Melia dubia, a fast-growing tree species with multi-various uses, is suitable species for agro and farm forestry. In an extensive survey in eight districts of Karnataka, India, it was observed that the majority of M. dubia natural populations are fragmented and tree number is very less. Sixty trees were selected to assess the genetic variability using SSR markers. Analysis revealed a moderate level of genetic diversity (Ho?=?0.47; He?=?0.69) and inbreeding as evinced by overall positive fixation index of 0.30. Mean Fst observed was 0.16 indicating moderate genetic differentiation among populations. The analysis of molecular variance ascribed 99% of total genetic diversity to within population variation. Cluster analysis by unweighted pair group method and genetic differentiation through structure analysis did not differentiate 10 populations as per geographic location, showing that moderate genetic diversity was not due to geographic distance. As genetic diversity was more within population, selection of individual plants would be more effective for genetic improvement to capture the natural variation within the population. An understanding of genetic diversity and differentiation of M. dubia natural populations will help in exploiting the genetic resource for tree improvement and also in formulating management and conservation strategies for this species.
In the present study, we examined the synergistic effect of phosphate solubilizing bacteria (PSB) and the chemical phosphate on the growth, photosynthetic efficiency, phosphorus (P) uptake, antioxidant activity, and yield of upland rice. Three effective bacterial strains with potent P solubilizing activity viz., Bacillus licheniformis (688.18 µg ml?1), Pantoea dispersa (570.90 µg ml?1), and Staphylococcus sp. (551.81 µg ml?1), were isolated from rice rhizosphere to study their impact on upland rice growth and yield under field conditions for two consecutive years. The experiment data revealed significant increments in shoot height, shoot dry weight, total chlorophyll, carotenoid, chlorophyll fluorescence (fv/fm), P uptake, antioxidant activity, and yield characteristics in upland rice treated with individual PSB or their consortia alone, compared to uninoculated control. However, the integrated use of PSB with 50% recommended P dose showed maximum increment in growth indices (21.25%, 21.86% increase in shoot length and 87.18%, 97.06% increase in shoot dry weight), P uptake (110.37%, 122.78% increase), and yield (50.58%, 35.64% increase) compared to uninoculated control for 2018 and 2019, respectively, indicating a reduction in the dependence of chemical P fertilizer by 50%. Therefore, it can be concluded that combined application of PSB and 50% recommended dose of chemical P can be implied for the sustainable cultivation of upland rice systems to give maximum benefits to the farmers and the environment.
Series of short amino terminal modified cationic peptides were designed and synthesized. All of the synthesized compounds were tested against gram-positive as well as gram-negative bacterial strain. Ionically bound peroxidases (POD) were salt extracted from the pulp of four Indian apple varieties, i.e., Golden delicious HP, Golden delicious JK, Red delicious, and Royal delicious. They were precipitated with chilled ethanol. Thermal treatments of partially purified enzymes were given from 40-70 degrees C for 30 minutes. Golden delicious HP peroxidase showed thermostability at 60 degrees C, while three other peroxidases were observed at 50 degrees C. Phenolic compounds (i.e., caffeic acid, ferulic acids, p-coumaric acid, protocatechuic acid) and metal ions (i.e., Cu2+ and Fe2+) activated all apple peroxidases. However, Mn2+ inhibited the peroxidases from Golden delicious HP, Golden delicious JK, and Red delicious, and a substantial increase was observed in Royal delicious peroxidase. Mg2+ inhibited the peroxidases from Golden delicious HP and Red delicious, but marginal activation was reported in peroxidases from Golden delicious JK and Royal delicious. Zn2+ established stimulation in Golden delicious HP and Golden delicious JK peroxidases, but inhibition was observed in peroxidases in Red delicious and Royal delicious.. The present study deals with the structure and functioning ofthree different forest communities, viz., horse chestnut, silverfir and kharsu oak forests, in a high altitude region of CentralHimalaya. The tree density and total basal cover of horse chestnutforest was 280 and 76, silver fir forest 355 and 106, and kharsuoak forest 480 trees ha-1 and 73 m2 ha-1, respectively. Allometricequations relating biomass of different tree components