The binding of certain growth factors and cytokines to components of the extracellular matrix can regulate their local availability and modulate their biological activities. We show that oncostatin M (OSM), a profibrogenic cytokine and modulator of cancer cell proliferation, specifically binds to collagen types I, III, IV, and VI, immobilized on polystyrene or nitrocellulose. Single collagen chains inhibit these interactions in a dose-dependent manner. Cross-inhibition experiments of collagen-derived peptides point to a limited set of OSM-binding collagenous consensus sequences. Furthermore, this interaction is found for OSM but not for other interleukin-6 type cytokines. OSM binding to collagens is saturable, with dissociation constants around 10(-8) m and estimated molar ratios of 1-3 molecules of OSM bound to one molecule of triple helical collagen. Furthermore, collagen-bound OSM is biologically active and able to inhibit proliferation of A375 melanoma cells. We conclude that abundant interstitial collagens dictate the spatial pattern of bioavailable OSM. This interaction could be exploited for devising collagenous peptide-antagonists that modulate OSM bioactivity in tumor growth and fibrotic disorders like rheumatoid arthritis and hepatic fibrosis. 相似文献

To investigate how plant diversity loss affects nitrogen accumulation in above‐ground plant biomass and how consistent patterns are across sites of different climatic and soil conditions, we varied the number of plant species and functional groups (grasses, herbs and legumes) in experimental grassland communities across seven European experimental sites (Switzerland, Germany, Ireland, United Kingdom (Silwood Park), Portugal, Sweden and Greece). Nitrogen pools were significantly affected by both plant diversity and community composition. Two years after sowing, nitrogen pools in Germany and Switzerland strongly increased in the presence of legumes. Legume effects on nitrogen pools were less pronounced at the Swedish, Irish and Portuguese site. In Greece and UK there were no legume effects. Nitrogen concentration in total above‐ground biomass was quite invariable at 1.66±0.03% across all sites and diversity treatments. Thus, the presence of legumes had a positive effect on nitrogen pools by significantly increasing above‐ground biomass, i.e. by increases in vegetation quantity rather than quality. At the German site with the strongest legume effect on nitrogen pools and biomass, nitrogen that was fixed symbiotically by legumes was transferred to the other plant functional groups (grasses and herbs) but varied depending on the particular legume species fixing N and the non‐legume species taking it up. Nitrogen‐fixation by legumes therefore appeared to be one of the major functional traits of species that influenced nitrogen accumulation and biomass production, although effects varied among sites and legume species. This study demonstrates that the consequences of species loss on the nitrogen budget of plant communities may be more severe if legume species are lost. However, our data indicate that legume species differ in their N2 fixation. Therefore, loss of an efficient N2‐fixer (Trifolium in our study) may have a greater influence on the ecosystem function than loss of a less efficient species (Lotus in our study). Furthermore, there is indication that P availability in the soil facilitates the legume effect on biomass production and biomass nitrogen accumulation. 相似文献

Malate occupies a central role in plant metabolism. Its importance in plant mineral nutrition is reflected by the role it plays in symbiotic nitrogen fixation, phosphorus acquisition, and aluminum tolerance. In nitrogen-fixing root nodules, malate is the primary substrate for bacteroid respiration, thus fueling nitrogenase. Malate also provides the carbon skeletons for assimilation of fixed nitrogen into amino acids. During phosphorus deficiency, malate is frequently secreted from roots to release unavailable forms of phosphorus. Malate is also involved with plant adaptation to aluminum toxicity. To define the genetic and biochemical regulation of malate formation in plant nutrition we have isolated and characterized genes involved in malate metabolism from nitrogen-fixing root nodules of alfalfa and those involved in organic acid excretion from phosphorus-deficient proteoid roots of white lupin. Moreover, we have overexpressed malate dehydrogenase in alfalfa in attempts to improve nutrient acquisition. Exposure to metals at workplaces is well known and in many cases occupational studies led to an
adoption of limit values. For airborne concentrations of substances as metals refer to the “Maximaleo
Arbeitsplatz-Konzentration” (MAK) in Germany or the “Threshold Limit Value” (TLV) in USA. Biological
monitoring consists of an assessment of overall exposure to chemicals at the workplace and in the
environment. The “Biologischer Arbeitsstoff Toleranzwert” (BAT) in Germany and the “Biological Exposure
Index” in the USA serve as reference values. Besides these occupational limit values, reference values exist
in Germany for the background exposure of the non occupationally exposed general population. In some
cases the reference values are exceeded without any occupational exposure. Several cases of unusual
environmental exposure to cobalt, mercury and manganese are reported. In such cases, it is often difficult to
evaluate the measured concentration. In Germany, therefore, the “Human-Biomonitoring-Werte” (HBMValues)
have been adopted in order to evaluate such high background exposures. The HBM-concept is
presented. Environmental exposure to metals is usual within some limits. Reference values are helpful for an
assessment. Unusual exposure occurs and the physician should be alert to symptoms of poisoning. 相似文献

