Proton transport on water wires, of interest for many problems in membrane biology, is analyzed in side-chain analogs of gramicidin A channels. In symmetrical 0.1 N HCl solutions, fluorination of channel Trp(11), Trp-(13), or Trp(15) side chains is found to inhibit proton transport, and replacement of one or more Trps with Phe enhances proton transport, the opposite of the effects on K(+) transport in lecithin bilayers. The current-voltage relations are superlinear, indicating that some membrane field-dependent process is rate limiting. The interfacial dipole effects are usually assumed to affect the rate of cation translocation across the channel. For proton conductance, however, water reorientation after proton translocation is anticipated to be rate limiting. We propose that the findings reported here are most readily interpreted as the result of dipole-dipole interactions between channel waters and polar side chains or lipid headgroups. In particular, if reorientation of the water column begins with the water nearest the channel exit, this hypothesis explains the negative impact of fluorination and the positive impact of headgroup dipole on proton conductance.  相似文献

The Bay of Santander constitutes one of the most important estuarine areas in northern Spain (Gulf of Biscay) which has been significantly stressed by sewage discharges. However, an improvement in the environmental quality of the bay is expected once the new sewer system comes into operation. The evaluation of such an evolution needs to be contrasted with the current situation, established in specific baseline studies. For this purpose, a field study was carried out on the soft bottom assemblages of the estuary in spring of 1998. At each station, the benthic fauna abundance, grain size, organic matter, carbon, nitrogen, phosphorus and heavy metals concentrations were analysed. The community structure and the physical and chemical characteristics of the sediment reflected the higher impacts both near the points of discharges and in the low water exchange zones, whereas those areas showing higher tidal flows presented an acceptable quality. High heavy metal concentrations for Cd, Hg, Pb and Zn indicated contamination of industrial sources in several areas of the Bay. This information allows to establish a reference situation in order to improve the design of a specific monitoring program for this area.  相似文献

Changes in the global environment are modifying the geographical locations of habitats suitable for plant growth. The capacity of plants to migrate to sites of suitable environmental quality will strongly influence future distributions of plant diversity. However, it is not well understood how rates of plant migration are influenced by the habitat loss and habitat fragmentation that characterise contemporary landscapes. In this study we develop a model that can predict migration rates in both intact landscapes (potential migration rate) and in fragmented landscapes (realised migration rates). Migration rates in fragmented landscapes might be slower for many reasons. In this study we focus on two, non‐exclusive reasons. First, the processes that move seeds may break down in fragmented landscapes causing seeds to be dispersed shorter distances. Second, in fragmented landscapes some proportion of seeds will not be deposited in habitats suitable for recruitment. We describe the breakdown of dispersal processes as a competing risk between the factors influencing dispersal in intact landscapes and the factors that may disrupt dispersal processes in fragmented landscapes. We show how the parameters that influence dispersal in fragmented landscapes can be estimated, and how these estimates can be used to forecast migration rates using an integrodifference equation (IDE). The forecasts of the IDE described the effects of reduced dispersal distances adequately. However, the IDE produced biased estimates of the effects of a reduction in plant habitat on migration rates. Model analyses showed that, although we can expect realised migration rates to be lower than potential migration rates, we can also expect the sensitivity of migration rate to habitat loss to vary. In addition, simulations showed that the qualitative nature of the responses of migration rate to habitat loss were variable – some model species responded non‐linearly to habitat loss, others responded linearly. While our method provides guidelines for empirical data collection and model parameterisation, we recognise that obtaining these data will be challenging.  相似文献
