A synthetic dipeptide, N-acetyl-5-hydroxytryptophyl-5-hydroxytryptophan amide, was shown previously to inhibit the binding of serotonin to a soluble specific serotonin binding protein as well as to alter brain serotonin levels. When injected into rats intraventricularly, the dipeptide caused an increase in pain threshold, lasting for several hours, as determined by either a flinch-jump test or a tail-flick test. This effect was reversed by naloxone. The dipeptide is a very weak inhibitor of the binding of labelled naloxone or dihydromorphine to a membranous opiate receptor preparation. The analgesic activity of the dipeptide was not diminished by p-chlorophenylalanine or the setonergic neurotoxin 5,7-dihydroxytryptamine, which depleted brain serotonin levels. This implies that the analgesic action of the dipeptide is not mediated directly by its effect on serotonin concentration. 相似文献

The complex embryonic phenotype of mutations in the faint little ball (flb) locus, encoding the Drosophila EGF receptor homolog (DER), was dissected by temperature shifts of a temperature-sensitive allele. We show that the phenotype can be resolved into at least five components, which are temporally and spatially distinct. Most notably, the central nervous system (CNS) phenotype is determined at two separate phases. A severe collapse results from early defects in the DER-expressing ectodermal cells from which neuroblasts and midline glial cells deaminate. We thus suggest that DER activity is crucial for interactions that occur in the ectoderm at an early stage, and determine the fate of neuronal and glial cell lineages. This finding explains how a severe CNS phenotype is generated in flb embryos, in spite of the absence of expression of the protein in neuronal cells. In a second phase, during germ band retraction, the flb function is required specifically in the three pairs of midline glial cells (MG). In the absence of a functional DER protein, these cells die or fail to differentiate correctly, resulting in a fused commissure phenotype. 相似文献

Hemopoiesis is a sequence of events initiated by the self-renewal of pluripotent stem cells followed by a series of differentiation steps and completed in the formation of distinct tissue patterns. Differentiation and self-renewal are antagonistic processes. A mechanism that attenuates the differentiation flow is obligatory to prevent the exhaustion of the stem cell pool. We suggest that stromal cells from the bone marrow control stem cell renewal through a mechanism that does not require colony-stimulating factors. The organization of cells within the tissue and their specific localization is suggested to be directed by stromal cell activities other than differentiation inducers. This study focuses on patterns of communication and interaction for peer support, which develop among parents of screen-positive children in the socio-medical space created by the diagnostic uncertainty of newborn screening, the limitations of established patient support groups and the daily challenges of screen-positive care management. Based on semi-structured interviews conducted in 2015–2017 with 34 Israeli parents whose child is screen positive, we describe and analyze the spontaneous grouping of parents using the WhatsApp instant messaging service for smartphones. These self-help groups did not reflect an attempt to forge an alternative movement of de-medicalization but rather to provide satisfactory answers to shared problems of care management under diagnostic uncertainty. These self-help groups are further discussed as a form of biosociality and lay expertise where elements of communication such as simplicity, reciprocity, and deliberative democracy serve as a complementary mirror-image of the asymmetrical patient–doctor communication. 相似文献
