Colon cancer is the second most common cause of cancer mortality in the Western world with metastasis commonly present at the time of diagnosis. Screening for propagation and metastatic behavior in a novel chimeric-mouse colon cancer model, driven by mutant p53 and β-Catenin, led to the identification of a unique, invasive adenocarcinoma. Comparison of the genome of this tumor, CB42, with genomes from non-propagating tumors by array CGH and sequencing revealed an amplicon on chromosome five containing CDK6 and CDK14, and a KRAS mutation, respectively. Single agent small molecule inhibition of either CDK6 or MEK, a kinase downstream of KRAS, led to tumor growth inhibition in vivo whereas combination therapy not only led to regression of the subcutaneous tumors, but also near complete inhibition of lung metastasis; thus, genomic analysis of this tumor led to effective, individualized treatment.  相似文献

Preexistent feline leukemia virus (FeLV) infection greatly potentiated the severity of the transient primary and chronic secondary stages of feline immunodeficiency virus (FIV) infection. Of 10 FeLV-FIV carrier cats, 5 died of experimentally induced FIV infection, compared with 2 deaths in 10 cats infected only with FeLV and 1 death in 7 cats infected only with FIV. FIV-infected cats with preexistent FeLV infections developed severe depression, anorexia, fever, diarrhea, dehydration, weight loss, and leukopenia 4 to 6 weeks after infection and were moribund within 2 weeks of the onset of signs, whereas cats infected only with FIV developed much milder self-limiting gross and hematologic abnormalities. Pathologic findings in dually infected cats that died were similar to those observed previously in cats dying from uncomplicated primary FIV infection but were much more widespread and severe. Coinfection of asymptomatic FeLV carrier cats with FIV did not increase the levels of FeLV p27 antigen present in their blood over that seen in cats infected with FeLV alone. The amount of proviral FIV DNA was much higher, however, in dually infected cats than in cats infected only with FIV; there was a greater expression of FIV DNA in lymphoid tissues, where the genome was normally detected, and in nonlymphoid tissues, where FIV DNA was not usually found. Dually infedted cats that recovered from the primary stage of FIV infection remained more leukopenic than cats infected with FIV or FeLV alone, and their CD4+/CD8+ T-lymphocyte ratios were inverted. One of these cats developed what was considered to be an opportunistic infection. It was concluded, therefore, that a preexistent FeLV infection in some way enhanced the expression and spread of FIV in the body and increased the severity of both the resulting transient primary and chronic secondary stages of FIV infection. This study also demonstrated the usefulness of the FIV model in studying the role of incidental infectious diseases as cofactors for immunodeficiency-causing lentiviruses.  相似文献

Photosynthetic rates and allocation of dry matter, nitrogen, and nonstructural carbohydrates were determined during onset of and recovery from a nitrogen stress for reproductive soybean (Glycine max [L.] Merrill cv Ransom) plants. Until the beginning of seed fill, non-nodulated plants were grown in flowing solution culture with 1.0 mM NO3- in a complete nutrient solution. One set of plants then was transferred to minus-nitrogen solution for 24 d of seed fill; a second set was transferred to a minus-nitrogen solution for 14 d followed by return to the complete solution with 1.0 mM NO3- for the remaining 10 d of seed fill; and a third set was continued on the complete solution. Net CO2 exchange rates of individual leaves, which remained nearly constant during seed fill for nonstressed plants, declined at an accelerated rate during onset of nitrogen stress as the specific content of reduced nitrogen in the leaves was decreased by remobilization of nitrogen to support pod growth. The rate of nitrogen remobilization out of leaves initially was relatively greater than the decrease in photosynthetic rate. While rate of pod growth declined in response to the developing nitrogen stress, photosynthetic assimilation of carbon exceeded reproductive demand and nonstructural carbohydrates accumulated within tissues. Following resupply of exogenous NO3-, specific rate of NO3- uptake by roots was enhanced relative to nonstressed plants. While there was little increase in content of reduced nitrogen in leaves, net remobilization of nitrogen out of leaves ceased, and the decline in photosynthetic rate stabilized at about 51% of that for nonstressed plants. This level of photosynthesis, combined with the availability of elevated pools of carbohydrates accumulated during stress, was sufficient to support the increases in both the specific rates of NO3- uptake and the rate of pod growth during recovery.  相似文献

The fifth base in human DNA, 5-methylcytosine, is inherently mutagenic. This has led to marked changes in the distribution of the CpG methyl acceptor site and an 80% depletion in its frequency of occurrence in vertebrate DNA. The coding regions of many genes contain CpGs which are methylated in sperm and serve as hot spots for mutation in human genetic diseases. Fully 30-40% of all human germline point mutations are thought to be methylation induced even though the CpG dinucleotide is under-represented and efficient cellular repair systems exist. Importantly, tumor suppressor genes such as p53 also contain methylated CpGs and these serve as hot spots for mutations in some, but not all, human cancers. Comparison of the spectrum of mutations present in this gene in different human cancers allows for predictions to be made on the molecular mechanisms of tumorigenesis.  相似文献

Histological features of adrenal glands from captive and unstressed free-ranging nine-banded armadillos were compared to determine if undefined stress factors associated with captivity were capable of causing changes in adrenal morphology. Animals in captivity less than 3 mo showed histological features identical to unstressed free-ranging animals. Animals in captivity 6 mo or longer, however, showed adrenal changes like those previously described in other mammals exposed to various types of stress, except that there was no accompanying increase in adrenal weight. Similar adrenal changes were found in free-ranging animals exposed to extremely harsh winter weather. Females from both the chronic captive population and this stressed free-ranging population were seen to suffer reproductive failure. Reproductive function in males was not affected. There was no detectable sex difference in gross or histological adrenal changes; however, previous reports indicate there is a sex difference in the secretory product from the adrenals of captive nine-banded armadillos. A possible relationship between reproductive failure in the female and the sex difference in adrenal secretory products is discussed.  相似文献

Experimental infection of white-tailed deer fawns with Salmonella meleagridis was accomplished. The fawns suffered clinical illness, similar to spontaneous cases observed in the field. This disease may be an important factor in fawn survival in wild herds based on the frequency with which Salmonellae could be isolated in wild fawns. The clinical disease was acute, characterized by rapid depression and dehydration. Death ensued in three of eight experimental cases. The survivors suffered clinical illness.  相似文献

A model of the human systemic arterial tree has been devised, based on a lumped-parameter-circuit approximate form. This model has been set up and studied on an analog computer. A feature of this simulation is the division of the arterial system into sections whose lengths are inversely proportional (approximately) to their cross-sectional area-or what is termed ‘equal-volume’ modeling.

Great care was exercised in the determination of the model parameters, using expressions for these parameters from a recent paper by Rideout and Dick on fluid flow in distensible tubes, with numerical values based on measurements reported in the medical literature.

The simulated pressure and flow waveforms obtained with the model compare favorably with data recorded from the normal adult human, and exhibit such well-known features as distal delay and peaking of pressure pulses. The aortic input impedance vs. frequency curve checks well against measurements on the human. The model also provides a simple means for determination of cardiac output, cardiac work and cardiac power under various assumed conditions such as variation of heart rate.  相似文献
