The present study was undertaken to elucidate the effect of alpha-linolenic acid (ALA, 18:3, ω-3) and gamma-linolenic acid (GLA, 18:3, ω-6) on experimental autism features induced by early prenatal exposure to valproic acid (VPA) in albino wistar pups. The pups were scrutinized on the accounts of behavioral, biochemical, and inflammatory markers, and the results suggested that the GLA can impart significant protection in comparison to ALA against VPA-induced autism features. When scrutinized histopathologically, the cerebellum of the GLA-treated animals was evident for more marked protection toward neuronal degeneration and neuronal loss in comparison to ALA. Concomitant administration of ALA and GLA with VPA demonstrated a marked cutdown in the Pgp 9.5 expression with GLA having more pronounced effect. Henceforth, it can be concluded that ALA and GLA can impart favorable protection against the VPA-induced autism-like features with GLA having pronounced effect.  相似文献

The intracellular redox environment of Staphylococcus aureus is mainly buffered by bacillithiol (BSH), a low molecular weight thiol. The identity of enzymes responsible for the recycling of oxidized bacillithiol disulfide (BSSB) to the reduced form (BSH) remains elusive. We examined YpdA, a putative bacillithiol reductase, for its role in maintaining intracellular redox homeostasis. The ypdA mutant showed increased levels of BSSB and a lower bacillithiol redox ratio vs. the isogenic parent, indicating a higher level of oxidative stress within the bacterial cytosol. We showed that YpdA consumed NAD(P)H; and YpdA protein levels were augmented in response to stress. Wild type strains overexpressing YpdA showed increased tolerance to oxidants and electrophilic agents. Importantly, YpdA overexpression in the parental strain caused an increase in BSH levels accompanied by a decrease in BSSB concentration in the presence of stress, resulting in an increase in bacillithiol redox ratio vs. the vector control. Additionally, the ypdA mutant exhibited decreased survival in human neutrophils (PMNs) as compared with the parent, while YpdA overexpression protected the resulting strain from oxidative stress in vitro and from killing by human neutrophils ex vivo. Taken together, these data present a new role for YpdA in S. aureus physiology and virulence through the bacillithiol system.  相似文献

Introduction

Widespread food insecurity in Africa continues to compromise an effective response to the AIDS epidemic. Health-related quality of life (HRQoL) is a comprehensive indicator of physical, mental, and social well-being that is associated with food insecurity and increasingly used to assess the well-being of people living with HIV/AIDS (PLHIV). We examined the impact of a food assistance intervention, previously shown to have reduced household food insecurity and improved nutritional status, on HRQoL of PLHIV.

Methods

We capitalized on an existing intervention targeting antiretroviral therapy (ART)- naïve PLHIV in Uganda, and conducted a prospective impact evaluation including a treatment and a comparison group. Data analyzed included 640 participants from two districts (318 in the intervention district) interviewed in both clinic and household settings at baseline and again approximately one year later. The main outcomes considered were physical and mental health dimensions of HRQoL, and other outcomes included self- and healthcare provider-reported symptoms. We utilized difference-in-difference propensity score matching methodologies to infer causality and examine program impacts.

Results

Over 12 months, food assistance significantly increased physical health scores (PHS) by 2.85 (P < .01) or approximately 0.35 SD, and reduced substantially the number of self- and healthcare provider-reported HIV-related symptoms by 3.83 and 2.68, respectively (P < .01). There was no significant impact, however, on mental health scores (MHS).

Conclusions

This study demonstrates the potential importance for HRQoL of including food assistance programming as part of the standard of care for PLHIV in areas of widespread food insecurity.  相似文献

Hepatocellular carcinoma (HCC) is the fifth most commonly diagnosed cancer worldwide and is associated with poor prognosis. The current study aimed to assess the therapeutic efficacy of resveratrol when administered alone and in combination with nicotinamide against alcohol‐aflatoxin B1‐induced HCC. Results reveal that during the development and progression of cancer, there was a decline in the level of antioxidant enzymes catalase, glutathione peroxidase, glutathione reductase (GR), antioxidant glutathione, and glutathione S‐transferase, which is an enzyme of detoxification pathways. Treatment of resveratrol restored the level of catalase and glutathione peroxidase toward normal in alcohol‐aflatoxin B1‐induced HCC; however, nicotinamide worked in concert with resveratrol only in upregulating the activity of glutathione reductase, glutathione level, and glutathione S‐transferase. SIRT1 agonist resveratrol was observed to modulate the activity of antioxidant enzymes by negatively regulating the expression of nuclear factor‐κB (NF‐κB) in alcohol‐aflatoxin B1‐induced HCC, thereby suggesting a cross‐talk between antioxidant enzymes SIRT1 and NF‐κB during the development and progression of HCC and its therapeutics by resveratrol and nicotinamide.  相似文献

Dietary deficiency of iron and zinc micronutrients affects more than two billion people worldwide. Breeding for micronutrient-dense crops is the most sustainable and cost-effective approach for alleviation of micronutrient malnutrition. Three accessions of Aegilops peregrina (Hack.) Maire & Weill (2n = 28, UPUPSPSP), selected for high grain iron and zinc concentration were crossed with Triticum aestivum L. cv. Chinese Spring (Ph I ). The sterile F1 hybrids were backcrossed with elite wheat cultivars to get fertile BC2F2 derivatives. Some of the fertile BC2F2 derivatives showed nearly 100% increase in grain iron and more than 200% increase in grain zinc concentration compared to the recipient wheat cultivars. The development of derivatives with significantly higher grain micronutrients, high thousand-grain weight and harvest index suggests that the enhanced micronutrient concentration is due to the distinct genetic system of Ae. peregrina and not to the concentration effect. Genomic in situ hybridization, comparison of introgressed chromosomes with the standard karyotype of Ae. peregrina and simple sequence repeat marker analysis revealed the introgression of 7SP chromosomes in five selected derivatives, 7UP in four, group 4 and 4SP in three and a translocated 5UP of Ae. peregrina in one of the selected derivatives. Molecular marker analysis using the introgressed chromosome markers indicated that two of the BC2F3 progenies were stabilized as disomic addition lines. It could, therefore, be concluded that the group 4 and 7 chromosomes of Ae. peregrina carry the genes for high grain iron and zinc concentration.  相似文献

The present investigation was carried out to isolate bacterial strains from soil/mud samples of metal-polluted environment to check whether the natural adaptation of microbes has equipped them for bioremediation of toxic heavy metals. The primary and secondary screening resulted in 50 mesophilic autotrophic isolates of microbial consortium adapted for metal tolerance and bioadsorption potentiality. The multimetal tolerance in bacterial strain was developed by sequential transfer to higher concentrations of Cd, Cr, Cu, Pb, Ni, and Zn. The isolates were checked for their biosolubilization potential with copper-containing metal sulfide ores, viz. chalcopyrite exhibited 64% and covellite 54% solubilization in the presence of 10?3 M multiple heavy metals on the fifth day at 35°C and pH 6.0. Metal adsorption of highly potential isolate, i.e., Paenibacillus validus MP5, studied by inductively coupled plasma optical emission spectroscopy (ICP-OES), showed maximum adsorption of Zn 27%, followed by Ni and Cd 16%, Cr 15%, Co 9%, and Pb 7.5% in chalcopyrite, which suggested its possible role in decontamination of metal-polluted sites.  相似文献
