近年来,植物遗传转化研究有了长足的发展。已经达到能够通过简单的遗传控制手段研究具有新表现型的植物,甚至达到进入商业化的程度。这些手段包括植物生物学的主要研究技术以及植物组织培养和树种改良的一些实用方法。尽管采用农瘤杆菌和鸟枪法等技术的植物遗传转化系统已经得到了广泛的应用,但是在如何开发具有能够得到控制表达的转基因高产植物方面,在如何使所得到的转基因植物远离遗传危害等方面,目前的转化系统遇到了极大的技术挑战。已经提出了各种各样的方法用于将新基因稳定地导入120多种不同植物的核基因组。本文将讨论这些遗传转化系统所需的生物学要求和实际应用方面的需求、基因转化和转基因表达的研究策略、遗传转化植物的鉴定以及转基因植物与大众的认可。本文将分为七个部分加以讨论:一、导言;二 、基因转化到细胞里的方法;三、植物遗传转化策略;四、植物遗传转化的鉴定;五、植物遗传转化的实际应用;六、转基因植物与环境;七、未来植物遗传转化的需求与发展方向。  相似文献

Using a nested plot design in oak forests in Minnesota, USA we measured the percent cover of young individuals of an exotic invading shrub, Rhamnus cathartica (common buckthorn), to assess the relationships at two scales between invasibility, propagule and light availability, and richness and cover of native species. Comparing patches (1 m2) within 17 Quercus -dominated stands (each 1 ha, within a 100 km by 150 km area), cover of young R. cathartica was negatively related to both species richness and cover of native species. In 1 m2 patches, native cover was positively associated with native richness and thus cover-related competition was a likely mechanism by which richness influenced R. cathartica . At the landscape scale (comparing the aggregate stand-scale metrics among the 17 stands), native cover and richness were still positively related, but had opposite relationships with R. cathartica cover. R. cathartica cover was positively related to species richness and negatively related to native species cover. The observed switch at different scales from a positive to a negative relationship between R. cathartica cover and native richness supported the hypothesized scale dependence of these relations. Propagule pressure, which we estimated by measuring the size of nearby mature R. cathartica shrubs, had a large positive effect on R. cathartica seedling cover at the landscape scale. These results suggest that landscape patterns of invasion may be best understood in light of the combination of many factors including native diversity, native cover, and propagule pressure.  相似文献

The sebum excretion times (SET) were studied in two breeds of cattle, Bos taurus (SH) and the more heat tolerant B. indicus (GB). Both breeds were maintained in thermoneutral (24 degrees C) and hot (32 degrees C) environments. SET was defined as the time between synthesis and release of sebum on to the skin surface. The SET was 7 days in SH and 6 days in GB animals at 24 degrees C but was a day earlier at 32 degrees C for each breed. The changing SET with environmental temperature was associated with an increased rate of synthesis of sebum with increased body temperature.  相似文献

Inherently curved DNA segments, associated with short runs of adenines, have been identified in many gene regulatory regions, yet their physiological significance remains unknown. The observations reported in this study indicate that intrinsically bent nucleic acid fragments are characterized by substantially attenuated affinities toward DNA-binding proteins involved in structural functions, such as H1 histone and protamine, as well as toward various DNA-modifying enzymes including ligases and exo- and endonucleases. Two mechanisms might be responsible for the altered binding properties. According to the first mechanism, the attenuated binding affinities and the bending represent two independent consequences of the unique structural parameters exhibited by A-tracts. Indeed, analysis of the degradation products obtained upon exposure of the curved sequences to various chemical nucleases points toward the narrowing of the DNA minor groove, a conformational modulation known to characterize A-tracts and to run along the axially-bent motifs, as a potential determinant of the observed binding attenuation. Alternatively, the conformational constraints which result from the stable bending might act to modulate the strength of DNA-protein interactions. Although the factor directly responsible for the altered binding affinities revealed by the bent sequences cannot as yet be conclusively resolved, it is proposed that a reiteration of this specific factor, being either an A-tract or a bend, in phase with the DNA helical repeat acts to amplify the modulation of the binding.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Microbial communities have important functions during spring phytoplankton blooms, regulating bloom dynamics and processing organic matter. Despite extensive research into such processes, an in-depth assessment of the fungal component is missing, especially for the smaller size fractions. We investigated the dynamics of unicellular mycoplankton during a spring phytoplankton bloom in the North Sea by 18S rRNA gene tag sequencing and a modified CARD-FISH protocol. Visualization and enumeration of dominant taxa revealed unique cell count patterns that varied considerably over short time scales. The Rozellomycota sensu lato (s.l.) reached a maximum of 105 cells L−1, being comparable to freshwater counts. The abundance of Dikarya surpassed previous values by two orders of magnitude (105 cells L−1) and the corresponding biomass (maximum of 8.9 mg C m−3) was comparable to one reported for filamentous fungi with assigned ecological importance. Our results show that unicellular fungi are an abundant and, based on high cellular ribosome content and fast dynamics, active part of coastal microbial communities. The known ecology of the visualized taxa and the observed dynamics suggest the existence of different ecological niches that link primary and secondary food chains, highlighting the importance of unicellular fungi in food web structures and carbon transfer.  相似文献

With an estimated $10.4 billion in medical costs and 31.4 million outpatient visits each year, influenza poses a serious burden of disease in the United States. To provide insights and advance warning into the spread of influenza, the U.S. Centers for Disease Control and Prevention (CDC) runs a challenge for forecasting weighted influenza-like illness (wILI) at the national and regional level. Many models produce independent forecasts for each geographical unit, ignoring the constraint that the national wILI is a weighted sum of regional wILI, where the weights correspond to the population size of the region. We propose a novel algorithm that transforms a set of independent forecast distributions to obey this constraint, which we refer to as probabilistically coherent. Enforcing probabilistic coherence led to an increase in forecast skill for 79% of the models we tested over multiple flu seasons, highlighting the importance of respecting the forecasting system’s geographical hierarchy.  相似文献

Patterns of laboratory-determined aggressive behaviour and exploratory activity were examined in wild-caught male beach voles (Microtus breweri), an island-endemic species. This species is ecologically interesting because it has been found not to exhibit the super-annual population cycles typical of other rodents in the subfamily Microtinae. We found that body weight and reproductive condition were good predictors of aggressive behaviour, with heavier and reproductively active voles showing higher incidences of several stereotyped aggressive acts. Aggression also increased as the percentage of males and females who were reproductively active in the control population increased. Aggression was negatively correlated with population density, and more or less independent of dispersal rate. Dispersers and residents exhibited similar aggressive behaviour patterns. These observations are consistent with models which have proposed that population cycles in other vole species are controlled through the social interactions of individuals.  相似文献

New and safe methods of investigating the kidney by x-ray visualization of its circulation are now available. This can be done either by injecting large amounts of contrast material through a vein and then taking multiple x-ray exposures of the kidneys, or by injecting the contrast material backward into the aorta to the level of the renal arteries.Another method of x-ray examination now available is taking body section films of the kidneys when the contrast medium is in the kidney substance. The synthesis of DNA was investigated inLactobacillus acidophilus R-26 during starvation for glutamic acid. The synthesis of RNA and of proteins was also examined. The content of DNA in the cell increased by 150–250%. The results show that new DNA-replication cycles can be initiated without simultaneous synthesis of proteins and of RNA.  相似文献
