The behaviour of gastrin (G) cells and somatostatin (D) cells in endoscopic antral biopsies and that of intraluminal gastrin (ILG) and somatostatin (ILS) release in the gastric juice were investigated in three groups of patients: control subjects, duodenal ulcer (DU) patients and DU patients treated by a superselective vagotomy (SSV). G and D cell densities were correlated in the three groups of subjects. The G/D cell ratio was significantly increased in SSV patients (P less than 0.001) as compared to control and DU patients. No correlation was found between gastrin or somatostatin cell densities and basal intraluminal levels of the two peptides. ILG output was significantly higher in DU patients than in control or SSV patients (P less than 0.001). ILS output was also higher in DU patients than in controls (P less than 0.001) and in SSV patients (P less than 0.05). It was also significantly augmented in SSV (P less than 0.001) as compared to control patients. ILG and ILS concentrations were only correlated in controls. Within each of the three groups of subjects, ILG and ILS release varied in function of the gastric juice pH. Our results emphasize the necessity to consider the intragastric pH as well as the physiological or pathological state to study intraluminal peptides in man.  相似文献

巴柑檬是我们第一次从国外引种成功的一种名贵香料植物。用色谱-质谱-计算机联用技术、毛细管气相色谱保留指数法和标准品叠加法分析了水蒸汽蒸馏巴柑檬叶和果皮精油的化学成分。从叶和果皮精油分离出来的220个和200个成分中,分别鉴定出48个和57个成分。鉴定组分的含量分别占叶和果皮精油的99.80%和98.54%。叶精油与果皮精油在化学成分方面的主要区别是叶油中萜烃化合物含量较低,而萜醇类化合物含量较高。  相似文献

To establish cell systems appropriate for investigating the mode of action of antiherpetic nucleoside analogues, mutant cell strains were constructed from murine mammary carcinoma FM3A cells, which were deficient in TK, but were transformed with a recombinant plasmid DNA containing the HSV-2 TK gene. The transformed cells incorporated the viral DNA, expressed viral TK activity and showed unusually high sensitivity to the cytostatic action of the antiherpetic nucleoside analogues ACV and IVDU, both of which were only weakly inhibitory to the growth of the parent cells. Curiously, the FM3A cell strains transformed with HSV-2 TK gene showed a higher sensitivity to ACV and IVDU than the previously established cell line transformed with HSV-1 TK gene. This contrasts with the inhibitory effects of ACV and IVDU on acute HSV infection, since HSV-2 infection is slightly or considerably less susceptible than HSV-1 infection to inhibition by ACV or IVDU, respectively.  相似文献

Redox titration of the electrochromic carotenoid band shift, detected at 50 μs after a saturating actinic flash, in spinach chloroplasts, shows that only one electron acceptor in Photosystem II participates in a transmembrane primary electron transfer. This species, the primary quinone acceptor, Q, shows only one midpoint potential (Em,7.5) of approx. 0 V and is undoubtedly equivalent to the fluorescence quencher, QH. A second titration wave is observed at low potential (Em,7.5 ? ? 240 mV) and at greater than 3 ms after a saturating actinic flash. This wave has an action spectrum different from that of Photosystem II centers containing Q and could arise from a secondary but not primary electron transfer. A low-potential fluorescence quencher is observed in chloroplasts which largely disappears in a single saturating flash at ? 185 mV and which does not participate in a transmembrane electron transfer. This low-potential quencher (probably equivalent to fluorescence quencher, QL) and Q are altogether different species. Redox titration of C550 shows that if electron acceptor Qβ is indeed characterized by an Em,7 of + 120 mV, then this acceptor does not give rise to a C550 signal upon reduction and does not participate in a transmembrane electron transfer. Redox titrations of the flash-induced formation of C550 (a linear indicator of Q?) were performed between pH 5.9 and 8.3 in Chlamydomonas Photosystem II particles lacking the secondary electron acceptor, B. One-third of the reaction centers show a pH-dependent midpoint potential (Em,7.5) = ? 30 mV) for redox couple QQ?, which varies by ?60 mV/pH unit. Two-thirds of the centers show a pH-independent midpoint potential (Emm = + 10 mV) for this couple. The elevated pH-independent Em suggests that in the latter centers the environment of Q has been modified such as to stabilize the semiquinone anion, Q?. The midpoint potentials of the centers having a pH-dependent Em are within 20 mV of those observed in chloroplasts having a secondary electron acceptor. It appears therefore that the secondary electron acceptor exerts little influence on the Em of QQ?. An EPR signal at g 1.82 has recently been attributed to a semiquinone-iron complex which comprises Q?. The similar redox behavior reported here for C550 and reported by others (Evans, M.C.W., Nugent, J.H.A., Tilling, L.A. and Atkinson, Y.E. (1982) FEBS Lett. 145, 176–178) for the g 1.82 signal in similar Photosystem II particles confirm the assignment of this EPR signal to Q?. At below ?200 mV, illumination of the Photosystem II particles produces an accumulation of reduced pheophytin (Ph?). At ?420 mV Ph? appears with a quantum yield of 0.006–0.01 which in this material implies a lifetime of 30–100 ns for the radical pair P-680+Ph?.  相似文献

