Immunological analyses of the gut contents of 1769 adult and 486 nymphal instars of the lesser waterboatman, Cymatia bonsdorfi, from Lough Corrib, western Ireland, were performed using a double immunodiffusion technique. Antisera against six potential prey categories (Oligochaeta, Cladocera, Copepoda, Gammarus, Diptera and Ephemeroptera), identified on the basis of field and laboratory observations, were utilized. An expectation maximization (EM) algorithm for the identification and characterization of common sites in unaligned biopolymer sequences 总被引:18,自引:0,他引:18
Statistical methodology for the identification and characterization of protein binding sites in a set of unaligned DNA fragments is presented. Each sequence must contain at least one common site. No alignment of the sites is required. Instead, the uncertainty in the location of the sites is handled by employing the missing information principle to develop an "expectation maximization" (EM) algorithm. This approach allows for the simultaneous identification of the sites and characterization of the binding motifs. The reliability of the algorithm increases with the number of fragments, but the computations increase only linearly. The method is illustrated with an example, using known cyclic adenosine monophosphate receptor protein (CRP) binding sites. The final motif is utilized in a search for undiscovered CRP binding sites. 相似文献

The distribution of blue whales, Balaenoptera musculus , in the eastern tropical Pacific (ETP) was analyzed from 211 sightings of 355 whales recorded during research vessel sighting surveys or by biologists aboard fishing vessels. Over 90% of the sightings were made in just two areas: along Baja California, and in the vicinity of the Costa Rica Dome (a large, stationary eddy centered near 9°N, 89°W), with the rest made along the equator near the Galapagos islands, the coasts of Ecuador and northern Peru. All sightings occurred in relatively cool, upwelling-modified waters. Because these areas are the most productive parts of the ETP, and have relatively large standing stocks of euphausiids, it seems possible that blue whales select low latitude habitats which permit foraging. The waters off western Baja California were occupied seasonally, with a peak in sightings coinciding with the spring peak in upwelling and biological production. The Costa Rica Dome area was occupied year round, suggesting either a resident population, or that both northern and southern hemisphere whales visit, with temporal overlap. The modal group size was one for all areas and seasons, but the frequency of groups with two or more whales was significantly higher in sightings made near the Galapagos Islands and the coast of Ecuador and northern Peru. 相似文献

The effects of dietary supplementation of dihydroxyacetone and pyruvate (DHAP) on metabolic responses and endurance capacity during leg exercise were determined in eight untrained males (20-30 yr). During the 7 days before exercise, a high-carbohydrate diet was consumed (70% carbohydrate, 18% protein, 12% fat; 35 kcal/kg body weight). One hundred grams of either Polycose (placebo) or dihydroxyacetone and pyruvate (treatment, 3:1) were substituted for a portion of carbohydrate. Dietary conditions were randomized, and subjects consumed each diet separated by 7-14 days. After each diet, cycle ergometer exercise (70% of peak oxygen consumption) was performed to exhaustion. Biopsy of the vastus lateralis muscle was obtained before and after exercise. Blood samples were drawn through radial artery and femoral vein catheters at rest, after 30 min of exercise, and at exercise termination. Leg endurance was 66 +/- 4 and 79 +/- 2 min after placebo and DHAP, respectively (P less than 0.01). Muscle glycogen at rest and exhaustion did not differ between diets. Whole leg arteriovenous glucose difference was greater (P less than 0.05) for DHAP than for placebo at rest (0.36 +/- 0.05 vs. 0.19 +/- 0.07 mM) and after 30 min of exercise (1.06 +/- 0.14 vs. 0.65 +/- 0.10 mM) but did not differ at exhaustion. Plasma free fatty acids, glycerol, and beta-hydroxybutyrate were similar during rest and exercise for both diets. Estimated total glucose oxidation during exercise was 165 +/- 17 and 203 +/- 15 g after placebo and DHAP, respectively (P less than 0.05). It is concluded that feeding of DHAP for 7 days in conjunction with a high carbohydrate diet enhances leg exercise endurance capacity by increasing glucose extraction by muscle. 相似文献