to cbh(circumference at breast height) were significant. Total vegetationbiomass was 505 t ha-1 in horse chestnut, 566 t ha-1 in silverfir and 593 t ha-1 in kharsu oak forests, of which maximum contributionwas by tree layer followed by shrub, herb, sapling and seedlinglayers. The forest floor litter biomass was 2·1, 4·7and 4·2 t ha-1 in horse chestnut, silver fir and kharsuoak forests, respectively. The total litter fall was 7·3,6·7 and 9·4 t ha-1 year-1, of which leaf littercontributed 48, 39 and 64% in horse chestnut, silver fir andkharsu oak forests, respectively. Turnover rate of tree litterwas 0·80 in horse chestnut, 0·61 in silver firand 0·71 in kharsu oak forests. Net primary productionof total vegetation was 19·6, 18·9 and 24·9t ha-1 year-1, of which tree layer contributed maximum proportionfollowed by herb, shrub, sapling and seedling layers. Persistent hypoxic pulmonary vasoconstriction (HPV) plays a significant role in the pathogenesis of pulmonary hypertension, which is an emerging clinical problem around the world. We recently showed that hypoxia-induced activation of glucose-6-phosphate dehydrogenase (Glc-6-PD) in pulmonary artery smooth muscle links metabolic changes within smooth muscle cells to HPV and that inhibition of Glc-6PD reduces acute HPV. Here, we demonstrate that exposing pulmonary arterial rings to hypoxia (20-30 Torr) for 12 h in vitro significantly (P < 0.05) reduces (by 30-50%) SM22α and smooth muscle myosin heavy chain expression and evokes HPV. Glc-6-PD activity was also elevated in hypoxic pulmonary arteries. Inhibition of Glc-6-PD activity prevented the hypoxia-induced reduction in SM22α expression and inhibited HPV by 80-90% (P < 0.05). Furthermore, Glc-6-PD and protein kinase G (PKG) formed a complex in pulmonary artery, and Glc-6-PD inhibition increased PKG-mediated phosphorylation of VASP (p-VASP). In turn, increasing PKG activity upregulated SM22α expression and attenuated HPV evoked by Glc-6-PD inhibition. Increasing passive tension (from 0.8 to 3.0 g) in hypoxic arteries for 12 h reduced Glc-6-PD, increased p-VASP and SM22α levels, and inhibited HPV. The present findings indicate that increases in Glc-6-PD activity influence PKG activity and smooth muscle cell phenotype proteins, all of which affect pulmonary artery contractility and remodeling. 相似文献
The effect of different molar ratios of polyethylene glycol (PEG) on the conformational stability of protein, bovine serum albumin (BSA), was studied. The binding of PEG with BSA was observed by fluorescence spectroscopy by measuring the fluorescence intensity after displacement of PEG with chromophore ANS and had further been confirmed by measuring the intrinsic fluorescence of tryptophan residues of BSA. Co-lyophilization of BSA with PEG at optimum BSA:PEG molar ratio led to the formation of the stable protein particles. Circular dichroism (CD) spectroscopy study suggested that a conformational change had occurred in the protein after PEG interaction and demonstrated the highest stability of protein at the optimum BSA:PEG molar ratio of 1:0.75. Additional differential scanning calorimetry (DSC) study suggested strong binding of PEG to protein leading to thermal stability at optimum molar ratio. Molecular mechanism operating behind the polyethylene glycol (PEG) mediated stabilization of the protein suggested that strong physical adsorption of PEG on the hydrophobic core of the protein (BSA) along with surface adsorption led to the stability of protein. 相似文献
UDP-glycosyltransferases (EC 2.4.1.x; UGTs) are enzymes coded by an important gene family of higher plants. They are involved in the modification of secondary metabolites, phytohormones, and xenobiotics by transfer of sugar moieties from an activated nucleotide molecule to a wide range of acceptors. This modification regulates various functions like detoxification of xenobiotics, hormone homeostasis, and biosynthesis of secondary metabolites. Here, we describe the identification of 96 UGT genes in Cicer arietinum (CaUGT) and report their tissue-specific differential expression based on publically available RNA-seq and expressed sequence tag data. This analysis has established medium to high expression of 84 CaUGTs and low expression of 12 CaUGTs. We identified several closely related orthologs of CaUGTs in other genomes and compared their exon-intron arrangement. An attempt was made to assign functional specificity to chickpea UGTs by comparing substrate binding sites with experimentally determined specificity. These findings will assist in precise selection of candidate genes for various applications and understanding functional genomics of chickpea. 相似文献
