Identification of a high molecular weight nervous system specific cell surface glycoprotein on murine neuroblastoma cells 总被引:1,自引:0,他引:1
An antiserum to N18 neuroblastoma cells has been used to identify a glycoprotein of apparent molecular weight greater than 200 000 D in SDS-polyacrylamide gels. This glycoprotein (Band 1) is found in culture medium of N18 cells. An immunologically similar component can be immunoprecipitated from detergent extracts of enzymatically iodinated or biosynthetically labelled viable cells. Anti-band 1 activity can be adsorbed from the antiserum by intact N18 cells but not four other cultured murine cell lines. Normal adult murine brain also adsorbs anti-band 1 activity but adult murine adrenal, heart, kidney, liver, lung, and spleen do not. Several experiments indicate that band 1 is not myosin heavy chain or the fibroblast LETS protein. Thus band 1 is a newly identified high molecular weight nervous system specific glycoprotein. 相似文献

A general procedure is described for measuring and testing population differences in gametic frequencies. The total dispersion among populations is subdivided in hierarchical fashion. The multiple-locus treatment is simply the sum of the single-locus analyses, provided gametic equilibrium obtains among the loci. In the event that gametic equilibrium does not obtain, correlations among loci need to be dealt with.—The analysis is then used to examine the genetic infrastructure of two Indian tribes from South America, the Ye'cuana (Makiritare) and the Yanomama. From historical evidence, we may identify several "clusters" of villages within each tribe. The demographic and cultural practices affecting village formation and the maintenance of peer integrity are rather different in these tribes, however, and lead us to postulate rather different patterns of genetic variation among villages. Analyses of five codominant two-allele loci, four dominant two-allele loci and two complex loci (with four codominant haplotypes each) demonstrate that Yanomama clusters are more disparate than Ye'cuana clusters, as would have been predicted on sociocultural grounds. 相似文献

Lysine vasopressin (LVP), arginine vasopressin, oxytocin, and arginine vasotocin administered intraventricularly (icv) to mice all provoked a dose-dependent behavioral response in the range 0.1 – 1.0 μg. This response included a pronounced hyperactivity, extensive foraging, increased grooming, and at higher doses, stereotyped scratching, squeaking, and occasional barrel rolling. The four hormones were all approximately equipotent. Desglycinamide lysine vasopressin and [desaminocys1, D-Arg8] vasopressin produced some of the characteristic behaviors, but were much less potent. While pretreatment of the animals with reserpine (5 mg/kg ip), haloperidol (0.5 mg/kg ip), or physostigmine (0.5 mg/kg ip) sedated the animals and attenuated the locomotion and grooming, these drugs did not substantially alter the characteristic behavioral responses to LVP. Pretreatment with α-methyl-p-tyrosine (400 mg/kg ip), p-chlorophenylalanine (320 mg/kg ip), 6-hydroxydopamine (100 μg icv), ergotamine (0.5 μg icv), ethoxolamide (52 ng icv), diphenhydramine (20 μg icv), prostaglondin F2α (2 μg icv), or naloxone (1 mg/kg ip) did not alter the LVP-induced behaviors. Transport properties of rabbit small intestinal mucosa were investigated to characterize the process by which epithelial cells maintain normal Na and K gradients across the cell membrane. Active transport of Na from the cell proceeded at a faster rate in the presence of K; and active transport of K into the cell was stimulated by the presence of Na. Following preincubation at 0°C to reduce tissue K content, a greater transmural electrical potential difference (PD) and short-circuit current (Isc) developed as the temperature was raised to 37°C if K was present in the bathing solution. The PD and Isc, which generally reflect the rate of active Na transport in ileum under control conditions, increased immediately upon raising the K concentration in the serosal solution from 0 to 10 nM.The results present the first direct indication in mammalian intestine of an interdependence of the Na and K active transport processes which regulate the intracellular content of these cations. 相似文献
