Freeze fracture and lanthanum tracer experiments have shown that gap junctions exist throughout folliculogenesis between granulosa cells and growing mouse oocytes (Anderson and Albertini, J. Cell Biol.71, 680–686, 1976). The following lines of experimentation in the present study suggest that metabolic cooperativity exists between granulosa cells and their enclosed oocytes, i.e., gap junctions are functional, and that in most cases examined, greater than 85% of the metabolites present in follicle-enclosed oocytes were originally taken up by the granulosa cells and transferred to the oocyte via gap junctions: (1) When incubated with various radiolabeled compounds, follicle-enclosed oocytes contained more intracellular radioactivity than did oocytes with no attached granulosa cells (denuded oocytes); (2) for two radiolabeled ribonucleosides examined, the distribution of phosphorylated metabolites in follicle-enclosed oocytes resembled that of granulosa cells and differed significantly from that in denuded oocytes; (3) pulse-chase experiments with radiolabeled ribonucleosides revealed that during the chase period more radioactivity became associated with the follicle-enclosed oocyte; (4) treatments known to disrupt gap junctions in other cell types were effective in reversibly uncoupling metabolic cooperativity between granulosa cells and oocytes; and (5) a series of control experiments using (a) medium conditioned by granulosa cells and (b) cocultures of denuded oocytes and granulosa cells in which physical contact between the two cell types was not permitted demonstrated that contact between follicle cells and oocytes was necessary for observing metabolic cooperativity. Metabolic cooperativity was also found between follicle cells and oocytes in the two culture systems which support growth of mouse oocytes in vitro. The fact that oocytes do not grow well, if at all, in the absence of follicle cells and the large contribution of nutrients apparently furnished to the oocyte by the granulosa cells is consistent with the concept that gap junction mediated metabolic cooperativity between follicle cells and their enclosed oocytes is vital for mammalian oocyte growth.  相似文献

We have analysed in detail the Na+ content and Na+ influx during fertilization and first divisions of the sea urchin egg (Paracentrotus lividus) using a filtration technique devised to eliminate rapidly contamination by the Na+ of external sea water. In the first 5 min following fertilization the egg fills up with Na+ (+ 30%). Thereafter Na+ is extruded and the Na+ content stabilizes at about 60% of the unfertilized egg level by the second cleavage (2 h). The initial increase in Na+ content is due to a large increase in Na+ influx already detected at 20 sec. The Na+ influx reaches its maximum at 1 min and its minimum at 5 min. H+ excretion follows the same kinetics. A second increase in Na+ influx is noted 5–10 min after fertilization; it reaches its maximum at prophase metaphase (30 min) and its minimum during cleavage (60 min). These oscillations in Na+ influx were observed for the first three divisions. Fertilization also immediately stimulates the Na+ efflux which remains elevated throughout the cell cycle and is responsible for the depletion of the Na+ content of the embryos. Activation of the eggs by weak amine bases (5 mM NH4Cl) which bypasses the early cortical reaction produces only a depletion in the Na+ content of the egg similar to that produced by fertilization. NH4Cl also increases the Na+ influx soon after fertilization, although no transient variations are noted.  相似文献

Analyses of four years of in situ sulfate uptake by microplankton communities in two,trophically different lakes showed that about 12% of the experiments had dark uptake equal to or higher than uptake at ambient light. Three axenic algal cultures subjected to different light intensities showed that sulfate uptake patterns, relative to primary productivity, vary with species and although sulfate uptake tends to decrease at lower light levels, at or very near darkness, in physiologically active (young) cultures sulfate uptake frequently increases dramatically. The field data, when summarized according to the light received, shows the same trends seen in the axenic cultures. It is concluded that sulfate uptake is only loosely associated with inorganic carbon uptake (primary productivity) and that under some circumstances a low level of light may increase the sulfate uptake rate.  相似文献

This investigation is concerned with a scattering matrix approach which is proposed for the non-destructive, differential diagnosis of muscle diseases. A number of muscle diseases are classified according to their various pathological indications and appropriate material parameters are derived for utilization as input data for a theoretical model. In the mathematical analysis phase of the model, Waterman's T-matrix approach in conjunction with statistical averaging for both position and orientation of muscle fibers are employed to obtain the attenuation due to geometric dispersion for a wide range of frequencies. The numerical results not only exhibit qualitative agreement with existing experimental data for normal muscle but also display differentiable patterns for the various muscle disease cases. The formulation is an improvement over the previously applied scattering theory in that it obtains the attenuation over a continuous frequency spectrum.  相似文献

Plasma renin activity (PRA) was studied in the rats bred by Dahl for susceptibility (S-strain) or resistance (R-strain) to salt (NaCl) induced hypertension. The pH curves for PRA had different shapes. The difference in shape of the pH curves was reflected in the ratio of PRA pH 8/PRA pH 6.5. This ratio was shown to be characteristic of the strain and to be independent of changes in absolute PRA level induced by variation in dietary NaCl. The ratio of PRA pH 8/PRA pH 6.5 was also different between strains in weanling as well as adult rats. The underlying cause for the strain difference in the effect of pH on PRA is unknown, but may involve molecular differences between strains in either renin or renin substrate.  相似文献

During microaerophilic growth of magnetic spirillum MS-1 on tartrate and nitrate, a maximal cell density was obtained at an initial oxygen partial pressure of 17 Pa. A transient accumulation of nitrous oxide and a 1:2 (mol/mol) stoichiometry between tartrate oxidation and nitrate reduction were observed, indicating that the organism carried out a respiratory type of metabolism.  相似文献
