Mycophenolic acid (MPA) can be produced in solid state fermentation. An isolate of Penicillium brevi-compactum ATCC 16024 grown on moist wheat bran produced a titre of 425 mg per kg of wheat bran. Central composite rotatable design
and response surface methodology were employed to derive a statistical model for media optimization towards production of
mycophenolic acid. Five levels with a five factorial design were adopted. The correlation coefficient was 0.82, ensuring a
satisfactory adjustment of the model to the experimental values. This statistical design was very effective in improving the
titre of mycophenolic acid up to 3286 mg per kg of wheat bran.

Summary The sclerotial actinomycete Chainia (NCL 82-5-1) secreted extracellular xylanase in submerged culture in media containing yeast extract and wheat bran or commercial xylan. A high activity (28 IU/ml) of xylanase was obtained in 72 h on a medium containing 3% xylan. Only a single species of xylanase (i.e. without isoenzymes) was detected by polyacrylamide gel electrophoresis. It had an optimum pH of 5.0 and optimum temperature of 65°C. It was stable at pH 6.0 to heating at 60°C for 10 min. Its pI was 8.0 and the Km was 0.4%. The results are discussed in relation to xylanase reported from actinomycetes such as Streptomyces xylophagus. 相似文献

Inhibitors of the thiol proteinase, papain (EC 3.4.22.2), were shown to be present in 11 species of 10 genera of plants. The inhibitor activity was nondialyzable, and precipitated by ammonium sulfate. Tissue cultures from a number of plant genera consisting of rapidly dividing cells contained latent papain inhibitor that could be activated upon heating. Four isoinhibitors of plant thiol proteinases from seeds of the legume Vigna unguiculata subsp. cyclindrica were purified to apparent homogeneity by acrylamide gel electrophoresis with or without sodium dodecyl sulfate. The inhibitors were present in very small amounts compared to the trypsin inhibitors and the degree of purification of the homogeneous isoinhibitors on the assumption that all were present initially in equal amounts was 15,000- to 60,000-fold. The isoinhibitors did not inhibit pepsin, bromelain, and the serine proteinases, trypsin, chymotrypsin, and subtilisin. They were specific for papain, chymopapain, and ficin but their inhibition of the proteinase, esterase, and amidase activities of the three enzymes differed. 相似文献

Background
Although Morbillivirus and Toxoplasma gondii have emerged as important pathogens for several cetaceans populations over the last 20 years, they have never been identified together in a Mysticete. In particular, morbilliviral infection has been never described in the Mediterranean fin whale population.Case presentation
On January 2011 an adult male of fin whale (Balaenoptera physalus) stranded along the Tyrrhenian coastline of Italy. During necropsy, tissue samples from heart, skeletal muscle, mesenteric lymph nodes, liver, spleen, lung, and kidney were collected and subsequently analyzed for Morbillivirus and Toxoplasma gondii by microscopic and molecular methods. Following the detailed necropsy carried out on this whale, molecular analysis revealed, for the first time, the simultaneous presence of a Dolphin Morbillivirus (DMV) and T. gondii infection coexisting with each other, along with high organochlorine pollutant concentrations, with special reference to DDT.Conclusion
This report, besides confirming the possibility for Mysticetes to be infected with DMV, highlights the risk of toxoplasmosis in sea water for mammals, already immunodepressed by concurrent factors as infections and environmental contaminants. 相似文献35.

Applications in the dissociation of cells for primary cell cultures, maintenance of cell lines and the production of G-bands on metaphase chromosomes are described. Summary An alkalotolerant Cephalosporium (NCL 87.11.9) strain capable of rapid growth and xylanase secretion over a wide pH range (pH 4–10) has been isolated from soil samples. When grown in shake flasks on a wheat bran, yeast extract medium for 96 h it produced 15 to 18 IU/ml. The novel feature of this study is that it is the first report of an extracellular fungal xylanase which is active and stable at high alkaline pH (8–9.5). The culture filtrate did not show any significant cellulase activity. Gel filtration studies indicated two peaks of xylanase activities corresponding to molecular weights of 70,000 and 30,000 in the proportion of 10:90. 相似文献
