Tissue biopsies from (1) some chronic inflammatory diseases, (2) a necrotic tumoral process, (3) normal human lymphatic ganglia, and (4) two congenital diseases of the adrenal cortex were selected for study. A block from each biopsy was fixed in glutaraldehyde-paraformaldehyde; a second block was fixed in 10% formaldehyde. In all cases septate junctions between digestive vacuoles did occur in phagocytic cells and some adrenal cortex cells fixed in formaldehyde. These junctions were similar to those reported recently for malakoplakia phagocytes. Consistently, they were not found to attach organelles other than lysosomes derivatives. Both phagocytes and adrenal cortex cells in the material fixed in glutaraldehyde-paraformaldehyde did not display adhesive specializations between digestive vacuoles. This suggests that the septate junctions described herein are artifactuous structures induced by formaldehyde. There is, however, a certain degree of specificity of cells having the capability of developing these septate junctions. It is assumed that the coating material of digestive organelles in phogocytes and some other cells would be responsible for both cell specificity and organelle specificity of the formaldehyde-induced septate junctions. 相似文献

Human erythrocytes are able to incorporate cyclic AMP (cAMP) in amounts larger than those required to saturate cAMP-dependent protein kinase. In contrast to previous observations in avian red blood cells in which cAMP stimulates the Na+/K+ cotransport system, we demonstrate that cAMP inhibits this system in human erythrocytes. The cotransport inhibition is enhanced by addition of phosphodiesterase inhibitor 1-methyl-3-isobutylxanthine to the incubation medium. The cAMP concentration giving half-maximal cotransport inhibition showed a wide variation among different individuals (from 0.1 to 5 mM external cAMP concentration). In contrast to cAMP, cyclic GMP showed little effect on the cotransport system. Ca2+ introduced into the cell interior was an inhibitor of the Na+/K+ cotransport system. These results suggest that in human cells in which endogeneous levels of cAMP and Ca2+ are modulated by hormones, the Na+/K+ cotransport system may be under hormonal regulation. 相似文献

Fruit distribution among individuals in populations of three orchid species was analyzed. Fruit set was low in the three species. The distribution of fruits in the non-autogamous species was skewed to the right, with high variance: mean ratios, and a moderate to high proportion of fruitless plants. Larger inflorescence size did not necessarily result in an increase in percent fruit set but tended to increase the probability to produce at least one fruit. Pollinator-mediated selection on inflorescence size through fruit production may be limited by the low overall level of visitation and the resulting uncertainty of pollination at the individual level. 相似文献

Volcanic eruptions play an important role in vegetation dynamics and its historical range of variability. However, large events are infrequent and eruptions with a significant imprint in today''s vegetation occurred far in the past, limiting our understanding of ecological processes. Volcanoes in southern Andes have been active during the last 10 ka and support unique ecosystems such as the Araucaria–Nothofagus forest. Araucaria is an endangered species, with a fragmented distribution and well‐adapted to fire and volcanic disturbances. Yet, it was suggested that volcanism might have increased the fragmentation. Through the use of pollen and tephra analysis from a sedimentary record, this paleoecological study aims to provide an insight into the vegetation responses to past volcanic disturbances, to assess the role of volcanic disturbance on the vegetation dynamics and to determine if the current fragmentation has been caused by volcanism. Results show that during the last 9 kyr, 39 tephra falls buried the vegetation around Lake Relem, more frequently between 4 and 2 ka. The pollen percentage indicates that the vegetation changed after small tephra fall but seldom caused significant changes. However, the large eruption of Sollipulli volcano (~3 ka) changed the environmental conditions affecting severely the vegetation. Ephedra dominated the early successional stage, perhaps facilitating Nothofagus recovering after ~500 years. Slight increase of Araucaria and Nothofagus obliqua‐type pollen percentages suggests that forest resisted without permanent changes and recovered relatively fast after the large eruption, perhaps because of sparse biological legacies distributed in the landscape. In the study area, the relative stability of Araucaria pollen after several tephra fall suggests no change in its past distribution at the current forest‐steppe ecotone, thus not affecting its current conservation status. Perhaps, random factors, the colonization patterns of the high elevations in the Andes after deglaciation and topography might play a more important role than previously thought. 相似文献

