Plasma hormone levels were determined at frequent intervals in 12 two-year-old Hereford heifers after treatment with prostaglandin F2α on day 267 of gestation to induce parturition. Of the 12 animals, six also received a daily injection of estradiol-17β until calving occurred. An additional six heifers received no treatment and therefore served as a control group. The treatment with PGF2α was effective since the treated heifers calved a mean of 12 days sooner than the untreated heifers (P < .01). A metabolite of PGF2α, 13, 14-dihydro-15-keto PGF2α, rose to a peak level in the plasma within one hour after injection (im) of PGF2α then declined to a baseline level within four hours. The plasma progesterone level had fallen significantly within two hours after injection, but no change in plasma estradiol-17β or estrone was observed. A Butyrivibrio fibrisolvens H17c glgB gene, was isolated by direct selection for colonies that produced clearing on starch azure plates. The gene was expressed in Escherichia coli from its own promoter. The glgB gene consisted of an open reading frame of 1,920 bp encoding a protein of 639 amino acids (calculated Mr, 73,875) with 46 to 50% sequence homology with other branching enzymes. A limited region of 12 amino acids showed sequence similarity to amylases and glucanotransferases. The B. fibrisolvens branching enzyme was not able to hydrolyze starch but stimulated phosphorylase alpha-mediated incorporation of glucose into alpha-1,4-glucan polymer 13.4-fold. The branching enzyme was purified to homogeneity by a simple two-step procedure; N-terminal sequence and amino acid composition determinations confirmed the deduced translational start and amino acid sequence of the open reading frame. The enzymatic properties of the purified enzyme were investigated. The enzyme transferred chains of 5 to 10 (optimum, 7) glucose units, using amylose and amylopetin as substrates, to produce a highly branched polymer. 相似文献

The pattern and opioidergic control of the secretion of gonadotrophins in prepubertal heifer calves were examined. Ten age-matched Hereford heifer calves were weighed and a blood sample was taken every 2 weeks from 2 to 25 weeks of age and then weekly until 60 weeks of age. At 60 weeks, a fertile bull was introduced and at 75 weeks of age pregnancy diagnosis was performed by transrectal ultrasonography. At 4, 12, 18, 24 and 32 weeks of age, the opioid antagonist naloxone was injected (i.v., n = 5; 1 mg kg-1 body weight) each hour for 12 h. Control heifers received sterile saline at the same ages. Blood samples were collected every 12 min for the 12 h treatment and serum samples were analysed for luteinizing hormone (LH) and follicle-stimulating hormone (FSH). Samples taken once every 2 weeks from 2 to 60 weeks were analysed for LH, FSH and oestradiol, and weekly samples were taken for progesterone determination. There was no effect of naloxone on the age at puberty, which was 56.2 +/- 0.7 weeks at a body weight of 388.5 +/- 8.0 kg. The mean age at conception was 63.4 +/- 0.5 weeks. On the basis of samples taken every other week, serum concentrations of LH were high at 10 weeks and between 40 and 60 weeks of age. Static pulmonary and total compliance during expiration was measured in fifteen healthy anesthetized ponies. Mean body weight of the ponies was 140.6 kg and ranged from 83.6 to 211.4 kg. For the fifteen ponies, mean values and SD were: total compliance (Ct), 168.7 plus or minus 47.2 ml/CMH2O, and mean pulmonary compliance (Cl), 368.4 plus or minus 90.3 ml/cmH2O. Both Ct and Cl were significantly dependent on body weight, calculated surface area, height at the withers, and circumference about the chest.
