In this report we present a series of experiments which have led us to support the notion of the presence of an ACTH receptor in the CNS. A short intense heat-stress (hot-plate) applied to the paws of rats will temporarily reduce activity. During the course of experimentation we were able to eliminate a number of logical mediators. Neither adrenalectomy, adrenal-medullectomy, naloxone administration, nor alpha-MSH-(1-12) were able to affect the observed akinesia. Hypophysectomy, however, was able to abolish or mask the behavior and the reduction in activity could be reinstated by the administration of ACTH-(4-10) to hypophysectomised rats. These data support the notion that a short intense stressor can release ACTH and that this ACTH can be responsible for mediating the short term reduction in activity. In addition, the fact that ACTH-(4-10) has only minimal steroidogenic properties and was able to reinstate the behavior led us to speculate that these effects were of central origin. Furthermore, since naloxone was not capable of altering the behavior, the suggestion is that ACTH in this paradigm acted at a receptor site apart from the naloxone sensitive receptor. This site may in fact be an ACTH specific receptor.  相似文献

A single dose of des-glycinamide-[Arg8]vasopressin (DGAVP, 2 mg intranasal) or oxytocin (OXT, 20 IU intranasal) was given to female and male volunteers, respectively, in a placebo-controlled double-blind trial. Memory, vigilance, attention, and mood were tested starting 10 minutes after treatment. The DGAVP dose improved delayed recognition of abstract words when measured 1 week after treatment and reduced the intercept of a memory comparison task (Sternberg paradigm). A trend was present for DGAVP and OXT to affect learning, i.e., storage processes of verbal memory in an opposite way; DGAVP improved, while OXT attenuated initial storage and the rate of storage. No treatment effects on visual memory and vigilance were found. Of the mood measures, vigor was reduced immediately after treatment with OXT.  相似文献

Erythroid Krüppel-like factor (EKLF) is atrans-acting factor that binds specifically to the β-globin promoter CACCC box. EKLF is essential for adult erythroid development in mice. We have cloned and sequenced a cDNA encoding the human homologue of this gene, which shares 69% identity with the mEKLF protein. The expression of hEKLF is restricted to fetal liver and adult bone marrow. The genomic locus of hEKLF was mapped to chromosomal band 19p13.12–p13.13, using fluorescencein situhybridization.  相似文献

During October-December 1987, chigger mites infested on the striped field mice (Apodemus agrarius coreae) collected at Pochun-gun, Gyeonggi Province and Chinhae city, Kyongnam Province were identified. 1. Of 171 back-striped mice collected, chigger mites were found from 58 mice, showing 33.9% of infestation rate. 2. Total 865 chigger mites collected were classified into seven species; Leptotrombidium pallidum (4.3%), L. palpale(23.0%), L. orientalis(20.2), L. zetum(19.0%). Neotrombicula tamiyai(32.9), N. japonica(0.2%) and Euschongastia koreaensis(0.3%). Though the most dominant species in number was N. tamiyai(32.9% of the total), this species was collected at very limited locality and for a short period. Therefore, most common species in Korea seems to be L. palpale which was found at all localities throughout the whole survey period. L. pallidum which is known as the vector species of tsutsugamushi disease in Korea was collected in a very low number(4.3% of the total chiggers collected). 3. The number of chigger mites infested on a host animal showed great variations from one chigger up to 207 chiggers. The present study has shown that there may be other Leptotrombidium species mite(s) for the vector and host of tsutsugamushi disease in Korea.  相似文献

Opioid peptides have been shown to enhance peripheral blood lymphocyte natural killer (NK) cell function. In this study, we report that certain non-opioid fragments of beta-endorphin (2-16, 2-17, and 6-17) enhance NK activity with peak dose responses seen at 10(-12) to 10(-15) concentrations. This effect can be inhibited by naloxone. We conclude that non-opioid fragments of beta-endorphin are more potent than either beta- or gamma-endorphin in enhancing human lymphocyte NK activity.  相似文献