Stream discharge?Cconcentration relationships are indicators of terrestrial ecosystem function. Throughout the Amazon and Cerrado regions of Brazil rapid changes in land use and land cover may be altering these hydrochemical relationships. The current analysis focuses on factors controlling the discharge?Ccalcium (Ca) concentration relationship since previous research in these regions has demonstrated both positive and negative slopes in linear log10discharge?Clog10Ca concentration regressions. The objective of the current study was to evaluate factors controlling stream discharge?CCa concentration relationships including year, season, stream order, vegetation cover, land use, and soil classification. It was hypothesized that land use and soil class are the most critical attributes controlling discharge?CCa concentration relationships. A multilevel, linear regression approach was utilized with data from 28 streams throughout Brazil. These streams come from three distinct regions and varied broadly in watershed size (<1 to >106 ha) and discharge (10?5.7?C103.2 m3 s?1). Linear regressions of log10Ca versus log10discharge in 13 streams have a preponderance of negative slopes with only two streams having significant positive slopes. An ANOVA decomposition suggests the effect of discharge on Ca concentration is large but variable. Vegetation cover, which incorporates aspects of land use, explains the largest proportion of the variance in the effect of discharge on Ca followed by season and year. In contrast, stream order, land use, and soil class explain most of the variation in stream Ca concentration. In the current data set, soil class, which is related to lithology, has an important effect on Ca concentration but land use, likely through its effect on runoff concentration and hydrology, has a greater effect on discharge?Cconcentration relationships. 相似文献

Digital radiographic imaging is increasing in veterinary practice. The use of radiation demands responsibility to maintain high image quality. Low doses are necessary because workers are requested to restrain the animal. Optimizing digital systems is necessary to avoid unnecessary exposure, causing the phenomenon known as dose creep. Homogeneous phantoms are widely used to optimize image quality and dose. We developed an automatic computational methodology to classify and quantify tissues (i.e., lung tissue, adipose tissue, muscle tissue, and bone) in canine chest computed tomography exams. The thickness of each tissue was converted to simulator materials (i.e., Lucite, aluminum, and air). Dogs were separated into groups of 20 animals each according to weight. Mean weights were 6.5 ± 2.0 kg, 15.0 ± 5.0 kg, 32.0 ± 5.5 kg, and 50.0 ± 12.0 kg, for the small, medium, large, and giant groups, respectively. The one-way analysis of variance revealed significant differences in all simulator material thicknesses (p < 0.05) quantified between groups. As a result, four phantoms were constructed for dorsoventral and lateral views. In conclusion, the present methodology allows the development of phantoms of the canine chest and possibly other body regions and/or animals. The proposed phantom is a practical tool that may be employed in future work to optimize veterinary X-ray procedures. 相似文献

Vaccine adjuvants are substances associated with antigens that are fundamental to the formation of an intense, durable, and fast immune response. In this context, the use of vaccine adjuvants to generate an effective cellular immune response is crucial for the design and development of vaccines against visceral leishmaniasis. The objective of this study was to evaluate innate inflammatory response induced by the vaccine adjuvants saponin (SAP), incomplete Freund's adjuvant (IFA), and monophosphoryl lipid A (MPL). After a single dose of adjuvant was injected into the skin of mice, we analyzed inflammatory reaction, selective cell migration, and cytokine production at the injection site, and inflammatory cell influx in the peripheral blood. We found that all vaccine adjuvants were able to promote cell recruitment to the site without tissue damage. In addition, they induced selective migration of neutrophils, macrophages, and lymphocytes. The influx of neutrophils was notable at 12 h in all groups, but at other time points it was most evident after inoculation with SAP. With regard to cytokines, the SAP led to production of interleukin (IL)-2, IL-6, and IL-4. IFA promoted production of tumor necrosis factor (TNF)-α, interferon (IFN)-γ, IL-6, IL-17, IL-4, and IL-10. We also observed that MPL induced high production of IL-2, TNF-α, and IFN-γ, in addition to IL-6, IL-17, and IL-10. In peripheral blood, values of certain cell populations in the local response changed after stimulation. Our data demonstrate that the three vaccine adjuvants stimulate the early events of innate immune response at the injection site, suggesting their ability to increase the immunogenicity of co-administered antigens.
