We studied the effect of triacontanol (TRIA) on shoot multiplication and rooting of in vitro derived shoot tips of Capsicum frutescens and Decalepis hamiltonii W & A. In both shoot multiplication and rooting phases, TRIA was administered at 2-20 g l–1. TRIA resulted in highest promotion of axillary shoot proliferation at 2 g l–1 in Capsicum frutescens and 20 g l–1 in Decalepis hamiltonii while rooting was maximum at 5 and 10 g l–1 for Capsicum frutescens and Decalepis hamiltonii respectively. TRIA enhanced shoot growth and chlorophyll content of leaves and also influenced root induction and supported growth of the roots. This work reveals that TRIA can be used as an effective growth regulator in the micropropagation of Capsicum frutescens and Decalepis hamiltonii, an endangered shrub of Deccan peninsular India.  相似文献

An efficient and highly reproducible protocol for micropropagation of bird eye chilli Capsicum frutescens was attempted. Murashige and Skoog (MS) medium containing 0.5-3.0 mgl(-1) of 6-benzyladenine (BA), 2-isopentenyl adenine (2iP), kinetin and 0.5-2.0 mg l(-1) of indole-3-acetic acid (IAA), indole-3-butyric acid (IBA) along with 1 gl(-1) activated charcoal (AC) were used for shoot regeneration from both shoot tip and nodal explants. Shoot tip explants (100%) grew well on medium containing 1 mgl(-1) of kinetin and 1 mgl(-1) of IBA. Shoot proliferation (1-3) from nodal explants was effective on this medium. The regenerated shoots with 4-7 nodes had further growth upon sub-culturing onto kinetin (1 mgl(-1)) and IBA (1 mgl(-1)) and rooted simultaneously. The rooted plants were transferred to pots after hardening under controlled conditions. The survival percentage in pots was 80-90%.  相似文献

Axillary buds obtained from field grown plants of D. hamiltonii were used to initiate multiple shoots on Murashige and Skoog's medium (MS) supplemented with 2 mg L(-1) 6-benzyl aminopurine (BA) and 0.5 mg L(-1) indole-3-acetic acid (IAA). Profuse rooting was achieved when the actively growing shoots were cultured on MS medium supplemented with 1.0 mg l(-1) indole-3-butyric acid (IBA). Regenerated plants were grown successfully in the plains, in contrast to wild growth in high altitudes and rocky crevices of hilly regions. Roots of different sizes from one-year-old tissue culture raised field grown plants had the same profile of 2-hydroxy-4-methoxybenzaldehyde as that of wild plants. A maximum of 0.14% and 0.12% 2-hydroxy-4-methoxybenzaldehyde was produced in roots of one year old tissue culture derived plants and greenhouse grown plants respectively.  相似文献

Although the bacterium E. coli is chosen as the host in many bioprocesses, products derived from the central aerobic metabolic pathway often compete with the acetate-producing pathways poxB and ackA-pta for glucose as the substrate. As such, a significant portion of the glucose may be excreted as acetate, wasting substrate that could have otherwise been used for the desired product. The production of the ester isoamyl acetate from acetyl-CoA by ATF2, a yeast alcohol acetyl transferase, was used as a model system to demonstrate the beneficial effects of reducing acetate production. All strains tested for ester production also overexpressed panK, a native E. coli gene that previous studies have shown to increase free intracellular CoA levels when fed with pantothenic acid. A recombinant E. coli strain with a deletion in ackA-pta produces less acetate and more isoamyl acetate than the wild-type E. coli strain. When both acetate-producing pathways were deleted, the acetate production was greatly reduced. However, pyruvate began to accumulate, so that the overall ester production remained largely unchanged. To produce more ester, a previously established strategy of increasing the flux from pyruvate to acetyl-CoA was adopted by overexpressing pyruvate dehydrogenase. The ester production was then 80% higher in the poxB, ackA-pta strain (0.18 mM) than that found in the single ackA-pta mutant (0.10 mM), which also overexpressed PDH.  相似文献

Perovskite solar cells (PSCs) exhibit a series of distinctive features in their optoelectronic response which have a crucial influence on the performance, particularly for long‐time response. Here, a survey of recent advances both in device simulation and optoelectronic and photovoltaic responses is provided, with the aim of comprehensively covering recent advances. Device simulations are included with clarifying discussions about the implications of classical drift–diffusion modeling and the inclusion of ionic charged layers near the outer carrier selective contacts. The outcomes of several transient techniques are summarized, along with the discussion of impedance and capacitive responses upon variation of bias voltage and irradiance level. In relation to the capacitive response, a discussion on the J–V curve hysteresis is also included. Although alternative models and explanations are included in the discussion, the review relies upon a key mechanism able to yield most of the rich experimental responses. Particularly for state‐of‐the‐art solar cells exhibiting efficiencies around or exceeding 20%, outer interfaces play a determining role on the PSC's performance. The ionic and electronic kinetics in the vicinity of the interfaces, coupled to surface recombination and carrier extraction mechanisms, should be carefully explored to progress further in performance enhancement.  相似文献

Plasmodium falciparum harbors an essential relict plastid called the apicoplast that is involved in several important biosynthetic processes. Over 500 nuclear encoded proteins are imported into the organelle that is now recognized as an important therapeutic target. These proteins contain an N-terminal transit peptide sequence essential for apicoplast targeting during which the P. falciparum Hsp70-1 plays an important role. In the present study, we have focused on the in vitro interactions of PfHsp70-1 with synthetic peptides endowed with transit peptide like features. The peptides exhibit higher affinity for PfHsp70-1 in the presence of ADP compared to ATP. The results highlight the positional importance of selected residues in the designed peptides for affinity. They suggest that better peptide affinity for the protein requires a Lys at second position, retention of aromatic residue at the last position, and absence of acidic residues at any position in the transit peptides. Overall, the present work is the first in vitro fluorescence-based study of PfHsp70-1 with peptides possessing transit peptide-like features.  相似文献