Photosystem I reaction center of the cyanobacterium Synechocystis sp. PCC 6803 contains seven different polypeptide subunits. The subunit with a molecular mass of about 8 kDa was isolated, and the sequence of its amino-terminal residues was determined. Oligonucleotide probes corresponding to this sequence were used to isolate the gene encoding this subunit. The gene, termed as psaE, codes for a polypeptide with a mass of 8075 Da. It is present as a single copy in the genome and is transcribed as a monocistronic messenger. The amino acid sequence of the 8-kDa subunit deduced from the gene sequence shows high homology with the deduced amino acid sequence of subunit IV of photosystem I from spinach. The DNA fragment sequenced in these studies also contains two other unidentified major open reading frames. A stable deletion mutation for the psaE gene was generated by transforming Synechocystis sp. PCC 6803 with a cloned DNA in which the psaE gene for 8-kDa subunit was replaced by a gene conferring resistance to kanamycin. The cyanobacterium Synechocystis sp. PCC 6803 carries out oxygenic photosynthesis analogous to higher plants. Its photosystem I contains seven different polypeptide subunits. The cartridge mutagenesis technique was used to inactivate the psaD gene which encodes subunit II of photosystem I. A mutant strain lacking subunit II was generated by transforming wild type cells with cloned DNA in which psaD gene was interrupted by a gene conferring kanamycin resistance. The photoautotrophic growth of mutant strain is much slower than that of wild type cells. The membranes prepared from mutant cells lack subunit II of photosystem I. Studies on the purified photosystem I reaction center revealed that the complex lacking subunit II is assembled and is functional in P700 photooxidation but at much reduced rate. Therefore, subunit II of photosystem I is required for efficient function of photosystem I. 相似文献

All tetrapods have the same four basic abdominal hypaxial muscle layers that wrap around the abdomen between the pelvis, ribcage, and spine. However, the marsupials and our immediate mammalian ancestors have epipubic bones extending anteriorly into the ventral hypaxial layers with two additional muscles connecting them to the ventral midline and femur. Studies of two marsupials have shown that all of the abdominal hypaxials play a part bilaterally in resting ventilation and during locomotion there is an asymmetrical pattern of activity as the hypaxial muscles form a cross‐couplet linkage that uses the epipubic bone as a lever to provide long‐axis support of the body between diagonal limb couplets during each step. The cross‐couplet epipubic lever system defines the earliest mammals and is lost in placental mammals. To expand our understanding of the evolution of mammalian abdominal muscle function and loco‐ventilatory integration we tested the generality of the cross‐couplet system in marsupials and conducted the first formal studies of hypaxial abdominal motor patterns in generalized placental mammals focusing on a representative rodent and insectivore. These new data reveal 1) that continuous abdominal muscle tonus during resting ventilation and a 1:1 breath to step cycle during locomotion appear to be the basal condition for mammals, 2) that the loss of epipubic bones in eutherians is associated with a shift from the cross‐couplet dominated motor pattern of marsupials to a shoulder‐to‐pelvis system with unilateral activation of abdominal muscles during locomotion and 3) that hypaxial function in generalized eutherians is more similar to marsupials than cursorial mammals. MOTIVATION: Although population-based association mapping may be subject to the bias caused by population stratification, alternative methods that are robust to population stratification such as family-based linkage analysis have lower mapping resolution. Recently, various statistical methods robust to population stratification were proposed for association studies, using unrelated individuals to identify associations between candidate genes and traits of interest. The association between a candidate gene and a quantitative trait is often evaluated via a regression model with inferred population structure variables as covariates, where the residual distribution is customarily assumed to be from a symmetric and unimodal parametric family, such as a Gaussian, although this may be inappropriate for the analysis of many real-life datasets. RESULTS: In this article, we proposed a new structured association (SA) test. Our method corrects for continuous population stratification by first deriving population structure and kinship matrices through a set of random genetic markers and then modeling the relationship between trait values, genotypic scores at a candidate marker and genetic background variables through a semiparametric model, where the error distribution is modeled as a mixture of Polya trees centered around a normal family of distributions. We compared our model to the existing SA tests in terms of model fit, type I error rate, power, precision and accuracy by application to a real dataset as well as simulated datasets. 相似文献
