The sensitivity of diploid human fibroblasts to the cytotoxic effects of diphtheria toxin (DT) depended on the cell growth status. Exponentially growing cells treated with 10?3-1 lethal flocculating units (LF) of DT/ml for 4 days survived with a frequency of 4 × 10?4. However, the DT-resistant phenotype of colonies isolated under these conditions was not stable. When the growth of the cells had been arrested by confluence or deprivation of serum growth factors prior to treatment with DT (4 days, 10?3-0.6 LF/ml), the survival decreased to 2 × 10?6 and the resistance of isolated colonies was stable. An in situ assay for induced DT-resistant mutants was developed in order to avoid problems associated with the possible reduced viability of the mutants relative to that of wild-type cells. A reproducible and linear dose response was obtained for the induction of DT-resistant mutants by ethylnitrosourea. The mutants were induced with high frequency by this compound (e.g., 10?3 mutants/viable cell at a 37% survival dose); complete expression of the mutant phenotype occurred after 6 generations of growth under nonselective conditions. Isolated mutant colonies showed stable resistance to DT and were cross-resistant to Pseudomonas aeruginosa exotoxin A.  相似文献

What was the environmental complex in which the Plio-Pleistocene hominids were evolving? What situations selected f o r increasing variation in hominid morphology? A n ap-preciation of ecological dynamics is important to develop answers to those questions. The circumstances that accompany periodically more severe semiarid successions appear to have promoted a shqt in early australopithecine morphology toward hyper-robust forms. Successional dynamics (including ameliorated conditions) may have resulted in repeated sympat y of potentially competing lines. Through the process of character divergence, some earliergracile populations appear to have emerged more Homo-like by the end of the Pliocene. Palaeodemographic analysis combined with detailed ecological modeling offers new possibilities f o r explorato y retrodictions concerned with common and divergent hominid adaptations. A 3-year study was conducted on an existing, one-cell, facultative sewage lagoon having a total surface area of 3.6 ha and receiving a BOD5 loading rate of 44 kglhald (40 Iblald). The comparative experimental periods ran from July through November for 3 consecutive years. During the first period, water hyacinths completely covered the lagoon. The water hyacinth coverage was reduced to 33% of the total surface area the second year. The lagoon, free of all vascular aquatic plants the third year, was monitored for comparative purposes. The most significant improvement overall in the effluent quality occurred when water hyacinths covered the entire lagoon. During this period the effluent BOD5 and TSS were 23 and 6 mgll, respectively. Without water hyacinths, the effluent BOD5 and TSS were 52 and 77 mgll, respectively. The effluent total organic carbon concentration with water hyacinths averaged 40 mgll, and without water hyacinths, 72 mgll. A discussion of the results from this 3-year study is presented in this paper along with associated problems that were observed when water hyacinths were introduced into the lagoon and altered its behavior from that of a normal facultative lagoon.  相似文献

Soybean (Glycine max (L.) Merr.) genotypes varying in area per nodal unit (usually a trifoliolate) and maturity class were grown in plots at the University of Illinois experimental farm. Leaf CO2-exchange rates per unit area (CER) were measured under sunlight on intact plants. In addition to previously reported correlations with specific leaf weight and chlorophyll, CER was positively correlated with ribulose bisphosphate carboxylase (RuBPcase) activity, specific activity, and soluble protein, and was negatively correlated with area per leaf unit. The CER: chlorophyll correlation was destroyed by high CER values in 2 chlorophyll-deficient lines. CER values for 27 of the 35 lines tested fell within the range of those for isolines of cultivar Clark varying in leaf characteristics. The CER values were highest for fully expanded leaves during rapid pod fill. These results suggested that photoperiod (maturity) genes and genes for leaf area growth interact with genes controlling photosynthetic CO2-exchange to produce the major differences in CER values among soybean genotypes.  相似文献

Polypeptides associated with the rabbit reticulocyte poly(A)-protein complex, isolated by oligo (dT)-cellulose chromatography, were analyzed by SDS-polyacrylamide gel electrophoresis. The complex derived from EDTA released total polysomal mRNP and 20S globin mRNP by RNase treatment contained four polypeptides of molecular weight 58000, 67000, 71000 and 79000. A 79000 molecular weight polypeptide, which was also a major component of the reticulocyte low molecular weight cytoplasmic fraction, was shown to form tenacious associations with poly(A) in vitro.  相似文献

Summary The endothelia of Tubifex tubifex Müller consist of myoendothelial cells, chloragocytes, or podocytes. The latter seem to occur only as windows on the ventral vessel which has an endothelium of myoendothelial cells elsewhere. The podocytes are large cells, with several processes on the inner side which ramify into several pedicels. These are aligned upon the outside of the basement membrane which lines the inside of the endothelium. The gaps between adjacent pedicels are about 40 nm wide. In capillaries fenestrated endothelia occur with irregular spacings measuring up to 0.4–1 m. A diaphragm in podocytes or capillary fenestrations do not seem to exist. The basement membrane is the only continuous layer lining the blood vessels and capillaries of Tubifex with a rather uniform diameter in the range of 50 nm. It is the only permeability barrier between blood and coelomic fluid.  相似文献

Summary A new variant of clinical galactosemia with two hitherto unidentified alleles on the transferase locus in one family is described. This new clinical variant of transferase has 25% of normal control activity in blood and in skin fibroblasts, and the patient accumulates galactose-1-phosphate in blood on an unrestricted galactose diet. Using starch gel electrophoresis on the hemolysate of the family members, a fast-moving transferase with mobility in between those of the normal control and of the Duarte variant is identified. This new allele is designated as (fast-moving Chicago variant). In addition, a second new allele was documented in this family by studying the instability of the transferase enzyme in hemolysates of family members at 50°C for various time intervals. This new allele is designated as (heat-labile Chicago variant). On the basis of the studies, the transferase genotype of this patients is thought to be a double heterozygote compound, /GALTG.  相似文献

The success of colchicine therapy in the management of familial Mediterranean fever has provided new direction to investigations into the pathogenesis of this disease. Examination of HLA antigen frequencies in 53 patients with familial Mediterranean fever and appropriate controls, as well as various immunologic studies have yielded no significant differences. However, B lymphocyte typing and assays for immune complexes, lymphokines and prostaglandins may be of potential interest. Preliminary studies indicate that leukocytes of patients with familial Mediterranean fever release increased amounts of lysozyme (P<0.01), when subjected to high temperatures, and of both lysozyme and myeloperoxidase at low osmotic concentrations. The known and potential effects of colchicine on leukocyte and cellular metabolism, and the current status of colchicine prophylaxis are reviewed. In patients receiving an optimum colchicine dose of 1.5 to 1.8 mg per day, side effects have been minimal and the frequency of attacks has been decreased significantly.  相似文献
