Interactions of intracellular membranes with microtubules play a fundamental role in the dynamic organization of cytoplasmic organelles. The microtubule-based motors kinesin and cytoplasmic dynein are responsible for directed movement of vesicles and organelles, but in vitro assays indicate the existence of another class of proteins linking membranes to microtubules. CLIP-170, a cytoplasmic linker protein that mediates binding of endosomes to microtubules, provides a paradigm for understanding how these proteins may complement the role of motors in regulating microtubule-dependent membrane trafficking.  相似文献

Colonies of the eusocial Damaraland mole-rat, Cryptomys damarensis , are founded from a single reproductive pair of animals genetically unrelated by common descent. All non-reproductive members of the colony are progeny of this reproductive pair. In colonies where the reproductive female has been experimentally removed or has died a natural death, there is a strict incest avoidance and the colony remains reproductively quiescent. Reinstatement of sexual activity in a queenless colony may be brought about in the laboratory by the introduction of an unfamiliar and unrelated adult male.
In the queenless colony under study, there was a marked change in social structure with an increase in Landau's index of linearity from 0.8 to 0.9 on introduction of the new male. The unrelated male became a high ranking dominant reproductive male. The youngest, but most dominant n on-reproductive female became sexually active and subsequently became pregnant and hence acquired the position of reproductive female. The new reproductive female exhibited heightened progesterone (9nmols/mmol creatinine) and oestradiol (3000pmols/mmol creatinine) concentrations in the urine relative to the other non-reproductive females. These hormone concentrations were indicative of a reproductively active female.
Behavioural and hormonal data are presented to show that sexual activity can be re-instated in queenless colonies of laboratory maintained mole-rats by the introduction of unrelated male mole-rats.  相似文献

The ovipositor of Spodoptera littoralis has many sensilla, which implies a distinct sensory function of this organ in the assessment of the egg laying substrates and placement of eggs. The ovipositor is composed of two papillae densely packed with sensilla. Scanning electron microscopy showed that there are four categories of sensilla chaetica (s. chaetica), characterized based on differences in distribution, length and morphology, distributed on each papilla: Type I with 69 ± 2.9 sensilla on each papilla (length: 29–163 μm) distributed at the base; Type II with 19 ± 1.8 sensilla (length: 287–446 μm) distributed medially; Type III with 120 ± 7.8 sensilla (length: 62–156 μm) distributed distally; and Type IV 8 ± 0.9 sensilla (length: 72–91 μm) scattered between Type III sensilla. The structure of s. chaetica Types I–III suggests a mechanosensory function, whereas electrophysiological analysis of s. chaetica Type IV showed responses from four gustatory neurons and one mechanosensory neuron. Of the chemoreceptive neurons, one responded to salt, one to the secondary compound caffeine, one to sugars, mainly sucrose and fructose, and lastly one potentially to water. Behavioral studies are needed to verify the role of these sensilla during choice of oviposition site.  相似文献

Activated oncospheres of T. hydatigena within filtration membrane diffusion chambers implanted intraperitoneally into dogs developed into larvae 3 mm in dia possessing a scolex anlage without hooks. Prophylactic vaccines can be expected to be one of the major practical outputs of parasitology research. Various groups within Australia have pursued the vaccine objective for several years, with particular emphasis on blood-stage falciparum malaria in man, intestinal helminths of sheep and cattle, cutaneous myiasis (blowfly strike) in sheep, cysticercosis in sheep and cattle, bovine babesiosis, and cattle ticks. Other vaccine programmes are concerned with giardiasis, filariasis, toxoplasmosis, fascioliasis, coccidiosis in poultry, cutaneous leishmaniasis and schistosomiasis japonica. For many years, the only available vaccine against a parasite in Australia has been the attenuated Babesia bovis vaccine produced by the Tick Fever Research Centre of the Queensland Department of Primary Industries. Strategies for achieving molecular vaccines are generally similar within the various research groups. They involve analysis of the immunology and immunochemistry of a model or in-vitro system; development of functional monoclonal antibodies; analysis of antibody specificities in clinically and/or functionally defined polyclonal sera; screening of cDNA or genomic expression libraries; peptide synthesis; identification of an appropriate vaccination schedule involving adjuvants or new recombinant DNA-based antigen delivery systems. Candida utilis was grown in batch culture with and without oxygen control. The concentrations of A-, B-, and C-type cytochromes were found to vary with the initial glucose concentration, with the dissolved oxygen concentration, and with time. A-type was the most sensitive. After glucose was essentially exhausted, the yeast catabolized ethanol, if it had been growing in a relatively low initial glucose concentration, or non-glucose carbohydrate, including some of that previously accumulated within the cell, if it had been growing in a high initial glucose concentration. This difference in metabolic pattern could explain why cytochrome derepression was initiated soon after glucose uptake ceased only if initial glucose had been relatively low. The effects of glucose and dissolved oxygen concentrations on yeast cytochromes and respiratory activity are discussed.  相似文献

