In the presence of a base such as ammonia liquid HCN (bp 25 °C) polymerizes readily to a black solid from which a yellow-brown powder can be extracted by water and further hydrolyzed to yield-amino acids. These macromolecules could be major components of the dark matter observed on many bodies in the outer solar system. The non-volatile black crust of comet Halley, for example, may consist largely of such polymers, since the original presence on cometary nuclei of frozen volatiles such as methane, ammonia, and water makes them possible sites for the formation and condensed-phase polymerization of hydrogen cyanide. It seems likely, too, that HCN polymers are among the dark —CN bearing solids identified spectroscopically by Cruikshanket al. in the dust of some other comets, on the surfaces of several asteroids of spectral class D, within the rings of Uranus, and covering the dark hemisphere of Saturn's satellite Iapetus. HCN polymerization could account also for the yellow-orange-brown coloration of Jupiter and Saturn, as well as for the orange haze high in Titan's atmosphere. Implications for prebiotic chemistry are profound. Primitive Earth may have been covered by HCN polymers through cometary bombardment or terrestrial synthesis, producing a proteinaceous matrix that promoted the molecular interactions leading to the emergence of life. 相似文献

Isolated rice embryos were used to investigate the regulatory effects of endosperm extracts and pure sugars on the expression of alpha-amylase gene RAmy3D and a sucrose synthase gene homologous to the maize isozyme Ss2. The high-level expression of RAmy3D in the scutella of isolated embryos could be inhibited by a variety of sugars as well as endosperm extracts from germinated rice grains. Glucose, at a concentration of 250 mM, was most effective in repressing RAmy3D mRNA accumulation. Furthermore, this repression was reversible. Interestingly, RAmy3D repression was always accompanied by the induction of sucrose synthase gene expression. These results support a model in which the expression of alpha-amylase and sucrose synthase genes in the rice scutellum are counter-regulated by the influx of sugars from the endosperm. 相似文献

Summary The structure of the basement membrane of the high endothelium of reactive human lymph nodes was investigated by techniques selective for carbohydrates (periodic acid-Schiff; critical electrolyte concentration staining with Alcian Blue; lectin histochemistry), specific proteins (immunohistochemistry for laminin and fibronectin) and by conventional techniques of light and transmission electron microscopy. Adjacent small lymphocytes were assigned to B and T cell subsets by use of monoclonal antibodies and they were analysed for non-specific esterase,-glucuronidase,-N-acetylglucaminidase and proteolytic activities. The basement membranes were shown to be distinctive and to contain three layers, of differing laminin, glycosaminoglycan and glycoprotein oligosaccharide content. Kinetic and equilibrium constants for lactose repressor-operator DNA interaction have been examined as a function of salt concentration, size and sequence context of the operator DNA, and temperature. Significant salt effects were observed on kinetic and equilibrium parameters for pLA 322-8, an operator-containing derivative of pBR 322, and pIQ, an operator and pseudooperator-containing derivative of pBR 322. The association rate constant and equilibrium constant for the 40 base pair operator fragment were also salt dependent. Data for all the DNAs were consistent with a sliding mechanism for repressor-operator association/dissociation [Berg, O. G., & Blomberg, C. (1978) Biophys. Chem. 8, 271-280]. Calculation of the number of ionic interactions based on salt dependence yielded a value of approximately 8 for repressor binding to pIQ and pLA 322-8 vs. approximately 6 for the repressor-40 base pair fragment. These data and the differences in binding parameters for the plasmids vs. the 40 base pair operator are consistent with the formation of an intramolecular ternary complex in the plasmid DNAs. Unusual biphasic temperature dependence was observed in the equilibrium and dissociation rate constants for pLA 322-8, pIQ, and the 40 base pair fragment. These observations coupled with a discontinuity found in the inducer association rate constant as a function of temperature suggest a structural change in the protein. The large positive entropy contributions associated with repressor binding to all the DNAs examined provide the significant driving force for the reaction and are consistent with involvement of ionic and apolar interactions in complex formation. 相似文献

Summary The reproductive strategy of the salamander Ambystoma talpoideum was studied on an age-specific basis in five breeding populations using ponds subjected to different annual probabilities of drying. Ambystoma talpoideum is facultatively paedomorphic in semi-permanent ponds but sexually mature individuals occur only as terrestrial morphs in temporary ponds. Larvae of paedomorphs and terrestrial morphs mature at the same age but different body sizes. Body mass and snout-vent length increased with age in both morphs. One-year old terrestrial morphs had larger snoutvent lengths than 1-year-old paedomorphs but were smaller in body mass. Clutch mass, number of ova, and diameter of ova increased with snout-vent length. After removing the effects of snout-vent length with an analysis of covariance, only the diameter of ova and the resultant clutch mass increased with age. One-year-old terrestrial morphs produced more ova than 1-year-old paedomorphs but paedomorphs have the potential to reproduce earlier than terrestrial morphs. Parental investment, as shown by the relationship between clutch size and egg size, increased with age. This suggested that as potential energy increased with age, A. talpoideum produced more eggs and larger eggs. Rather than optimizing egg size, individuals may produce a range of egg sizes that theoretically fare better in spatially and temporally varying environments. 相似文献

This work describes a neutral and alkaline elution method for measuring DNA single-strand breaks (SSBs), DNA double-strand breaks (DSBs), and DNA-DNA crosslinks in rat testicular germ cells after treatments in vivo or in vitro with both chemical mutagens and gamma-irradiation. The methods depend upon the isolation of testicular germ cells by collagenase and trypsin digestion, followed by filtration and centrifugation.
137Cs irradiation induced both DNA SSBs and DSBs in germ cells held on ice in vitro. Irradiation of the whole animal indicated that both types of DNA breaks are induced in vivo and can be repaired. A number of germ cell mutagens induced either DNA SSBs, DSBs, or cross-links after in vivo and in vitro dosing. These chemicals included methyl methane sulfonate, ethyl methane sulfonate, ethyl nitrosurea, dibromochlorpropane, ethylene dibromide, triethylene melamine, and mitomycin C. These results suggest that the blood-testes barrier is relatively ineffective for these mutagens, which may explain in part their in vivo mutagenic potency.This assay should be a useful screen for detecting chemical attack upon male germ-cell DNA and thus, it should help in the assessment of the mutagenic risk of chemicals. In addition, this approach can be used to study the processes of SSB, DSB, and crosslink repair in DNA of male germ cells, either from all stages or specific stages of development.Abbreviations DBCP

Changes with time in the distribution of virus and PR protein around single local lesions of TMV infected tobacco 总被引:1,自引:0,他引:1
Summary ELISA was used to determine PR la protein and TMV accumulation in local necrotic lesions produced on salicylic acid and water sprayed Nicotiana tabacum cv Xanthi-nc leaves. The amount of PR la protein produced is the result of an interaction between the salicylic acid treatment and lesion growth. The implication of these observations for experiments investigating the relationship between PR proteins and resistance are discussed.The distribution of TMV and PR la protein in and around single local necrotic lesions up to 14 days after inoculation was measured by ELISA. The highest concentration of TMV was in the centre of the lesion and decreased rapidly with distance from the centre. In contrast there was very little PR la protein in the centre of the lesion, the largest amounts were just outside the centre, and the concentration then decreased with distance from the centre. This is the distribution that might be expected for a substance closely associated with the restriction of virus spread. 相似文献
