献

Introduction

Statins, widely prescribed drugs for treatment of cardiovascular disease, inhibit the biosynthesis of low density lipoprotein cholesterol (LDL-C). Despite providing major benefits, sub populations of patients experience adverse effects, including muscle myopathy and development of type II diabetes mellitus (T2DM) that may result in premature discontinuation of treatment. There are no reliable biomarkers for predicting clinical side effects in vulnerable individuals. Pharmacometabolomics provides powerful tools for identifying global biochemical changes induced by statin treatment, providing insights about drug mechanism of action, development of side effects and basis of variation of response.

Objective

To determine whether statin-induced changes in intermediary metabolism correlated with statin-induced hyperglycemia and insulin resistance; to identify pre-drug treatment metabolites predictive of post-drug treatment increased diabetic risk.

Methods

Drug-naïve patients were treated with 40 mg/day simvastatin for 6 weeks in the Cholesterol and Pharmacogenetics (CAP) study; metabolomics by gas chromatography-time-of-flight mass-spectrometry (GC–TOF–MS) was performed on plasma pre and post treatment on 148 of the 944 participants.

Results

Six weeks of simvastatin treatment resulted in 6.9% of patients developing hyperglycemia and 25% developing changes consistent with development of pre-diabetes. Altered beta cell function was observed in 53% of patients following simvastatin therapy and insulin resistance was observed in 54% of patients. We identified initial signature of simvastatin-induced insulin resistance, including ethanolamine, hydroxylamine, hydroxycarbamate and isoleucine which, upon further replication and expansion, could be predictive biomarkers of individual susceptibility to simvastatin-induced new onset pre-type II diabetes mellitus. No patients were clinically diagnosed with T2DM.

Conclusion

Within this short 6 weeks study, some patients became hyperglycemic and/or insulin resistant. Diabetic markers were associated with decarboxylated small aminated metabolites as well as a branched chain amino acid directly linked to glucose metabolism and fatty acid biosynthesis. Pharmacometabolomics provides powerful tools for precision medicine by predicting development of drug adverse effects in sub populations of patients. Metabolic profiling prior to start of drug therapy may empower physicians with critical information when prescribing medication and determining prognosis.

Genes of the Major Histocompatibility Complex ( Mhc ) play a fundamental role during the immune response because MHC molecules expressed on cell surface allow the recognition and presentation of antigenic peptides to T-lymphocytes. Although Mhc alleles have been found to correlate with pathogen resistance in several host-parasite systems, several studies have also reported associations between Mhc alleles and an accrued infection risk or an accelerated disease progression. The existence of these susceptibility alleles is puzzling, as the cost generated by the infection should rapidly eliminate them from the population. Here, we show that susceptibility alleles may be maintained in a population of house sparrows ( Passer domesticus ) if they have antagonistic effects on different malaria parasites. We found that one Mhc class I allele was associated with a 2.5-fold increase in the risk to be infected with a Plasmodium strain, but with a 6.4-fold reduction in the risk to harbour a Haemoproteus strain. We suggest that this antagonistic effect might arise because Mhc genes can alter the competitive interactions between malaria parasites within the host.  相似文献
