Laccases (EC 1.10.3.2) are multicopper oxidases able to oxidize various substrates, such as phenolic subunits of lignin. The substrate range can be widened to non-phenolic units by the use of mediators. Since discovery of the laccase-mediator system, direct reactions of lignin and laccase without mediated electron-transfer have gained much less attention. The objective of this study was to investigate lignin as a substrate for fungal laccases by using lignin model compounds. These model compounds contained guaiacylic and syringylic moieties and also compounds of guaiacylic origin at a higher oxidation level. Some of these compounds are commercially available, but most of them were synthesized. The oxidation reaction rates of the lignin model compounds were studied by monitoring consumption of the co-substrate oxygen, in reactions catalyzed by laccases from two different fungi; Melanocarpus albomyces and Trametes hirsuta, possessing different molecular and catalytic properties. These reaction rate studies were compared to physicochemical properties of the lignin model compounds: relative redox potentials determined using cyclic voltammetry and pKa-values. Docking of syringylic and biphenylic compounds to the active sites of both laccases was performed and the resulting model complex structures were used to further interpret the reaction rate results. Reaction rates of laccases are mainly affected by the ability of a lignin model compound to be oxidized and the pKa-value of the substrate seems to be less important. As a consequence, syringylic compounds are oxidized with the highest rates and compounds at a higher oxidation level and redox-potential, such as vanillin, are oxidized at a much lower rate. Both guaiacylic and syringylic type compounds fit well in the active sites of both laccases. Only for a biphenylic compound, steric clashes were observed, and they are likely to have an effect on the reaction rate. When the oxidation rates on the selected model compounds with the two different laccases were compared, the redox-potential difference between laccases T1 copper and the lignin model compound (ΔE) was not the only property that determined the oxidation rate. Mast cells are a significant source of tumor necrosis factor (TNF) superfamily members, such as TNF-α, CD30 ligand/CD153 (CD30L)
and CD40L/CD154. Furthermore, the expression of some of these proteins in mast cells has been associated with tumorigenesis,
and mast cells have been found to be increased in number in the basal cell carcinoma (BCC) lesion. In this study, we have
examined the expression of TNF-α, CD30L and CD40L immunoreactivity in mast cells in the healthy-looking skin and lesional
skin of ten patients with superficial spreading BCC. Also, the counterparts of these molecules, TNF receptor (TNFR) I and
II as well as CD30 and CD40, were analysed immunohistochemically. We found that numbers of mast cells and Kit-positive cells
were significantly increased in the dermal BCC lesion. The percentage of CD30L-positive mast cells and the number of CD30-positive
cells were significantly increased in the upper dermis of the BCC lesion as well. In addition, the numbers of TNF-α-positive
mast cells and cells with TNFRI and TNFRII were markedly increased in the upper lesional dermis. In contrast, no mast cells
positive for CD40L could be detected, even though the lesional dermis contained increased numbers of CD40 positive cells.
The BCC epithelium was positive for TNFRI, TNFRII and CD40, but not for CD30, though the larger basal buds appeared to be
less intensely stained for TNFRI and CD40. In conclusion, mast cells positive for CD30L and TNF-α, but not CD40L, are increased
in number in the lesional dermis in BCC. These data suggest plausible pathways whereby mast cells can be activated and to

