The effects of different NaCl concentrations on the growth andnitrogen fixation activity of white lupin (Lupinus albus [L.])was studied over a 6 d period. Plant growth parameters, photosynthesisand shoot respiration were unaffected by NaCl concentrationsup to 150 mol m–3. However, nitrogenase activity decreasedwith increased NaCl concentration up to 100 mol m–3, whilstthe O2 diffusion resistance increased with 100 mol m–3NaCl, but showed no further change when 150 mol m–3 NaClwas applied for 6 d. Increases in NaCl concentration decreasednodular starch content while increasing sucrose content, suggestingan osmotic regulation. These changes were associated with a77% decrease in sucrose synthase activity. The effect on theO2 diffusion resistance was paralleled by changes in glycoproteincontent of the nodules, as determined by immunogold localizationand ELISA. X-ray microanalysis studies of nodules showed that,following a 6 d exposure to 150 mol m–3 NaCl, Na+ ionswere largely excluded from the infected zone, whilst only lowlevels of Cl- ions penetrated into this region. Na+ entry intoroots and leaves was also at a low level. Leghaemoglobin contentdecreased with saline stress, as did superoxide dismutase; whichdecreased by 36% following exposure to 100 mol m–3 saltfor 6 d. These results are discussed in relation to the relativesalt tolerance of the Multolupa/ISLU-16 symbiosis. Leishmaniasis, a multi-faceted ethereal disease is considered to be one of the World's major communicable diseases that demands exhaustive research and control measures. The substantial data on these protozoan parasites has not been utilized completely to develop potential therapeutic strategies against Leishmaniasis. Dihydrofolate reductase thymidylate synthase (DHFR-TS) plays a major role in the infective state of the parasite and hence the DHFR-TS based drugs remains of much interest to researchers working on Leishmaniasis. Although, crystal structures of DHFR-TS from different species including Plasmodium falciparum and Trypanosoma cruzi are available, the experimentally determined structure of the Leishmania major DHFR-TS has not yet been reported in the Protein Data Bank. A high quality three dimensional structure of L.major DHFR-TS has been modeled through the homology modeling approach. Carefully refined and the energy minimized structure of the modeled protein was validated using a number of structure validation programs to confirm its structure quality. The modeled protein structure was used in the process of structure based virtual screening to figure out a potential lead structure against DHFR TS. The lead molecule identified has a binding affinity of 0.51?nM and clearly follows drug like properties.  相似文献

Tea plants (Camellia sinensis) contain 5-6% caffeine that is responsible for the stimulating effect of the beverage. As the tolerance to caffeine varies among individuals, low caffeine tea would be an ideal alternative. While assessing the potential of a few selected bacteria-Bacillus licheniformis, B. subtilis and B. firmus, to multiply on nutrient medium supplemented with glucose (5%) and tea leaf extract (2%), it was observed that only B. licheniformis could proliferate on this medium. Hence, B. licheniformis was used for further studies. Tea plants were sprayed with a suspension of B. licheniformis at a dilution of 5 x 10(8) CFU/ml containing 0.1% Tween 80 as surfactant. In situ lowering of caffeine from tea leaves was evident without affecting the quality of the other tea components. Further, there was no change in the morphological and physiological characteristics as well. It is suggested that spraying of B. licheniformis may be useful in yielding decaffeinated tea with good flavour and aroma.  相似文献

Groups of albino rats were fed low and high protein diets and the former group was fed with different supplements. The supplements used were niacin, pyridoxine and glutamic acid. A low protein diet was found to decrease the activities of l -glutamate-NAD-oxidoreductase and l -glutamate-1-carboxy-lyase in the cerebrum; this confirmed the results of previous work in this laboratory. Addition of glutamic acid to a low protein diet was found to reverse the effects of a low protein diet to some extent. Groups of animals were fed kodri (Paspalum scorbiculatum), a millet deficient in lysine, with or without lysine itself or foodstuffs rich in it, e.g. moth bean (Phaseolus aconitifolius Jacq.), peas (Pisum sativuni) and skim milk powder. When combined, the groups fed on the lysine or lysine-source supplemented diets were found to perform better in a water maze on the basis of the relative ease with which they could reverse a previous learned pattern. They also had higher activities of l -glutamate-NAD-oxidoreductase and l -glutamate-1-carboxy-lyase than groups with no lysine or lysine source added. There was no significant difference between the groups receiving the different lysine supplements.  相似文献

We have shown that lipopolyamines bind to the lipid A moiety of lipopolysaccharide, a constituent of Gram-negative bacterial membranes, and neutralize its toxicity in animal models of endotoxic shock. In an effort to identify non-polyamine scaffolds with similar endotoxin-recognizing features, we had observed an unusually high frequency of hits containing guanylhydrazone scaffolds in high-throughput screens. We now describe the syntheses and preliminary structure-activity relationships in a homologous series of bis-guanylhydrazone compounds decorated with hydrophobic functionalities. These first-generation compounds bind and neutralize lipopolysaccharide with a potency comparable to that of polymyxin B, a peptide antibiotic known to sequester LPS.  相似文献

 Mangroves, the intertidal ecosystems occurring primarily in the tropical regions of the world, are valuable natural resources with high productivity and unique habitat value. However, the genetic structure of plant species within the mangrove ecosystem is poorly understood. The present communication is the first report on the use of molecular markers in assessing intra-site and intra-specific polymorphism in one of the mangrove species, Acanthus ilicifolius, for identifying/ detecting distinct genotypes for long-term conservation. Random amplified polymorphic DNAs (RAPDs) and restriction fragment length polymorphisms (RFLPs) were used to elucidate the intra- and inter-population variability in this widely distributed mangrove species. In all, 48 genotypes representing eight distinct populations were analysed. A low level of polymorphism was detected at the intra-population level through both RAPD (3.8–7.3%) and RFLP (3.2–9.1%) analyses. At the inter-population level, 25 of the 73 RAPD loci (34%) detected through the use of 13 random primers and 44 of the 96 RFLP loci (45.8%) revealed through 15 probe/enzyme combinations were polymorphic. RFLP analyses were carried out using genomic clones developed from the same species. The somatic cells of the species displayed 48 chromosomes, with no numerical changes at either intra- or inter- population levels. Received: 5 June 1996 / Accepted: 8 November 1996  相似文献
