A high-performance liquid chromatographic procedure was developed for the determination of caffeine in various biologic fluids and coffee. A reversed-phase column and UV detection at 254 nm were used to obtain a sensitivity of 0.1 μg/ml caffeine in serum and saliva using a sample volume of 0.1 ml. Caffeine metabolites and commonly ingested xanthines do not interfere with the assay. The within-day coefficients of variation were 9.8 and 9.9% at plasma caffeine concentrations of 2 and 10 μg/ml, respectively. The day-to-day coefficients of variation were 6.8 and 6.6% at plasma caffeine concentrations of 2 and 10 μg/ml, respectively. Serum and saliva caffeine concentrations were determined following a single oral dose of coffee and an intravenous infusion of caffeine in one subject. Computer estimates of caffeine pharmacokinetic parameters in one subject are in excellent agreement with previously published values. 相似文献

Modulation of epithelial cell proliferation by the dissolved oxygen concentration (PO2) of the growth medium was assessed with primary human foreskin epithelium and a continuous monkey kidney epithelial cell line (LLC-MK2). Direct measurement of the growth medium PO2 provides the first quantitative evaluation of epithelial cell proliferation as a function of PO2 provides the first quantitative evaluation of epithelial cell proliferation as a function of PO2. Sustained proliferation of LLC-MK2 cells occurs in serum-free medium equilibrated with a gas phase containing 18% or 30% O2 v/v. Mid-logarithmic phase cultures rapidly consume dissolved oxygen; this results in a 60–70 mm Hg decline in PO2 and leads to a stable growth medium PO2 between 70 and 100 mm Hg, well above anoxic values. In contrast, if culture medium is equilibrated with a gas phase containing 0% or 1% O2 v/v to yield a growth medium PO2 ~ 20–40 mm Hg, proliferation of LLC-MK2 and primary foreskin epithelial cells is retarded, and LLC-MK2 cells use little dissolved oxygen. Gentle, continuous rocking to prevent diffusion gradient formation enhances proliferation slightly at the higher PO2, but neither periodic fluid renewals nor continued rocking stimulates cells retarded by a lowered oxygen concentration to resume proliferation. The data collectively demonstrate that epithelial cell proliferation requires a PO2 > 40 mm Hg, and threshold requirements are probably closer to 70 mm Hg. Glycolysis continues at a PO2 insufficient for proliferation, but more lactic acid accumulates in actively proliferating cultures than in cultures equilibrated with 0% oxygen. We conclude that epithelial cells in vitro both consume more oxygen and require a higher PO2 for continued proliferation, and that the oxygen requirement for epithelial cell proliferation exceeds that of a comparable population of fibroblasts for which low oxygen may enhance survival and proliferation. 相似文献

The optimum temperatures for population development were determined for six species of bacterial-feeding nematodes from among eight temperatures, ranging from 5 to 40 C. Four of the species are cohabiting species. The range of temperatures over which population development occurs (temperature niche breadth) is different for the cohabiting species. The green alga Stichococcus bacillaris Naeg. is able to take up at least eleven amino acids. All of these except glutamic and aspartic acids are transported by carrier systems that obey saturation kinetics. The acidic amino acids enter the cell by passive diffusion. Michaelis-Menten parameters (Ks and Vmax) were calculated for several amino acids. All obey simple Michaelis-Menten behavior except for 2-methylalanine and leucine which may have double carrier systems of different affinities. Interactions between pairs of amino acids suggest that there is at least one carrier system specific for basic amino acids and probably several systems specific for neutral amino acids. Further analysis of neutral amino acid interactions reveal that the uptake of several amino acids is incompletely inhibited by competitor uptake at infinite concentration. The simplest interpretation of the data is the operation of three carrier systems for neutral amino acids, one of which has higher affinity and broader specificity than the other two. The amino acid carrier systems appear to operate by an active mechanism. The metabolic poison DCCD inhibits uptake up to 99%. The capacities of the neutral amino acid carrier systems are increased when cells are grown in medium containing suboptimal concentrations of nitrogen. 相似文献

Aspects of membrane stucture and functions were studied in ethidium bromide resistant cells. Submitochondrial particles were solubilized and electrophoresed. The gel patterns, representing mitochondiral membrane proteins, demonstrated qualitative and quantitative alterations in mitochondrial preparations derived from virus-transformed cells and ethidium bromide resistant cells as compared to the control cells. The plasma membrane glycoproteins were labelled by the sodium borohydride method. The glycoporteins were released with Triton X-100 and electrophoresed. Fluorograms of the gels demonstratred some marked differences between the ethidium bromide resistant cells and their parental strain. The observed alterations in the membrane glycoproteins did not result in altered glucose transport properties or in the elution patterns of plasma membrane glycopeptides as analyzed by Sephadex G-50 chromatography. Dye uptake and binding studies with intact parental and drug resistant cells and their isolated mitochondria demonstrated no alteration of the membrane permeability or the number of binding sites for ethidium bromide. Similar results were also obtained with a cyanine dye. This latter finding was significant in that it permitted one to exclude dye exclusion as a mechanism for ethidium bromide resistance. 相似文献

Abstract A possible involvement of amino acid uptake mechanisms in the etiology of the human neurodegenerative disease, Huntington's disease (HD), was investigated. Measurement of glutamate (Glu) and aspartate (Asp) uptake was performed in blood platelets, which have previously been shown to constitute a peripheral model system for central amino acid uptake processes. Analyses of Glu and Asp accumulation at 10−7 M and kinetic examination of the high affinity site for Glu indicate no significant differences between control and HD platelets. A genetically determined defect in amino acid uptake therefore does not seem to underlie the nerve cell loss observed in HD patients. 相似文献

Summary Periodical cicadas (Magicicada spp.) rarely flew distances greater than 50 m across an open field or along a forest edge. Most cicadas caught after flying 50 m or more were females and all of these females had mated. Flights were most common when adults were 2 to 3 weeks old. Among insects in general, most dispersing individuals are barely post-teneral or extremely young. Cicadas are discussed as an exception to this generality. Both sexes of cicadas are attracted by the male song to chorusing centers for mating (Alexander and Moore 1958). Trees which were chorusing centers had more eggnests than those which were not, suggesting a lack of postmating dispersal from the chorusing trees. 相似文献

Estimates are made of the effects which a steady magnetic field may have on the action potential. These are divided into two classes: Current distortion and inductive effects. Employing relations of the Hall Effect, it is found that for distortion of the action-potential current-pattern of the squid axon, magnetic field strengths of the order of Gauss are required. Changes in the current pattern due to inductive effects similarly imply that a magnetic field kgauss distributed over ? 1 cm of axon would measurably perturb current flow in the action potential. The phenomenological coefficient α characterizes the asymmetry of the current pattern. For the case of a cylindrically symmetric response, α ? 0, in which case inductive perturbation of the action potential is nullified. Finite inductive effects for the case of the cylindrically asymmetric action potential are found also to be dependent on the orientation of the magnetic field with respect to the action potential. The effects of the extent of the applied magnetic field are also discussed. 相似文献
