The real-time three-dimensial CSD-analysis was modified for application to the acoustic synaptic regions (acoustic neuropiles) in the migratory locust,Locusta migratoria. The advantages compared with single cell recordings and field potential analysis are discussed. An appropriate conductivity measurement and processing is adapted to this problem. Influences of errors are estimated. Application to the frontal acoustic neuropile of the metathoracic ganglion ofLocusta migratoria leads to first results. In 25 day old rats monocularly deprived by unilateral eyelid suture on postnatal day 10 (MD), [3H]quinuclidinyl benzylate (3H-QNB) binding was significantly reduced in the visual cortex (VC) of both sides, but elevated in both superior colliculi (SC). Muscarinic receptor binding in the frontal cortex (FC), a non-visual brain area, in the lateral geniculate nucleus (LGN), and in the retina was not affected. In 25 day old rats raised in complete darkness from birth (DR) similar changes in3H-QNB binding were found in VC and SC. However, binding levels were also decreased in the FC and significantly increased in the retina. In adult (6 month old) MD and DR rats the differences in3H-QNB binding as compared to age-matched controls had disappeared completely in all visual brain areas studied. Detailed Scatchard analyses indicate that the alterations in the3H-QNB binding were due to changes in receptor number only.This paper is dedicated to Dr. Derek Richter on his seventy-fifth birthday.  相似文献

Summary Developmental profiles of 11 gangliosides, concentration of lipid- and glycoprotein-bound sialic acid, and activity of AChE of the rat and mouse cerebral cortex were followed from the 7th day of gestation to the 21st postnatal day.There are three main changes in ganglioside concentration, which are similar in both species. The first occurs from gestation day 10 until birth: parallel to decreased proliferation, cell migration, and neuroblast differentiation, GM3 and GD3 in mouse cortex and GD3 in the rat's decreases in favor of GQ1b, GT1b, and GD1a.The second occurs from birth until the first postnatal week: Parallel to increased growth and arborization of dendrites and axons as well as synaptogenesis in rats and mice, there is a two-fold rise of GD1a, whereas GQ1b and GT1b remain on a nearly constant level. Concomitantly, GM3 and GD3 decreases. The third period of ganglioside changes starts in the second postnatal week, parallel to onset of myelination, and is characterized by an increase of GM1 in parallel with a decrease of the polysialogangliosides GT1b and GQ1b.  相似文献

  • 1 . In 30 Hochmooren der Bundesrepublik Deutschland werden verschiedene Arten des Sphagnetum magellanici auf ihren Gehalt an Pb, Cd, Cu, Zn und Mn untersucht (Sphagnum magellanicum, Sphagnum rubellum, Polytrichum strictum, Vaccinium oxycoccus).
  • 2 . Während die Sphagnen besonders viel Pb anreichern, zeigt Polytrichum strictum hohe Cu-Gehalte. In Vaccinium oxycoccus treten sehr hohe Mn-Konzentrationen auf.
  • 3 . Von Vaccinium oxycoccus werden Blättchen, Sprosse und Wurzeln getrennt untersucht. Pb, Cd, Cu und Zn reichern sich besonders in den Wurzeln an, Mn dagegen in den Blättchen.
  • 4 . Es werden darüber hinaus vertikale Torfprofile auf ihr Schwermetallverteilungsmuster untersucht. Während sich vor allem Pb und Cd, aber auch Cu und Zn in den unteren Moosabschnitten und in den anschließenden Torfschichten anreichern, ist die Mn-Konzentration an der Polsteroberfläche am höchsten.
  • 5 . In der Schwermetallbelastung der Hochmoore treten deutliche regionale Unterschiede auf. Die höchste Pb-Belastung findet sich in den Hochmooren des Harzes, während die Pb-Belastung in NW-Deutschland, im südlichen Schwarzwald und im westlichen Alpenvorland am geringsten ist.
  • 6 . Beim Schwermetallgehalt in Sphagnum magellanicum bestehen zwischen Pb einerseits und Cd, Cu und Zn andererseits signifikante Korrelationen. Eine Sonderstellung unter den gemessenen Mikronährstoffen nimmt Mn ein.
  • 7 . Die Jahresnettoproduktion von Sphagnum magellanicum wird mit verschiedenen Methoden bestimmt. Sie beträgt im Mittel 3,0 g. dm?2 a?. Daraus lassen sich Immissionsraten für die einzelnen Schwermetalle berechnen, die für Cd und Zn rund die Hälfte der Emission dieser Metalle betragen.

