Workshop on the ecosystem and fisheries of the Coral Sea: an Australian perspective on research and management

This report summarizes a workshop on the Coral Sea to determine key research findings and identify the research gaps needed to support sustainable management of a proposed Coral Sea Marine Reserve. Key research questions included determining the connectivity of apex predators with the broader southwest Pacific Ocean, and assessing the regions?? biodiversity in relation to seabed topography and oceanographic processes. The workshop concluded noting the importance of engaging surrounding countries in maintaining the sustainability and uniqueness of the Coral Sea.     

Reporting of Quantitative Protein Electrophoresis in Australia and New Zealand: a Call for Standardisation

Background

The lack of guidelines on reporting standards for protein electrophoresis may have led to significant differences in reports from different laboratories.

Objective

To determine the extent of variation in reporting of protein electrophoresis results in Australia and New Zealand.

Method

Questionnaires were distributed to laboratories throughout Australia and New Zealand asking about protein electrophoresis practices and reporting.

Results

Extensive variation was found in the following reporting practices: (a) units for urine Bence Jones protein (BJP); (b) reporting absence of a paraprotein rather than a normal pattern; (c) numerical reporting of all protein fractions or only the paraprotein; (d) warning of possible inaccuracy in the serum immunoglobulin result of the paraprotein type; (e) co-migration of a paraprotein with a normal serum protein; (f) use of a confirmatory test when a known paraprotein is no longer detectable.

Conclusions

A working party should be established to make recommendations on the reporting of protein electrophoresis. Implementation of such recommendations should reduce both report variation between laboratories and the risk of misinterpretation of reports.     

Ectomycorrhizal species associated with Pinus radiata in New Zealand including novel associations determined by molecular analysis

Ectomycorrhizal (ECM) associates of the exotic plantation species Pinus radiata were investigated above and below ground over two years in the North Island of New Zealand. ECM species were identified using morphological and molecular (restriction fragment length polymorphism and DNA sequencing) analysis. Eighteen ECM species were observed fruiting above ground; 19 ECM species were identified below ground. In the above ground study, Wilcoxina mikolae, Rhizopogon pseudoroseolus and Inocybe sindonia were noted for the first time as ECM associates of P. radiata in New Zealand. Below ground, the species W. mikolae, R. pseudoroseolus, Rhizopogon luteorubescens, Pseudotomentella sp., Pseudotomentella tristis and Tomentella sp. were found as new associates of P. radiata in New Zealand. Additionally, six ECM types were found that could not be identified with molecular analysis. The putative ECM taxa Tricholoma pessundatum, Laccaria laccata and Hebeloma crustuliniforme were examined by molecular analysis, and species identifications were proposed to be changed to Tricholoma sp., L. laccata and Hebeloma sp. for specimens associated with P. radiata in New Zealand. The species identity of I. sindonia, previously unidentified to species level, was determined with direct sequencing.     

Diet-induced obesity in rats leads to a decrease in sperm motility

Background  

Obesity is rapidly becoming a worldwide epidemic that affects children and adults. Some studies have shown a relationship between obesity and infertility, but until now it remains controversial. Thus, the aim of the present study was to investigate the effect of high-fat diet-induced obesity on male reproductive parameters.     

Thyroid hormone-responsive pituitary hyperplasia independent of somatostatin receptor 2.

Mice homozygous for the targeted disruption of the glycoprotein hormone alpha-subunit (alphaGsu) display hypertrophy and hyperplasia of the anterior pituitary thyrotropes. Thyrotrope hyperplasia results in tumors in aged alphaGsu(-/-) mice. These adenomatous pituitaries can grow independently as intrascapular transplants in hypothyroid mice, suggesting that they have progressed beyond simple hyperplasia. We used magnetic resonance imaging to follow the growth and regression of thyrotrope adenomatous hyperplasia in response to thyroid hormone treatment and discovered that the tumors retain thyroid hormone responsiveness. Somatostatin (SMST) and its diverse receptors have been implicated in cell proliferation and tumorigenesis. To test the involvement of SMST receptor 2 (SMSTR2) in pituitary tumor progression and thyroid hormone responsiveness in alphaGsu(-/-) mutants, we generated Smstr2(-/-), alphaGsu(-/-) mice. Smstr2(-/-), alphaGsu(-/-) mice develop hyperplasia of thyrotropes, similar to alphaGsu(-/-) mutants, demonstrating that SMSTR2 is dispensable for the development of pituitary adenomatous hyperplasia. Thyrotrope hyperplasia in Smstr2(-/-), alphaGsu(-/-) mice regresses in response to T4 treatment, suggesting that SMSTR2 is not required in the T4 feedback loop regulating TSH secretion.
