In vitro lymphocyte stimulation of sensitized rabbit lymphocytes to specific antigen (ovalbumin) was found to depend on thymic-dependent lymphocytes. This conclusion is based on an enhanced response upon enrichment with T lymphocytes by passage of lymphocytes through nylon wool, and on the elimination of the response after treatment of lymphocytes with complement and an antiserum to rabbit thymus cells prepared in a goat. Specificity of the antiserum was demonstrated by elimination of in vitro T-cell function and retention of in vitro B-cell functions. 相似文献

In the renal glomerulus, the narrow slits between adjacent epithelial podocytes are bridged by a diaphragm (2, 8, 11). In rat and mouse kidneys fixed by perfusion with tannic acid and glutaraldehyde (TAG), it has recently been discovered that this diaphragm has a highly ordered, isoporous substructure (9). It consists of a regular array of alternating cross bridges extending from the podocyte plasma membranes to a centrally running filament. This zipperlike pattern results in two rows of rectangular pores, approximately 40 X 140 A in cross section, dimensions consistent with the proposed role of the diaphragm as an important filtration barrier to plasma proteins (6). In the present study, we found in freeze-cleaved and in freeze-etched normal rat glomeruli that the surface of the slit diaphragm has an appearance conforming to the pattern found in sectioned material. 相似文献

Near surface thermoclines form each day in the limnetic waters of Lake Titicaca (Peru-Bolivia) and thereby retain phytoplankton under extreme irradiances. This bright light exposure results in strongly depressed chlorophyll fluorescence and photosynthesis which both decay (bright light) and recover (dim light) by first order rate kinetics. During each afternoon the phytoplankton are redistributed by wind-induced mixing, and full recovery is accomplished soon after nightfall. In vivo fluorescence was measured over this diet cycle both with (Fb) and without (Fa 3-(3,4-dichlorophenyl)-1,1-dimethyl urea. Strongest bright light effects were on the parameter (Fa– Fa), a crude measure of operational photosystem II reaction centers (RC IIs). On dates of strong thermocline development, surface (FFa– Fa) was reduced to 5% or less of that for the mixed layer maximum. Fluorescence depression was greater in the lake than in Pyrex bottles incubated at fixed depths for 4 h. Ultraviolet light intensified the photoinhibitory response, but strong (Fb– Fa) depression could be induced by photosynthetically available radiation alone. In Lake Titicaca, photoinhibition apparently operates by reversible in-activation of RC IIs. It occurs in the natural water column and is not simply an artifact affixed bottle incubations. 相似文献

New insights in Salicornia L. and allied genera (Chenopodiaceae) inferred from nrDNA sequence data 总被引:1,自引:0,他引:1
A phylogenetic analysis was performed based on ITS DNA sequences of fourteen samples from different sources of six species of Salicornia, the three allied genera Arthrocnemum, Sarcocornia and Halocnemum of the same tribe Salicornieae, and other genera of the subfamily Salicornioideae used in previous studies. Bassia hirsuta, Camphorosma monspeliaca (subfamily Chenopodioideae) and four species of Suaeda (subf. Suaedoideae) were chosen as outgroups. Results show that the annual genus Salicornia is a sister group to the perennial genera Sarcocornia, Arthrocnemum and Halocnemum. Moreover, the phylogenetic analysis based on ITS results distinguished two groups of Salicornia species which fitted with ploidy level: one group consisted of diploid species, and the second of tetraploid ones. Sarcocornia and Arthrocnemum are shown to be closely related, even though the species investigated here exhibited an evident distance between their ITS sequences. On the basis of our results, these two genera should be united. Bienertia (already separated as Bienertieae) was confirmed as probable outgroup to the subf. Salicornioideae, while Kalidium (subf. Salicornioideae, tribe Halopeplideae) was an outgroup to the rest of the Salicornioideae (tribe Salicornieae). The group Allenrolfea plus Halocnemum was the most basal of the tribe Salicornieae amongst those investigated in this study. The two samples of Halocnemum strobilaceum used in this work displayed numerous changes (transitions and transversions) in their respective sequences, probably related to their morphological and chorological differentiation. On the basis of our analysis, the most probable basal chromosome number for Salicornieae appears to be 2n = 18. The same number would also be the base number for the annual genus Salicornia and the perennial Arthrocnemum ( + Sarcocornia), with polyploidy arising independently in the two groups. 相似文献

Horizontal transects measuring phytoplankton biomass and temperaturewere made in Lake Tahoe in the nearshore epilimnion in 1976and 1977, near the deep chlorophyll maximum in midlake in 1977,and in the deep thermocline in 1976. Variance spectra from thesetransects indicate that large length scale patchiness dependson large-scale habitat variability, manifested as nitrate patchiness,caused by stream inflow in the nearshore epilimnion and by differentialtransport processes in the deeper water. When large-scale sourcesof habitat variability are absent, phytoplankton spatial patternsare not dominated by large-scale patches and are consistentwith a random distribution. No significant small-scale patchinesswas seen under any environmental conditions.

Subunit structure in the walls of sectioned microtubules was first noted by Ledbetter and Porter (6), who clearly showed that certain microtubules of plant meristematic cells have 13 wall protofilaments when seen in cross section. Earlier, protofilaments of microtubular elements had been described in negatively stained material, although exact counts of their number were difficult to obtain. In microtubular elements of axonemes, some success has been achieved in visualizing protofilaments in conventionally fixed and sectioned material (8, 10); much less success has been achieved in identifying and counting protofilaments of singlet cytoplasmic microtubules. By using glutaraldehyde-tannic acid fixation, as described by Misuhira and Futaesaku (7), Tilney et al. (12) studied microtubules from a number of sources and found that all have 13 protofilaments comprising their walls. These authors note that "...the number of subunits and their arrangement as protofilaments appear universal...". Preliminary studies of ventral nerve cord of crayfish fixed in glutaraldehyde-tannic acid indicated that axonal microtubules in this material possess only 12 protofilaments (4). On the basis of this observation, tannic acid preparations of several other neuronal and non-neuronal systems were examined. Protofilaments in microtubules from these several cell types are clearly demonstrated, and counts have been made which show that some kinds of microtubules have more or fewer protofilaments than the usual 13 and that at least one kind of microtubule has an even rather than an odd number. 相似文献