Summary Calorimetric estimates of the utilization efficiency of the free-energy derived from substrate oxidation by cell suspensions of two nitrifying bacteria, Nitrosomonas and Nitrobacter, provided two ranges of values: 11 to 27% and 15 to 51%, respectively. About 15 to 30% of the utilized free-energy is used for driving endergonic reactions other than CO2 fixation, probably the synthesis of polyphosphates.The molar heat of substrate oxidation does not seem to be influenced by the age of cells harvested during growth or by the length of the incubation period during which cells have been kept in a buffer suspension in a starved condition. The loss of respiratory activity measured either by oxygen uptake or heat evolution in the presence of the specific substrate, nitrite or ammonium, decreases according to kinetics which are influenced by the aerobiosis of the suspension. The viability of the starved cells decreases in a way which is similar to that of the respiratory activity. It seemed impossible to obtain cells which had lost their viability but kept the ability to oxidize their substrate.Two inhibitors of the respiratory chain, quinacrine and cyanide, are without effect on the molar heat of substrate oxidation and consequently on the free-energy utilization efficiency. 2.4 dinitrophenol did decrease the rate of heat evolution during substrate oxidation at concentrations at which the rate of oxygen uptake was not depressed, with the consequences that free-energy efficiency was apparently increased.  相似文献

