Cyclo(l-Pro)3 (CP3) is a synthetic peptide created to model cis and torsionally strained peptide bonds that also exhibits a strong distinctive UV circular dichroic (CD) spectrum. Circular dichroic spectra were computed for the amide pi-pi* transition using the dipole interaction model for various conformations of the peptide. Conformations of CP3 were created initially from crystal data, and followed by energy minimizations via molecular mechanics using the cvff force field; the effects of additional geometric optimizations by semi-empirical and ab initio quantum mechanics were investigated. The CD spectra for each conformation were calculated using a variety of different parameters, and each result was compared with the published experimental spectrum [Deber, C.M., Scatturin, A., Vaidya, V.M. & Blout, E.R. (1970) Small cyclic proline peptides: UV absorption and CD. Herein, two distinct conformations, a C3 symmetric and an asymmetric form, gave CD predictions that separately did not resemble the experimental spectrum. Energy differences were predicted at various theoretical levels, including MP2 and density functional theory. When the predicted CD spectra for each conformation were multiplied by Boltzmann weighting factors created using heats of formation determined by the AM1 optimizations, the weighted composite CD spectrum created did resemble experiment for the pi-pi* transition indicating that both conformations may exist simultaneously in solution.  相似文献

Chikungunya virus (CHIKV) is primarily transmitted by Aedes spp. mosquitoes. The present study investigated vector competence for CHIKV in Aedes aegypti and Aedes albopictus mosquitoes found in Madurai, South India. The role of receptor proteins on midguts contributing to permissiveness of CHIKV to Aedes spp. mosquitoes was also undertaken. Mosquitoes were orally infected with CHIKV DRDE‐06. Infection of midguts and dissemination to heads was confirmed by immunofluorescence assay at different time points. A plaque assay was performed from mosquito homogenates at different time points to study CHIKV replication. Presence of putative CHIKV receptor proteins on mosquito midgut epithelial cells was detected by virus overlay protein binding assay (VOPBA). The identity of these proteins was established using mass spectrometry. CHIKV infection of Ae. aegypti and Ae. albopictus midguts and dissemination to heads was observed to be similar. A plaque assay performed with infected mosquito homogenates revealed that CHIKV replication dynamics was similar in Aedes sp. mosquitoes until 28 days post infection. VOPBA performed with mosquito midgut membrane proteins revealed that prohibitin could serve as a putative CHIKV receptor on Aedes mosquito midguts, whereas an absence of CHIKV binding protein/s on Culex quinquefasciatus midguts can partially explain the non‐permissiveness of these mosquitoes to infection.  相似文献

We report a simple route to design highly sensitive triangular silver nanoplates (TSNPs)-based SERS substrate for the trace-level detection of explosive molecules. The size-dependent localized surface plasmon resonance (LSPR) tunability for the synthesis of TSNPs is achieved by controlling reaction kinetics and seed volume in a modified seed-mediated approach. The computed extinction spectra of TSNP, using the finite-difference time-domain (FDTD) method, are in excellent agreement with the experimental results, therefore assisting further in the investigation of the plasmonic properties of TSNP. The higher electric field enhancement offered by TSNP is systematically investigated by performing the FDTD simulations for various sizes and corner rounding of TSNP. The FDTD results show that the dipolar plasmon resonance wavelength, size, and corner rounding of TSNP are the principal contributing factors for designing the high-performance SERS substrate. Herein, we have used a portable Raman system for the SERS-based detection of three important explosive molecules: picric acid (PA), ammonium nitrate (AN), and 2, 4-dinitrotoluene (DNT). The TSNP-based SERS substrates display excellent intensity enhancement factors of?~?107 for rhodamine 6G (R6G) and PA and?~?105 for AN. The high sensitivity of SERS substrate with limit-of-detection (LOD) of value 2.3?×?10?11 M for PA and 3.1?×?10?8 M for AN and effective batch-to-batch reproducibility for DNT, thus offering its potentials for field detection of explosive molecules at trace-level.