The growth of Saccharomyces carlsbergensis in continuous culture has been studied when dissolved oxygen and glucose concentrations were held constant at a series of steady-state levels. Both oxygen and glucose controlled the degree of aerobic metabolism and of ethanolic fermentation. When the glucose uptake rate was low (between 1.2 and 2.8 mmoles per hour per gram of yeast) the relative distribution of glucose between ethanolic and aerobic fermentation was sensitive to oxygen: when dissolved oxygen was near to saturation, glucose metabolism was 0.98 aerobic; when dissolved oxygen was 0.01 saturated, 0.8 of intake glucose metabolism was by ethanolic fermentation. On the other hand when glucose intake was high (between 7.6 and 18.2 mmoles) metabolism was predominately by ethanolic fermentation even when dissolved oxygen concentration was at saturation. The extent, to which catabolism proceeded by an anaerobic or aerobic pathway, as judged by ethanol production, was controlled more by the uptake of glucose than of oxygen.  相似文献

Mature eggs of Taenia taeniaeformis hatched readily in the presence of sodium hypochlorite and no loss in infectivity of oncospheres for mice was observed after hatching. Crude and sodium deoxycholate-solubilized antigens (termed TtO-DOC) prepared from such oncospheres stimulated high levels of protection against T. taeniaeformis infection in immunized mice similar to those described previously for oncospheres prepared by other methods. Mice immunized with TtO-DOC antigens that had been exposed to potassium metaperiodate remained significantly protected against infection. Exposure of TtO-DOC antigens to pronase and thermolysin, or to trypsin, significantly reduced the ability of these antigens to protect mice against infection. These data suggest that the antigens which immunize mice against infection include protein components. 125I-labelled TtO-DOC antigens were immunoprecipitated with sera from mice infected with T. taeniaeformis and the immunoprecipitates analysed by sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE). Immunoprecipitation with sera from C3H/He mice infected for 28 days revealed a single major labelled protein antigen having a relative molecular mass (Mr) of 31,000. Sera from 5-month infected C3H/He mice immunoprecipitated at least thirteen labelled antigens, including one at Mr 31,000. Attempts to use SDS-PAGE separated proteins to immunize mice showed that oncosphere antigens exposed to the reducing conditions prior to SDS-PAGE lost their ability to protect mice against infection. It was concluded that SDS-PAGE was an unsatisfactory technique for the isolation of a host protective fraction of TtO-DOC antigens. TtO-DOC proteins were resolved by PAGE performed in the presence of sodium deoxycholate (DOC-PAGE) and mice were vaccinated with cut-outs from the gel. A fraction of the DOC-polyacrylamide gel was found to be effective in immunizing mice against infection. Thus, although the characteristics of the protein antigens in this DOC-PAGE fraction have yet to be determined, an important fractionation technique has been identified. It was shown that partial removal of DOC from oncosphere antigen preparations solubilized in 1% DOC was required for the antigen to stimulate protective immunity. These findings will facilitate further antigen characterization studies towards the development of a defined-antigen vaccine in murine cysticercosis. This is particularly so as attempts to raise anti-oncospheral monoclonal antibodies capable of passively transferring protection to mice by using crude antigen preparations to immunize donor mice have not been successful.  相似文献
