The function of occludin (Occ) in the tight junction is undefined. To gain insight into its role in epithelial cell biology, occludin levels in Madin-Darby canine kidney II cells were suppressed by stably expressing short interfering RNA. Suppression of occludin was associated with a decrease in claudins-1 and -7 and an increase in claudins-3 and -4. Claudin-2 levels were unaffected. The tight junction "fence" function was not impaired in suppressed Occ (Occ) clones, as determined by BODIPY-sphingomyelin diffusion in the membrane. The most striking changes were those related to control of the cytoskeleton and the "gate" function of tight junctions. A reduced ability of Occ clones to extrude apoptotic cells from the monolayers suggested that neighbors of apoptotic cells either failed to sense their presence or were unable to coordinate cytoskeletal activity necessary for their extrusion. To further test the extent to which actin cytoskeletal activity depends on the presence of occludin, Occ and Occ+ monolayers were depleted of cholesterol. Previous studies showed that cholesterol depletion is associated with reorganization of the actin cytoskeleton and a fall in transepithelial electrical resistance. In contrast to control Occ (Occ+) cells, transepithelial electrical resistance did not fall significantly in cholesterol-depleted Occ monolayers and they failed to generate Rho-GTP, one of the signaling molecules involved in regulating the actin cytoskeleton. While steady-state transepithelial electrical resistance was similar in all clones, tight junction permeability to mono- and divalent inorganic cations was increased in Occ monolayers. In addition, there was a disproportionately large increase in permeability to monovalent organic cations, up to 6.96 Å in diameter. Chloride permeability was unaffected and there was little change in mannitol flux. The data suggest that occludin transduces external (apoptotic cells) and intramembrane (rapid cholesterol depletion) signals via a Rho signaling pathway that, in turn, elicits reorganization of the actin cytoskeleton. An overview of the morphology and life cycle of Batrachochytrium dendrobatidis, the cause of chytridiomycosis of amphibians, is presented. We used a range of methods to examine stages of the life cycle in culture and in frog skin, and to assess ultrastructural pathology in the skin of 2 frogs. Methods included light microscopy, transmission electron microscopy with conventional methods as well as high pressure freezing and freeze substitution, and scanning electron microscopy with critical point drying as well as examination of bulk-frozen and freeze-fractured material. Although chytridiomycosis is an emerging disease, B. dendrobatidis has adaptations that suggest it has long been evolved to live within cells in the dynamic tissue of the stratified epidermis. Sporangia developed at a rate that coincided with the maturation of the cell, and fungal discharge tubes usually opened onto the distal surface of epidermal cells of the stratum corneum. A zone of condensed, fibrillar, host cytoplasm surrounded some sporangia. Pollen-limited plants are confronted with a difficult tradeoff because they must present showy floral displays to attract pollinators and yet must also minimize their apparency to herbivores. In these systems, traits that increase pollinator visitation may also increase herbivore oviposition and overall plant resistance may therefore be constrained to evolve largely as a correlated response to selection on plant apparency or vigor. We used a family-structured quantitative genetic experiment to evaluate the importance of ungulate browsing, flowering date and plant height (traits that are related to overall vigor), and variation in a putative phytochemical defense (cucurbitacin production) on patterns of seed fly attack in a scarlet gilia population. We found significant genetic variation in the amount of insect damage plants experience in the field, providing evidence that resistance may evolve. In addition, we found that browsing reduced seed fly attack and that oviposition is strongly related to plant size and flowering date; large, early flowering plants experience high attack. In addition, we found that high cucurbitacin production was correlated with low seed fly damage, although this effect was relatively weak.We found directional selection on final plant height and flowering date; tall, early flowering plants had the highest reproductive success. In addition, we found negative directional selection on cucurbitacin production, which may indicate a high cost of cucurbitacin or other functions of this phytochemical. Although seed fly herbivory arguably decreases plant fitness, we found an unexpected positive relationship between damage and fitness. A negative relationship between fitness and damage may be masked in this system through strong positive indirect correlations between patterns of damage and levels of pollinator visitation. Finally, we found significant genetic variation in flowering date, plant height, and cucurbitacin production. Resistance to seed flies may evolve in this population, but largely as a non-adaptive correlated response to selection on overall plant vigor. Molecularly imprinted thin films consisting of proteins embedded in polymerised aminophenyl boronic acid have been made on glass supports. The protein contents of the films have been optimised to achieve a maximum energy of interaction between the film and the native template. The fabrication of the films and the subsequent removal from their surfaces of the imprint proteins has been shown to be a facile and easily reproduced process. The enthalpy changes associated with the rebinding of the films with their original templates (lysozyme and cytochrome c) and with non-native templates has been examined by micro-calorimetry. The results demonstrate that thin films can be successfully imprinted as shown by the significant reduction in the enthalpy (DeltaH) observed when the films were rebound with proteins other than the original templates. Additionally, it was shown that after binding, non-template proteins could be removed by washing and a greater enthalpy again observed when the films were rebound with the native protein compared to that which had been found with the non-native protein. 相似文献
