Inoculum-dependent division lag of Bacillus cultures and its relation to an endogenous factor(s) ("schizokinen"). J. Bacteriol. 91:1070-1079. 1966.-When cells of Bacillus megaterium, grown on Brain Heart Infusion Agar, were inoculated into a chemically defined medium, they exhibited a division lag which was an inverse function of inoculum size. The addition of filtrates of cultures from the same medium eliminated the inoculum-dependent component of lag, but not an inoculum-independent residual lag of constant duration. Culture filtrate of B. subtilis var. niger not only eliminated its inoculum-dependent lag but also was required to sustain exponential division. Dose-response and "growth time" bioassays, developed to measure lag-reducing activity of filtrates, demonstrated accumulation of active filtrate factor to a "critical" concentration prior to division initiation. Addition of this concentration to cultures eliminated the inoculum-dependent lag. Accumulation of the factor ceased temporarily at onset of division, but excretion was resumed later during exponential growth. Accumulation of a lag-reducing, cell-associated factor followed a similar course. Chromatographic and bioautographic analyses of culture filtrates of B. megaterium indicated that a single substance was primarily responsible for their activity. Results of dose-response tests for reciprocal activities of filtrates of different Bacillus species and strains suggested production of different factors by some, and of different quantities of similar factors by others. It is proposed that such endogenous factors which are synthesized and accumulate to a population-dependent concentration as a requisite to initiation and maintenance of division be designated as "schizokinens."  相似文献

Summary The present studies examined some of the properties of Cl channels in renal outer medullary membrane vesicles incorporated into planar lipid bilayers. The predominant channel was anion selective having aP Cl/P K ratio of 10 and a unit conductance of 93 pS in symmetric 320mm KCl. In asymmetric KCl solutions, theI-V relations conformed to the Goldman-Hodgkin-Katz equation. Channel activity was voltage-dependent with a gating charge of unity. This voltage dependence of channel activity may account, at least in part, for the striking voltage dependence of the basolateral membrane Cl conductance of isolated medullary thick ascending limb segments. The Cl channels incorporated into the planar bilayers were asymmetrical: thetrans surface was sensitive to changes in ionized Ca2+ concentrations and insensitive to reducing KCl concentrations to 10mm, while thecis side was insensitive to changes in ionized Ca2+ concentrations, but was inactivated by reducing KCl concentrations to 50mm.  相似文献

The O antigen of Salmonella group B strains contains the sugar abequose, whereas those from group A and D strains contain paratose or tyvelose in its place. This is the essential difference between these Salmonella groups. Only the final step in the biosynthesis of abequose differs from that of paratose, and the abequose confers on group B strains their specific O4 antigen. The gene, rfbJ, encoding the enzyme abequose synthase for this last specific step has been cloned, identified, and sequenced and has been shown to function in group A and D strains to make them O4+. This one gene thus differentiates group B from group A or group D salmonellae. The enzyme abequose synthase appears to be related to galactose epimerase, and the significance of this is discussed. The rfbJ gene and adjacent DNA is of much lower G+C content than is usual for salmonellae, indicating that the region did not originate in a salmonella but was transferred from outside.  相似文献

The protamines are small, arginine-rich nuclear proteins that replace histones and transition proteins late in the haploid phase of spermatogenesis in mammals. The two mouse genes encoding protamines--Prm-1 and Prm-2--have been molecularly cloned and mapped to mouse chromosome 16 (MMU 16). A cDNA clone of mouse Prm-1 that hybridized to the corresponding human gene was used to analyze a panel of somatic cell hybrids made between human lymphoblasts and the E36 hamster cell line. The human gene, which we have designated PRM 1, was syntenic with human chromosome 16 (HSA 16) and discordant with all other human chromosomes. Linkage analysis in the mouse was accomplished using the backcross (Czech II x BALB/c Pt) x Czech II to map Prm-1 and Prm-2 to a position near the 5' terminus of MMU 16. No recombination between Prm-1 and Prm-2 was observed among 89 progeny of the Czech II x BALB/c cross or among 94 progeny of the backcross (CBA/J x BALB/cJ) x BALB/cJ, demonstrating that the two loci are separated by less than 1.6 cM on MMU 16. This tight linkage may be of functional significance, as Prm-1 and Prm-2 are among a limited number of genes known to be expressed postmeiotically in male haploid germ cells.  相似文献

Interactive effects of seven years of compaction due to wheel traffic and tillage on root density, formation of arbuscular mycorrhizae, above-ground biomass, nutrient uptake and yield of corn (Zea mays L.) were measured on a coastal plain soil in eastern Alabama, USA. Tillage and soil compaction treatments initiated in 1987 were: 1) soil compaction from tractor traffic with conventional tillage (C,CT), 2) no soil compaction from tractor traffic with conventional tillage (NC,CT), 3) soil compaction from tractor traffic with no-tillage (C,NT), and, 4) no soil compaction from tractor traffic with no-tillage (NC,NT). The study was arranged as a split plot design with compaction from wheel traffic as main plots and tillage as subplots. The experiment had four replications. In May (49 days after planting) and June, (79 days after planting), root biomass and root biomass infected with arbuscular mycorrhizae was higher in treatments that received the NC,NT treatment than the other three treatments. In June and July (109 days after planting), corn plants that received C,CT treatment had less above-ground biomass, root biomass and root biomass infected with mycorrhizae than the other three treatments. Within compacted treatments, plants that received no-tillage had greater root biomass and root biomass infected with mycorrhizae in May and June than plants that received conventional tillage. Corn plants in no-tillage treatments had higher root biomass and root biomass infected with mycorrhizae than those in conventional tillage. After 7 years of treatment on a sandy Southeastern soil, the interactive effects of tillage and compaction from wheel traffic reduced root biomass and root biomass infected with mycorrhizae but did not affect plant nutrient concentration and yield. ei]J H Graham  相似文献

Escherichia coli strains ECOR2, ECOR3 and K-12 are very closely related in genotype as indicated by multilocus enzyme electrophoresis. We show that they have very different rfb regions indicating that recombination has occurred in this region, and we suggest that it may be associated with niche adaptation.  相似文献
