Three different earthworm species Eisenia fetida, Eudrilus eugeniae and Perionyx excavatus in individual (Monocultures) and combinations (Polycultures) were utilized to compare the suitability of worm species for vermicomposting of filter mud as well as the quality of the end product. The filter mud blended with saw dust can be directly converted into good quality fertilizer (vermicompost). Eight different reactors including three monocultures and four polycultures of E. fetida, E. eugeniae and P. excavatus and one control were used for the experiment. Vermicomposting resulted in significant reduction in C/N ratio, pH, total organic matter (TOC) but increase in electrical conductivity (EC), total nitrogen (TN), total phosphorus (TP) and macronutrients (K, Ca and Na). Oxygen uptake rate (OUR) dropped up to 1.64–1.95 mg/g (volatile solids) VS/day for monoculture reactors and 1.45–1.78 mg/g VS/day for polycultures reactors, respectively, after 45 days of vermicomposting. Cocoon production and the earthworm biomass increased as vermicomposting progressed. On an overall the mono as well as polyculture reactors produced high quality stable compost free from pathogens and no specific differentiation could be inferred between the reactors. 相似文献

Opercular and surfacing activity were observed in the Indian catfish Clarias batrachus and Heteropneustes fossilis . In the series A experiments, the opercular activity was monitored under two experimental conditions, viz., (1) surfacing allowed and (2) surfacing prevented. A statistically significant elevation of the rate of opercular activity was observed when air breathing was prevented in both species. In addition, a significant prevention effect and a time of day dependence of that were noticed in both species. In the series B experiments, temporal patterns of surfacing and air-gulping activities were examined under an artificial LD 12:12 schedule at 2-h intervals over a period of 48 h. The inter air-gulping interval in minutes between two consecutive bouts was also recorded four times each day in both species. A significant 24-h rhythm was found for the rate of surfacing activity and length of the inter air-gulping interval in both species. However, the overall activity appears to be much higher in Heteropneustes fossilis as compared to Clarias batrachus . 相似文献

Vaccination is the attempt to mimic certain aspects of an infection for the purpose of causing an immune response that will protect the individual from that infection. Malaria, a disease responsible for immense human suffering, is caused by infection with Plasmodium spp. parasites, which have a very complex life cycle--antigenically unique stages infect different tissues of the body. It is a parasitic disease for which no successful vaccine has been developed so far, despite considerable efforts to develop a subunit vaccine that offers protective immunity. Due to the spread of drug-resistant malaria, efforts to develop an effective vaccine have become increasingly critical. DNA vaccination provides a stable and long-lived source of protein vaccine capable of inducing both antibody- and cell-mediated immune responses to a wide variety of antigens. Injected DNA enters the cells of the host and makes the protein, which triggers the immune response. According to present needs, the flexibility of DNA vaccine technology permits the combination of multiple antigens from both the preerythrocytic and erythrocytic stages of malaria parasite. DNA vaccines with genes coding for different antigenic parts of malaria proteins have been created and presently some of these are undergoing field trials. The results from these trials will help to determine the likelihood of success of this technology in humans. The entire coding sequence of the bi-functional enzyme, Δ1-Pyrroline-5-carboxylate synthetase (P5CS) from Arabidopsis thaliana was reverse-transcribed, amplified and expressed under the control of CaMV 35S promoter in transgenic tobacco plants. Several lines were established and tested for the expression of P5CS. Drought and salinity were applied as osmotic stresses and proline content of the transformed plants was compared with that of non-transformed controls. Results indicate that transgenic lines express higher levels of proline and show enhanced resistance to the applied osmotic stress as compared to the non-transgenic plants. 相似文献

So far, it has been difficult to generate embryonic stem (ES) cell from early stage preimplantation embryos of buffalo. These ES cells will be more helpful for efficient embryo cloning and generation of body cells as they are more primitive than inner cell mass (ICM)-derived ES cells. The present study was conducted to find the effect of lipopolysaccharide (LPS), melatonin (N-acetyl-5-methoxytryptamine, a pineal gland product), and citral (3,7-dimethyl-2,6-octadienal and a retinoic acid synthesis blocker) on establishment of primary ES cell colonies, the comparative size of the ES cell colonies, and expression of pluripotent genes during extended period of culture in buffalo. Zona-free eight-cell stage in vitro fertilization (IVF) embryos were cultured in ES cell medium supplemented with none (media I as control), LPS (media II), citral melatonin (media III), or melatonin (media IV). The multiplication of blastomere leading to ES cell colony formation and expression of pluripotent genes were assessed up to day 20 of culture. The primary colony formation, the comparative size of the ES cell colonies, and expression of pluripotent genes in these colonies were better in the medium supplemented with melatonin in all days of culture. Within melatonin supplementation, the colony size was comparatively larger on day 8 and day 12 of culture. Further, with this supplementation, the Oct-4 and Nanog expression was comparatively higher on all days of culture. The results indicated that supplementation of melatonin helped in the formation of better primary ES cell colony as well as in the maintenance of pluripotency. The results also indicated that primary colonies developed on day 8 to day 12 of culture may be better for passaging them for establishment of ES cell line from early stage preimplantation IVF embryos of in buffalo. 相似文献