We have studied the fate of olfactory afferents during metamorphic transformation of Drosophila melanogaster. Intracellular labeling of afferents from larval head chemosensilla suggests that the larval antennal lobe may be an olfactory target, whereas tritocerebral and suboesophageal centers are likely targets of gustatory sensilla. Application of monoclonal antibody 22C10 shows that the larval antennal nerve is the precursor of the adult antennal nerve and is used as a centripetal pathway for the adult afferents. Likely guidance cues are larval olfactory afferents that persist during early metamorphosis. P[GAL4] enhancer trap lines are introduced as efficient markers to follow the establishment of adult sensory projections. β-Galactosidase and the bovine TAU protein were used as reporter proteins, and their expression patterns are compared. P[GAL4] lines MT14 and KL116 demonstrate that adult antennal afferents have arrived in the antennal lobe 24 h after pupariation and extend to the contralateral lobe 6 h later. Line MT14 expresses GAL4 mostly in basiconic sensilla and in certain trichoid sensilla, whereas KL116 is specific for trichoid and a small subset of basiconic sensilla. In the antennal lobe, largely complementary subsets of glomeruli are labeled by the two lines, in agreement with the observation that particular types of sensilla project to particular target glomeruli. © 1997 John Wiley & Sons, Inc. DNA oligonucleotides with dA and dU residues can form duplexes with trans d(A · U) base pairing and the sugar-phosphate backbone in a parallel-stranded orientation, as previously established for oligonucleotides with d(A · T) base pairs. The properties of such parallel-stranded DNA (ps-DNA) 25-mer duplexes have been characterized by absorption (uv), CD, ir, and fluorescence spectroscopy, as well as by nuclease sensitivity. Comparisons were made with duplex molecules containing (a) dT in both strands, (b) dU in one strand and dT in the second, and (c) the same base combinations in reference antiparallel-stranded (aps) structures. Thermodynamic analysis revealed that total replacement of deoxythymine by deoxyuridine was accompanied by destabilization of the ps-helix (reduction in Tm by −13°C in 2 mM MgGl2, 10 mM Na-cacodylate). The U-containing ps-helix (U1 · U2) also melted 14°C lower than the corresponding aps-helix under the same ionic conditions; this difference was very close to that observed between ps and aps duplexes with d(A · T) base pairs. Force field minimized structures of the various ps and aps duplexes with either d(A · T) or d(A · U) base pairs ps/aps and dT/dU combinations are presented. An artificial neural network has been developed for the recognition and prediction of transmembrane regions in the amino acid sequences of human integral membrane proteins. It provides an additional prediction method besides the common hydrophobicity analysis by statistical means. Membrane/nonmembrane transition regions are predicted with 92% accuracy in both training and independent test data. The method used for the development of the neural filter is the algorithm of structure evolution. It subjects both the architecture and parameters of the system to a systematical optimization process and carries out local search in the respective structure and parameter spaces. The training technique of incomplete induction as part of the structure evolution provides for a comparatively general solution of the problem that is described by input-output relations only. Seven physicochemical side-chain properties were used to encode the amino acid sequences. It was found that geometric parameters like side-chain volume, bulkiness, or surface area are of minor importance. The properties polarity, refractivity, and hydrophobicity, however, turned out to support feature extraction. It is concluded that membrane transition regions in proteins are encoded in sequences as a characteristic feature based on the respective side-chain properties. The method of structure evolution is described in detail for this particular application and suggestions for further development of amino acid sequence filters are made. © 1996 John Wiley & Sons, Inc.  相似文献

Abstract An inter- and intra-species correlation was found between the intracellular potassium concentration and growth temperature within the Methanobacteriales , comprising mesophiles as well as moderate ( Methanobacterium thermoautotrophicum ) and extreme thermophiles ( Methanothermus fervidus, Mt. sociabilis ). Potassium concentrations in different species were determined at optimal growth temperatures and for the same species cultured at different temperatures. The main anionic component was found to be the unusual trianionic cyclic 2,3-diphosphiglycerate. In vitro experiments with the thermolabile enzymes glyceraldehyde-3-phosphate dehydrogenase and malate dehydrogenase from Mt. fervidus indicated that the potassium salt of the cyclic diphosphoglycerate acts as potent thermostabilizer. Thus it appears that, for the methanogens, changes in the intracellular ion concentration are the basis of thermoadaptation.  相似文献

Recently we reported the characterization of simian immunodeficiency virus (SIVlhoest) from a central African l'hoest monkey (Cercopithecus lhoesti lhoesti) that revealed a distant relationship to SIV isolated from a mandrill (SIVmnd). The present report describes a novel SIV (SIVsun) isolated from a healthy, wild-caught sun-tailed monkey (Cercopithecus lhoesti solatus), another member of the l'hoest superspecies. SIVsun replicated in a variety of human T-cell lines and in peripheral blood mononuclear cells of macaques (Macaca spp.) and patas monkeys (Erythrocebus patas). A full-length infectious clone of SIVsun was derived, and genetic analysis revealed that SIVsun was most closely related to SIVlhoest, with an amino acid identity of 71% in Gag, 73% in Pol, and 67% in Env. This degree of similarity is reminiscent of that observed between SIVagm isolates from vervet, grivet, and tantalus species of African green monkeys. The close relationship between SIVsun and SIVlhoest, despite their geographically distinct habitats, is consistent with evolution from a common ancestor, providing further evidence for the ancient nature of the primate lentivirus family. In addition, this observation leads us to suggest that the SIVmnd lineage should be designated the SIVlhoest lineage.  相似文献
