Ouabain binding to erythrocyte membranes is increased in obese subjects. Three study groups are compared: 14 reference subjects, 102 +/- 16% of ideal weight; 9 obese on unrestricted diets, 207 +/- 16% of ideal weight; 11 obese on restricted diets, 202 +/- 35% of ideal weight. A reproducible (CV = 11.3%) ouabain-binding assay is used to measure Na+-K+ ATPase sites in erythrocyte membranes. The number of binding sites per red blood cell for obese subjects on unrestricted diets, 431 +/- 30, is greater than for the reference group, 346 +/- 66 (p less than 0.01), or for obese subjects on restricted diets, 371 +/- 68 (p less than 0.05). These data suggest that caloric intake influences the number of Na+-K+ ATPase sites. Scatchard plots indicate only one type of binding site for ouabain with an affinity constant of about 3 X 10(8) M-1. 相似文献

Summary Quantitative analysis of the X-linked enzyme, glucose 6-phosphate dehydrogenase (G-6-PD), was performed on tumor cells lines
from two human females. Both tumor cells were hyperdiploid, having complete or redundant C+X groups. One, no. 930, lacked
the X chromatin body and exhibited twice the level of G-6-PD as in the X chromatin-positive tumor cells, ME-180. Hence, in
the no. 930 cell, reversal of X chromosomal condensation was associated with loss of the X chromatin body and doubling of
genetic activity.
Cells of no. 930 were subsequently placed in culture where after three passages they developed an X chromatin body (or bodies).
G-6-PD determinations made at that time showed enzyme levels comparable to the X chromatin-positive tumor cells (ME-180).

Biological selectivity is shown to vary with medium osmotic strength and temperature. Selectivity reversals occur at 4°C and at an external osmolality of 0.800 indicating that intracellular hydration and endosolvent (intracellular water) structure are important determinants in selectivity. Magnetic resonance measurements of line width by steady-state nuclear magnetic resonance (NMR) indicate a difference in the intracellular water signal of 16 Hz between the K form and Na form of Escherichia coli, providing additional evidence that changes in the ionic composition of cells are accompanied by changes in endosolvent structure. The changes were found to be consistent with the thermodynamic and magnetic resonance properties of aqueous electrolyte solutions. Calculation of the dependence of ion-pairing forces on medium dielectric reinforces the role of endosolvent structure in determining ion exchange selectivity. 相似文献

Summary Crystalline inclusions in parathyroid gland cell nuclei of Rana temporaria were studied by electron microscopy using a specimen tilting stage. Images were analysed by optical diffraction. The eruption and wear of the cheek teeth of male Fallow deer from a park and wild population has been studied for age determination purposes. It was found possible to age accurately up to 48 months animals whose date of death was known, whether they were from a wild or park population. It was found that up to at least this age individual and environmental differences affecting animals born in successive years did not mask those resulting fromthe restricted seasonal breeding of this species.
