To determine if molybdenum (Mo) is a chemical stressor, fingerling and juvenile rainbow trout (Oncorhynchus mykiss) were exposed to waterborne sodium molybdate (0, 2, 20, or 1,000 mg l-1 of Mo) and components of the physiological (plasma cortisol, blood glucose, and hematocrit) and cellular (heat shock protein [hsp] 72, hsp73, and hsp90 in the liver, gills, heart, and erythrocytes and metallothionein [MT] in the liver and gills) stress responses were measured prior to initiation of exposure and at 8, 24, and 96 h. During the acute exposure, plasma cortisol, blood glucose, and hematocrit levels remained unchanged in all treatments. Heat shock protein 72 was not induced as a result of exposure and there were no detectable changes in total hsp70 (72 and 73), hsp90, and MT levels in any of the tissues relative to controls. Both fingerling and juvenile fish responded with similar lack of apparent sensitivity to Mo exposure. These experiments demonstrate that exposure to waterborne Mo of up to 1,000 mg l-1 did not activate a physiological or cellular stress response in fish. Information from this study suggests that Mo water quality guidelines for the protection of aquatic life are highly protective of freshwater fish, namely rainbow trout. 相似文献

The causes of the exceptionally early blooms of Ceratium spp.in the Southern Bight of the North Sea during the early 1970sare re-assessed. This localised event is found to coincide witha change in the meridional circulation over the North Sea inthe late winter and early spring, with abnormally strong northerliesduring the 1960s giving way to strengthened southerlies duringthe early 1970s. Such a change in wind direction can be expected(both from observation and simulation) to bring about a lesswidespread distribution of turbid coastal water in the SouthernBight and the marked reduction in windfetch may also have servedto decrease watercolumn turbidity through its effect on theincidence of long-period swell. These processes may be reinforcedby associated changes in the quantity of turbid runoff dischargedby the Rhine. The advanced blooms of Ceratium spp. in the early1970s are attributed both to these effects and to an increasedfrequency of light winds (decreased wind mixing) in the earlymonths of the year. 相似文献

Proteins with RER-specific signal sequences are cotranslationally translocated across the rough endoplasmic reticulum through a proteinaceous channel composed of oligomers of the Sec61 complex. The Sec61 complex also binds ribosomes with high affinity. The dual function of the Sec61 complex necessitates a mechanism to prevent signal sequence-independent binding of ribosomes to the translocation channel. We have examined the hypothesis that the signal recognition particle (SRP) and the nascent polypeptide-associated complex (NAC), respectively, act as positive and negative regulatory factors to mediate the signal sequence-specific attachment of the ribosome-nascent chain complex (RNC) to the translocation channel. Here, SRP-independent translocation of a nascent secretory polypeptide was shown to occur in the presence of endogenous wheat germ or rabbit reticulocyte NAC. Furthermore, SRP markedly enhanced RNC binding to the translocation channel irrespective of the presence of NAC. Binding of RNCs, but not SRP-RNCs, to the Sec61 complex is competitively inhibited by 80S ribosomes. Thus, the SRP-dependent targeting pathway provides a mechanism for delivery of RNCs to the translocation channel that is not inhibited by the nonselective interaction between the ribosome and the Sec61 complex. 相似文献

1. Climate effects on plant defences, and subsequently insect herbivores, have received considerable attention. However, climate also directly affects the physiological condition of insects that may influence their ability to combat plant defences. 2. This study tested the effects of body fat and water content on the ability of mountain pine beetles, Dendroctonus ponderosae Hopkins, to tolerate monoterpenes, the primary defences of their host pine trees. To manipulate their physiological condition, we exposed beetles to one of three environmental treatments: cold (4 °C, 30% RH), humid (21 °C, 70% RH) or dry (21 °C, 30% RH). Beetles had the highest fat and water content in the cold treatment, followed by lower fat but the same water content in the humid treatment, and the lowest of both in the dry treatment. Following environmental treatments, beetles were exposed to 0, 312.5 or 625 ppm vapours of monoterpenes (−)-α-pinene or (R)-(+)-limonene. 3. Survival during both environmental and monoterpene treatments was highest for the cold treatment, less for the humid treatment, and lowest for the dry treatment. Fat and water content positively predicted survival. In response to increasing monoterpene concentration, survival, body condition and water content, but not fat, declined. 4. These results indicate that water content of insect herbivores was important for the realised toxicity of plant defences, presumably due to the process of excreting detoxified compounds. Energetic costs of detoxification were less evident. Dehydration of insects has not been widely considered in insect–plant interactions, but effects of climate change on humidity may become increasingly important for these interactions. 相似文献

Monoclonal antibody 10.2-16 is directed toward the mouse class II major histocompatibility complex gene product 1-Ak expressed on the cell line LK35.2. Instead of activating cells by fluorophor we used (acrylamide-coated) heavy and magnetic microspheres of 0.6 micron in radius. These microspheres are chemically coupled (carbodiimide method) with the antibody toward the surface antigen. The cells are observed through a microscope with horizontal alignment, as they sediment in a (temperature controlled) tube with square cross-section. Stokes Law allows the determination of the density of cells (first alone) using viscosity and density of Dulbecco's modified Eagle's Medium together with the observed mean sedimentation velocity (66 microns/min) and a mean diameter of 10 microns. We found a density of 1.0558 +/- 0.0028 g/cm3 at 10 degrees C. Independently, thinly coated, heavy (and magnetizable) microspheres with the cited antibody are attached to cells and observed likewise. The increased sedimentation velocity permits us to show that the cells were fully covered with microspheres (290 per cell). A magnetic field gradient opposing gravity moved these cells against gravity with two different mean velocities, 340 microns/min and 850 microns/min. The higher velocity resulted in 290 particles per cell, the lower one in 130 particles per cell. The limits for the expansion of this method to smaller particle sizes (down to 10 nm) are evaluated. 相似文献