Conclusions La Provence apparaît, en définitive, comme un territoire des plus variés aux divers points de vue climatique, édaphique, biotique.C'est une région d'autant plus favorable aux études phytosociologiques que la variété des milieux s'y double d'une brutalité souvent étonnante des contrastes, conduisant à la juxtaposition parfois extrêmement précise d'associations végétales très différentes, dont les limites peuvent alors être cartographiquement figurées avec une très suffisante exactitude.Cette vigueur des contrastes est surtout marquée en Provence calcaire où les divers termes des séries évolutives dérivant du Quercetum ilicis climacique ou y conduisant, se juxtaposent souvent sans transition de quelque importance.Les contrastes sont cependant moins accusés en Provence cristalline et dans la Basse-Vallée du Rhône.En Crau comme en Camargue, les associations végétales se juxtaposent souvent en ceintures parallèles ou concentriques — elles s'individualisent alors facilement —, souvent en mosaïques irrégulières, au grè des variations locales des deux facteurs déterminants: l'eau en Crau, le sel en Camargue.En Provence cristalline, les Maures étant surtout faites de roches se délitant facilement (grès, micaschistes et phyllades) sous un climat chaud et humide, le sol se reconstitue facilement après sa destruction partielle par l'érosion; les contrastes sont moins tranchés; on observe des transitions nombreuses entre les divers termes des séries évolutives, dont les limites ne peuvent être fixées qu'approximativement. Nous y avons notamment abordé le problème de la forêt de chêne-liège dont la signification est loin d'être connue, non seulement en Provence mais sur toute l'étendue de son aire, en Afrique du Nord, en Espagne, au Portugal, en France méridionale, en Italie et dans les Iles de la Méditerranée occidentale.En provence cristalline, il est certain que l'homme en a considérablement assuré l'extension, directement pour l'utilisation du liège, indirectement par la coupe et le feu, et, en bien des points de la Méditerranée occidentale, le chêne-liège se présente comme une essence cultivée.Par ailleurs rien ne permet de mettre en doute son indigénat. Mais, dans les conditions actuelles du milieu en Provence cristalline:le chêne vert est possible partout;dans les conditions naturelles de la concurrence vitale le chêne vert l'emporte sur le chêne-liège.Si, donc, la forêt originelle a comporté le chêne-liège, ce ne peut être qu'à l'état d'essence secondaire et seulement en quelques points où une faible couverture forestière lui permettait de cohabiter avec le chêne vert.Les documents cartographiques au I/20 000e tendent à montrer, d'aileurs que, dans les conditions naturelles de reconstitution de la forêt ou dans le peuplement naturel des sols neufs (Isthme de Giens), c'est la chênaie de chêne vert, non la chênaie de chêne-liège qui s'installe.Les grandes unités phytosociologiques de la Provence paraissent être maintenant convenablement définies floristiquement; il convient donc d'en poursuivre l'étude écologique, dont certains aspects seulement les plus apparents—mais peut-être pas toujours les plus actifs-, ont été abordés.Notons enfin la démonstration faite, au cours de l'excursion, de la parfaite application possible des méthodes phytosociologiques à l'étude des groupements marins et la nécessité, particulièrement apparente en milieu marin, de considérer tout autant les animaux que les végétaux. L'avenir est bien aux biocénoses dont les grands traits ont été mis en évidence, quant à la côte de la presqu'île de Giens, par Roger Molinier.Reçu par la rédaction le 20. Regulation of glycogenolysis in human skeletal muscle   总被引:7,自引:0,他引:7  
The role of inorganic phosphate on the regulation of glycogenolysis in resting and contracting muscle was studied in human quadriceps muscle. Increased Pi content was achieved by intermittent electrical stimulation of the muscle followed by occlusion of the blood flow. Occlusion resulted in the maintenance of a high Pi content over a 60-s observation period during which the muscle was either at rest or was stimulated electrically. The study was performed with and without infusion of epinephrine (EPI). In the absence of EPI the phosphorylase a fraction was 50% immediately at the end of the initial stimulation period, declining to 22% after 60 s. With EPI corresponding values for phosphorylase a were 91% initially, 56% after 30 s, and 33% after 60 s, respectively. In both cases the Pi content was increased by approximately 35 mmol/kg dry muscle during the stimulation and remained constant during the occlusion. In neither of these situations was significant degradation of glycogen observed during the occlusion. In the study performed with electrical stimulation during the occlusion period, muscle glycogen degradation was observed both with and without EPI. Phosphorylase a fractions and Pi contents in this study were similar to those observed when muscle was rested over the 60-s occlusion period. The paradox of a high Pi content and extensive transformation of phosphorylase to the a form but low glycogenolytic activity points to additional factors in the regulation of glycogen breakdown.  相似文献

用改良的Giemsa C-带技术以单株为基础分析了八倍体小黑麦×普通小麦的杂种BC_1,F_(?)和F_(?)代植株的核型。在鉴定了C-带核型的1098株杂种后代植株中,发现了78条小麦-黑麦和277条黑麦-黑麦易位染色体。在不同的世代和株系中,小麦-黑麦染色体易位率变化在4.35—14.07%之间,平均7.10%;黑麦-黑麦染色体易位率在0.48—52.78%之间,平均25.23%。鉴定的小麦-黑麦易位染色体涉及了黑麦的14条不同的染色体臂和小麦的A、B和D组染色体。易位的48.57%发生在小麦和黑麦的部分同源染色体之间,51.43%发生在非部分同源染色体之间。不同的黑麦染色体臂参与易位的频率不同。小麦-黑麦染色体易位主要发生在杂种的早期世代,使用适当的选择技术在F_3获得了纯合的易位植株。文中讨论了快速选育易位系的技术和它们在小麦育种中的应用问题。  相似文献
