1. A protein, aggregating at body temperature and solubilizing when cooled, was isolated from fresh human milk at neutral pH and studied for some of its physical, chemical and immunological properties. The name ;galactothermin' is proposed for this protein. 2. Isolation and purification of galactothermin involved casein removal from skim milk at pH4.64 followed by centrifugal fractionation of residual protein-containing solutions repeatedly heated and cooled between 40 degrees C and 0 degrees C at pH7.3. 3. The molecular weight by ultracentrifugal analysis and the minimum molecular weight by sum of amino acid residues were 11400 and 14000 respectively. The sedimentation coefficient s(25,w) was 1.05S and the diffusion coefficient was 7.15x10(-7). Reversible aggregation is favoured by increase in protein concentration, ionic strength, temperature, time and approach to the isoionic point of 7.27 from either acidic or alkaline conditions. 4. Among the amino acid residues, proline predominates and non-polar species account for two-thirds of the total. Cysteine and cystine are absent. Analysis of galactothermin showed it to be essentially free of hexose, sialic acid, calcium and phosphate. 5. Galactothermin is antigenic in the rabbit as evidenced by the passive cutaneous anaphylaxis test. Single precipitin lines are produced in immunodiffusion tests. 6. The first photographs of Cardioceratinae out of exactly defined horizons from the Latest Callovian to the Middle Oxfordian of northern Switzerland are given in this article. Most specimens were either collected in large-scale excavations, or from measured sections. The Cardioceratinae from the Cordatum Subzone are important because they are very numerous, and because particularly well-preserved specimens are abundant. Cardioceratinae of this subzone are uncommon elsewhere in Europe. All representatives of the subfamily from the Cordatum Subzone, macro- as well as microconchs, are grouped into the single genus Cardioceras. An abrupt morphological change has been recognized between the Cardioceratinae of the Densiplicatum Zone and those of the Transversarium Zone.Most of the figured specimens have been found in an incomplete sequence of thin or even condensed horizons. These beds are the distal, starved basin facies of a thick, argillaceous, shoaling-up succession in northwestern Switzerland. Sedimentologic and taphonomic methods have been used in order to work out the time sequence of sediments and ammonite faunas in condensed beds, and to arrive at a correct zonation. These methods as well as observations on the distribution of chamosite and glauconite, and the ammonite assemblages, in two coeval horizons lead us to the conclusion that the Cardioceratinae of the Cordatum Subzone in northern Switzerland lived preferentially in water less than about 100 m deep. 相似文献

A technique for the rapid preparation of mitochondria from rat liver is described. Tissue fractionation is performed by a single centrifugation step with a discontinuous Percoll density gradient. Total preparation times of 5--6 min are achieved by using this method. The mitochondrial fraction obtained is relatively free of contaminating organelles, as judged by marker-enzyme activity determinations. Mitochondria isolated by Percoll-density-gradient centrifugation differ from mitochondria obtained by differential centrifugation [Taylor, Prpić, Exton & Bygrave (1980) Biochem. J. 188, 443--450] in that the former exhibit a higher acceptor control ratio and a higher calcium content. Values obtained for the protonmotive force are not significantly different between the two preparations.