New analogues of triscatecholate siderophores based on linear or tripodal triamines with or without spacer groups or lipophilic and hydrophilic substituents were synthesized. The catecholate moieties were prepared in OH-forms, as acetylated compounds or masked as 8-methoxycarbonyloxy-2,4-dioxo-1,3-benzoxazine derivatives. Some of the new compounds were active as siderophores tested by growth promotion assays using various Gram-negative bacteria and mycobacteria under iron limitation and by CAS-assay. To assess the influence of bacteria inoculation on carbon flow through maize plant and rhizosphere,14C allocation after14CO2 application to shoots over a 5-day period was determined. Plants were grown on C- and N-free quartz sand in two-compartment
pots, separating root and shoot space. While one treatment remained uninoculated, treatments two and three were inoculated
withPantoea agglomerans (D5/23) andPseudomonas fluorescens (Ps I A12), respectively, five days after planting. Bacterial inoculation had profound impacts on carbon distribution within
the system. Root/rhizosphere respiration was increased and more carbon was allocated to roots of plants being inoculated.
After five days of14CO2 application, more ethanol-soluble substances were found in roots of inoculated treatments and lower rhizodeposition indicated
intensive C turnover in the rhizosphere. In both inoculated treatments the intensity of photosynthesis measured as net-CO2-assimilation rates were increased when compared to the uninoculated plants. However, high C turnover in the rhizosphere reduced
shoot growth of D5/23 inoculated plants, with no effect on shoot growth of Ps I A12 inoculated plants. A separation of labeled
compounds in roots and rhizodeposition revealed that neutral substances (sugars) constituted the largest fraction. The relative
fractions of sugars, amino acids and organic acids in roots and rhizodeposition suggest that amino acid exudation was particularly
stimulated by bacterial inoculation and that turnover of this substance group is high in the rhizosphere. 相似文献

Fusobacterium nucleatum (Fn) is a Gram‐negative oral commensal, prevalent in various human diseases. It is unknown how this common commensal converts to a rampant pathogen. We report that Fn secretes an adhesin (FadA) with amyloid properties via a Fap2‐like autotransporter to enhance its virulence. The extracellular FadA binds Congo Red, Thioflavin‐T, and antibodies raised against human amyloid β42. Fn produces amyloid‐like FadA under stress and disease conditions, but not in healthy sites or tissues. It functions as a scaffold for biofilm formation, confers acid tolerance, and mediates Fn binding to host cells. Furthermore, amyloid‐like FadA induces periodontal bone loss and promotes CRC progression in mice, with virulence attenuated by amyloid‐binding compounds. The uncleaved signal peptide of FadA is required for the formation and stability of mature amyloid FadA fibrils. We propose a model in which hydrophobic signal peptides serve as “hooks” to crosslink neighboring FadA filaments to form a stable amyloid‐like structure. Our study provides a potential mechanistic link between periodontal disease and CRC and suggests anti‐amyloid therapies as possible interventions for Fn‐mediated disease processes. 相似文献
