α-AMANITIN, a cyclic peptide of the toadstool Amanita phalloides1,2, causes necrosis of liver and kidney cells, the first morphological lesions occurring in the nuclei3,4. It acts by binding to RNA polymerase in eukaryotic .cells and inhibiting the enzyme5–9. The hepatotoxicity of amanitin increases several times when it is conjugated to albumin, probably because of a slower rate of elimination of the toxin through the glomeruli4,10. It is unlikely that the amanitin-albumin conjugate enters the hepatocyte by a mechanism involving its albumin moiety; it was therefore suggested11 that penetration of the liver cells is consequent on binding of the amanitin group to the carrier involved in transport of this peptide. This led us to consider more generally the facilitation of penetration into cells by large molecules by means of binding to another molecule for which a carrier exists on the cell membrane12,13.  相似文献

This is an analytic study of mucous flow caused by ciliary motion. The computed flow data may be compared with that already found exprimentally. The effects of mucous density, viscosity and layer depth on flow phenomena are investigated. The effects of cilia diameter, length, spacing and oscillation frequency are determined from the equations governing the flow of the mucous blanket. A pertinent finding of the analysis is that the mucous flow in the airway tubes can satisfy physical constraints only through the assumption of a variable viscosity in the covering mucous blanket. The mucous viscosity must increase considerably from the low value at the cillium layer to a much higher value at the air-mucus interface.  相似文献

Interactions of certain naturally occurring, amphiphilic polypeptides with membranes were investigated. Mastoparan (wasp venom toxin), melittin (bee venom toxin), cardiotoxin (cobra venom toxin), and polymyxin B (antibacterial antibiotic) inhibited protein kinase C stimulated by phosphatidylserine bilayer or arachidonate monomer and blocked binding of [3H] phorbol 12,13-dibutyrate to protein kinase C in the presence of phosphatidylserine bilayer, with IC50 values (concentrations causing 50% inhibition) of 1-8 microM. Mastoparan and polymyxin B were much less inhibitory (IC50, 10-20 microM), whereas melittin and cardiotoxin were similarly inhibitory (IC50, 1-4 microM), when protein kinase C was activated instead by synaptosomal membrane. Kinetic analysis indicate that mastoparan inhibited protein kinase C, assayed using phosphatidylserine or synaptosomal membrane as the phospholipid cofactor, competitively with the phospholipid cofactor, in a mixed manner with CaCl2 or diacylglycerol, noncompetitively with histone, and uncompetitively with ATP, with apparent Ki values of 1.6-18.7 microM. Inhibition of Na,K-ATPase in the membrane by these polypeptides had relative potencies different from those for their inhibition of protein kinase C activated by the same membrane preparation; mastoparan and melittin inhibited the two activities with comparable potencies, but polymyxin B and cardiotoxin were far less effective in inhibiting Na,K-ATPase. The same relative inhibitory potencies of the polypeptides (melittin greater than mastoparan greater than polymyxin B) for inhibition of Na,K-ATPase were also noted for their inhibition of Ca2+/calmodulin-dependent protein kinase II, 86Rb uptake (Na+ pump) by HL60 cells and the phorbol ester-induced differentiation of the leukemia cells. These findings were consistent with discrete interactions of the polypeptides with functionally distinct sites on the membrane, leading to differential inhibition of biological activities associated with the membrane. Actions of certain polypeptides appeared to be more specific compared to those of lipid second messengers such as lyso-phosphatidylcholine and sphingosine, and the antineoplastic ether lipid analogs such as 1-O-octadecyl-2-methyl-rac-glycero-3-ophosphocholine.  相似文献

The ability of prey to detect predators directly affects their probability of survival. Chemical cues are known to be important for predator detection in aquatic environments, but the role of other potential cues is controversial. We tested for changes in behaviour of Rana temporaria tadpoles in response to chemical, visual, acoustic, and hydraulic cues originating from dragonfly larvae (Aeshna cyanea) and fish (Gasterosteus aculeatus). The greatest reduction in tadpole activity occurred when all cues were available, but activity was also significantly reduced by visual cues only. We did not find evidence for tadpoles lowering their activity in response to acoustic and hydraulic cues. There was no spatial avoidance of predators in our small experimental containers. The results show that anuran larvae indeed use vision for predator detection, while acoustic and hydraulic cues may be less important. Embryo dormancy of Taxus baccata var. fastigiata is eliminated when cultured continuously in nutritive liquid medium. An equivalent percentage of germination is obtained when the embryos are transferred to agar medium after 8 days of liquid culture. There is no morphological development of the embryo during the period in the liquid medium. But we have ascertained that water-soluble germination inhibitors present in the embryo are leached out into the medium, permitting germination. Germination is totally absent when the embryos are cultured continuously in distilled water, alone or with minerals; incidental in sucrose solution; and maximal when the medium contains sucrose and Ca2+ or K+ ions. The extent of germination on agar medium depends upon the composition of the liquid medium in which the embryos are cultured for the initial 8 days. But this preliminary culture in the liquid medium does not always remove the endogenous inhibitors, irrespective of its composition. This can be achieved only in the presence of sucrose; and this process can be made more effective by the addition of Ca2+ ions.  相似文献

The effect of exposure to freezing temperature (?15°C) on leaf phospholipid composition of hardened rye (Secale cereale L.) and hardened wheat cultivars (‘Miranovskaja 808’, ‘Bezostaja 1’, ‘Short Mexican’ and ‘Penjamo 62’), which differ in their resistance to frost, was investigated. Hardening took place under natural conditions. All the seedlings attained an equal level of linolenic acid in their leaves during hardening. Exposure to freezing temperatures resulted in a loss of phosphatidyl choline and accumulation of phosphatidic acid in the leaves. The ratio of phosphatidic acid to phosphatidyl choline, but not the level of poly-unsaturated fatty acids in the leaves, was related to their ability to survive at low temperatures. As freezing injury is caused by the formation of ice crystals in both extra- and intracellular space, it is probable that the plasma membranes of the investigated cultivars differed with respect to their water permeability. It is concluded that the plants, depending on the degree of their resistance to cold, produce an unknown substance of lipidic nature upon exposure to cold, with the aid of which they adjust the transitional state of their membranes to the prevailing temperature and, at the same time, facilitate the efflux of water from the cells.  相似文献
