Starch supported production of maximum α-amylases (dextrinizing and saccharifying) byFusarium oxysporum andF. scirpi. Addition of gibberellic acid resulted in an increased production of α-amylase. Presence of glucose depressed the enzyme production. pH 4.5 and 4.0 was found to be optimum for the dextrinizing enzyme secreted by both species. The temperature of 25 and 40 °C was optimum for the dextrinizing enzyme secreted byF. oxysporum andF. scirpi, respectively. Saccharifying enzymes of both species showed their optimum at pH 6.9. The optimum temperature for the activity of the saccharifying enzyme was 30 and 40 °C, respectively.  相似文献

We present methods for the rapid, simple purification of Enzyme I, HPr, and Protein IIIGlc of the Escherichia coli phosphoenolpyruvate:sugar phosphotransferase system (PTS) using plasmids overproducing gene products. The gene for HPr (ptsH) was cloned into the expression vector pKC30. A simple procedure was devised for the purification to homogeneity of this protein from extracts of heat-induced cells containing pKC30/ptsH recombinant clone. The genes for Enzyme I (ptsI) and Protein IIIGlc (crr) were cloned separately into the expression vector pRE1. Rapid purification procedures were developed for the isolation of homogeneous preparations of these two proteins from extracts of heat-induced cells containing pRE1/ptsI and pRE1/crr recombinants. From about 6 g of cells, these procedures yielded 100, 86, and 50 mg of Enzyme I, HPr, and Protein IIIGlc, respectively. The activity of the proteins purified by these methods was comparable to that of the proteins isolated by previously published less efficient procedures.  相似文献

Bilateral cryptorchidism was induced surgically in albino rats and pattern of protein profiles was studied in reproductive organs. Cryptorchidism activated tissue proteolysis leading to overall degradation in soluble and structural protein fractions and in amino acids leading to prevalence of negative nitrogen balance in the reproductive organs. The testicular hypoalbuminic and hypoglobulinic conditions seem to be responsible for oligo-astheno-spermia associated with cryptorchidism.  相似文献

We have studied the activities of 2′,3′-cyclic nucleotide 3′-phosphohydrolase, 1,2-diacylglycerol: CDPethanolamine phosphoethanolamine transferase (EC 2.7.8.1), and 1,2-diacylglycerol: CDPcholine phosphocholine transferase (EC 2.7.8.2) in developing rat brain gray matter and white matter. The specific activity of cyclic nucleotide phosphohydrolase was 5–8 fold higher in white matter than in gray matter at all ages. No significant changes were observed during development. The specific activity of phosphocholine transferase was 2 to 3 fold higher than phosphoethanolamine transferase at all ages both in gray and white matter. Both phosphocholine transferase and phosphoethanolamine transferase increased more than 2 fold in specific activity between 14 and 90 days of age. The total activity of phosphocholine transferase also showed an increase during development. The apparentK m values for nucleotides and dicaprin were similar in gray matter and white matter. Except for lowK m values for nucleotides at 14 days of age, no significant changes were observed during development. Changes in rates of glycerophospholipid synthesis may be partly due to the specific activities of these enzymes but are also determined by the quantities of substrates and inhibitors and by affinities for the substrates. Changes in the cell surface of vaginal epithelial cells were studied by scanning microscopy and fluorescence spectroscopy. Microvilli which are prominent features of the vaginal epithelial cells in proestrus and diestrus are replaced by sheet-like structures in the estrus phase. Surface morphology of vaginal epithelial cells of estradiol primed rat resembles the vaginal cells from estrus phase rats whereas vaginal cells from control rats resembles the diestrus phase. Measurement of the fluidity of the membranes indicated that the vaginal epithelial cell membrane of estrus rats is more fluid compared to proestrus and diestrus. Similarly, estradiol primed immature rat vaginal epithelial cell membrane was observed to be more fluid than the corresponding control.  相似文献

We report the use of a sensitive and specific enyzme-linked immunosorbent assay (ELISA) and polymerase chain reaction (PCR) to study the expression of protein D, a major androgen-regulated sperm-binding glycoprotein at the protein and mRNA level in different anatomical regions of the rat epididymis. The concentration of protein D in the caput, corpus and cauda region of the epididymis was 10.2 +/- 0.67, 7.3 +/- 0.61 and 22.8 +/- 1.34 ng/micrograms total protein, respectively. The total RNA extracted from the caput, corpus and cauda regions of the rat epididymis was amplified by PCR with oligonucleotide primers specific for the 5' and 3' portion of protein D cDNA. Compared to the caput and cauda region, a significant reduction (greater than 82 +/- 3%) in the expression of protein D mRNA levels was observed for corpus epididymal RNA. This data demonstrates regional differences in the concentration of protein D and suggests that protein D expression may be regulated at the level of mRNA within the corpus epididymidis.  相似文献

The heterogeneous vegetation mosaic of the South Turkana region of north Kenya is associated with diversity in the region's physical environment. The abundance and distribution of the dominant species are related to gradients in those abiotic factors that influence water availability, including precipitation, soil texture, and topographic relief. Research focused on three Acacia species that are a major component of the Turkana vegetation; A. tortilis, A. senegal, and A. reficiens. These species each exhibit a different response to variations in abiotic factors. Consequently, species abundance varies independently across the landscape, creating a continuum of intergrading populations. Community types can be identified within the mosaic of intergrading populations. Although community borders are not discrete due to continual change in species abundance, types are identifiable and are repeated in areas with similar environmental conditions. The landscape patterns are representative of Whittaker's (1953) climaxas-pattern, with communities created by individual patterns of populations responding to environmental gradients, creating a continuum of community change across the landscape.  相似文献

Transforming growth factor β1 (TGFβ1) inhibits epithelial cell proliferation late in the G1 phase of the cell cycle. We examined the effect of TGFβ1 on known late G1 cell cycle regulators in an attempt to determine the molecular mechanism of growth inhibition by this physiological inhibitor. The results demonstrate the TGFβ1 inhibits the late G1 and S phase specific histone H1 kinase activity of p33cdk2. This inhibitiion is not dur to TGFβ1's effect on p33cdk2 synthesis, but rather due to its negative effect on the late G1 phosphorylation of p33cdk2. It is also shown that TGFβ1 inhibits both late G1 cyclin A and cyclin E associated histon H1 kinase activities. The inhibitor has no effects on the synthesis of cyclin E but to inhibit the synthesis of cyclin A protein in a cell cycle dependent manner. If TGFβ1 is added to cells which have progressed futher than 8 hours into G1, then it is without inhibitory effect on cyclin A synthesis. These effect on TGFβ1 on late G1 cell cycle regulators correlate well with its inhibitory effects on cellular growth and suggest that these G1 cyclin dependent kinases might serve as targets for TGFβ1-mediated growth arrest.  相似文献