This study demonstrated that transposable elements in Pseudomonas cepacia could be inserted upstream of a poorly expressed gene and increase its expression more than 30-fold. Five elements, TnPc1, IS402, IS403, IS404, and IS405, were isolated by their ability to increase expression of the beta-lactamase gene of the broad-host-range plasmid pRP1. Increased expression resulted only from insertion of these elements, suggesting that insertional activation is an important means of elevating gene expression in this organism. Four of the elements inserted between a PstI site within the beta-lactamase gene and a BamHI site located 375 base pairs upstream of its promoter. The element IS403 inserted distal to the BamHI site within the coding region for the gene tnpR, suggesting that insertional activation can act over greater than expected distances. In addition, the element IS402 activated the beta-lactamase genes carried on plasmids pRP1 and pMR5 (temperature-sensitive pRP1) equally well in opposite orientations, demonstrating that insertional activation by this element occurs independent of its orientation.  相似文献

A mathematical model previously developed to explain beta-cell membrane potential oscillations has been modified to accommodate the external variation of K+, Na+ and Ca2+ concentrations. Our model, which is applicable to excitable cells, incorporates the barrier kinetics. Hodgkin-Huxley-type gating mechanism, and an electrogenic Na+-K+ pump. Numerical solutions of our model are in agreement with many of the experimental results reported in the literature on excitable cells.  相似文献

In view of the close structural similarity of the pro-opiocortin fragment γ-MSH and of ACTH/MSH type peptides, the behavioral profile of γ-MSH was explored. Attention was first focussed on behavioral procedures in which ACTH/MSH related neuropeptides have been found effective. Using different procedures to test avoidance behavior, it was found that γ-MSH and ACTH-like neuropeptides had opposite effects. In this respect the activity of γ-MSH resembles that of opiate antagonists rather than that of β-endorphin. Accordingly, ACTH1–24-induced excessive grooming which is blocked by opiate antagonists, is attenuated by γ-MSH. In addition, γ-MSH injected into the periaqueductal gray matter of the brainstem of opiate naive rats elicited symptoms reminiscent of those seen after opiate withdrawal. γ-MSH attenuated more or less several effects of intracerebroventricularly administered β-endorphin (e.g. antinociception, hypothermia, α-MSH release) and decreased acquisition of heroin self-administration. Although γ-MSH at rather high doses displaced naloxone from its specific binding sites in brain homogenates, it did not interfere with β-endorphin-induced effects on in vitro muscle preparations (guinea pig ileum, rat rectum). Interestingly, γ-MSH induced relaxation of the rat rectum in vitro. It is postulated that γ-MSH may attenuate β-endorphin-induced effects by acting via γ-MSH receptor sites (functional antagonism), although a pharmacological antagonism cannot be excluded as yet.  相似文献

Epidemiological studies on host rodents of tsutsugamushi disease were carried out during the period of July-September 1990 at nine localities of central Korea. Among total 111 wild rodents trapped by the modified Sherman live traps, 103 were Apodemus agrarius (92.8%), seven were Crocidura lasiura (6.3%) and one was Microtus fortis (0.9%), showing 24.0% of trapping rate in winter, 11.7% in spring, 11.2% in summer and 12.0% in autumn. Out of 103 A. agrarius 84 were parasitized by chiggers, showing 81.6% of the infestation rate and 43.0 of the chigger index. The antibody positive rate of A. agrarius sera to Rickettsia tsutsugamushi was significantly variable by locality, being in the range of 0-78.6%. The seasonal change of the antibody positive rate at Dorai 5-ri, Goyang-gun was 75.8% in average during November-March, decreased to 30.3% in April and further decreased to 13.3% in average during May-August. Among 33 antibody positives, 31 were Karp strain and two were Gilliam. Seven Crocidura lasiura sera showed all negative. R. tsutsugamushi organisms were isolated from three A. agrarius out of 94 mice tested, showing 3.2% of the infection rate.  相似文献