The aim of our work was to study the working‐age population's self‐reported physical symptoms associated with using mobile phones and other electrical devices. A qualitative method was applied using an open‐ended question in a questionnaire, which included questions about the possible influence of new technical equipment on health. We then created subgroups of respondents for different self‐reported symptoms associated with mobile phones and other electrical devices. The research questions were: (1) how the respondents described physical symptoms associated with using mobile phones and other electrical devices and (2) how the answers can be classified into subgroups based on symptoms or devices. We identified the following categories: (1) respondents with different self‐reported symptoms which they associated with using mobile phones (headache, earache, or warmth sensations), (2) respondents who had skin symptoms when they stayed in front of a computer screen, (3) respondents who mentioned physical symptoms associated with using mobile phones and other electrical devices. Total prevalence of self‐reported physical symptoms associated with using mobile phones and other electrical devices (categories 1 and 2) was 0.7%. Lactobacillus acidophilus NCFM is a probiotic bacterium adapted to survive in the gastrointestinal tract and with potential health benefits to the host. Lactitol is a synthetic sugar alcohol used as a sugar replacement in low calorie foods and selectively stimulating growth of L. acidophilus NCFM. In the present study the whole-cell extract proteome of L. acidophilus NCFM grown on glucose until late exponential phase was resolved by 2-DE (pH 3-7). A total of 275 unique proteins assigned to various physiological processes were identified from 650 spots. Differential 2-DE (DIGE) (pH 4-7) of L. acidophilus NCFM grown on glucose and lactitol, revealed 68 spots with modified relative intensity. Thirty-two unique proteins were identified in 41 of these spots changing 1.6-12.7-fold in relative abundance by adaptation of L. acidophilus NCFM to growth on lactitol. These proteins included β-galactosidase small subunit, galactokinase, galactose-1-phosphate uridylyltransferase and UDP-glucose-4-epimerase, which all are potentially involved in lactitol metabolism. This first comprehensive proteome analysis of L. acidophilus NCFM provides insights into protein abundance changes elicited by the prebiotic lactitol. 相似文献

Dispersal is an important mechanism used to avoid inbreeding. However, dispersal may only be effective for part of an individual's lifespan since, post-dispersal individuals that breed over multiple reproductive events may risk mating with kin of the philopatric sex as they age. We tested this hypothesis in black grouse Tetrao tetrix, and show that yearling females never mated with close relatives whereas older females did. However, matings were not with direct kin suggesting that short-distance dispersal to sites containing kin and subsequent overlap of reproductive lifespans between males and females were causing this pattern. Chick mass was lower when kinship was high, suggesting important fitness costs associated with inbred matings. This study shows that increased inbreeding risk might be a widespread yet rarely considered cost of ageing. 相似文献

The heritability estimate (±SE) for tarsus length in the pied flycatcher is 0.53 ± 0.10, based on mother-offspring regressions. The heritability is almost the same (0.50 ± 0.22) for offspring transferred to other nests and reared by foster parents, whereas there is no resemblance (0.04 ± 0.23) between the offspring and their foster mothers. Hence, the nest environment does not affect parent-offspring resemblance. However, a full-sib correlation yields an estimate of the heritability twice as high as the parent-offspring regression did, indicating that shared environment effects, which are not correlated with the tarsus length of parents, must be important. An environmental deviation due to food factors affecting tarsus length is demonstrated. The most important food factors are probably associated with 1) polygyny, which leads to reduced paternal feeding at secondary nests, 2) high breeding density, and 3) progress of the breeding season. All three are associated with reductions in offspring mean tarsus length. We estimate selection on tarsus length for the major components of lifetime reproductive success. Offspring with the shortest tarsi have reduced survival from fledging until breeding, and males with tarsus length close to the mean are most successful in attracting mates. Clutch size increases with female tarsus length, except for individuals with very long tarsi. In general, directional selection is weak, but stabilizing selection is rather strong for survival and male mating success. 相似文献

Good genes models of sexual selection assume that sexual advertisement is costly and thus the level of advertisement honestly reveals heritable viability. Recently it has been suggested that an important cost of sexual advertisement might be impairment of the functioning of the immune system. In this field experiment we investigated the possible trade-offs between immune function and sexual advertisement by manipulating both mating effort and activity of immune defence in male pied flycatchers. Mating effort was increased in a non-arbitrary manner by removing females from mated males during nest building. Widowed males sustained higher haematocrit levels than control males and showed higher expression of forehead patch height, suggesting that manipulation succeeded in increasing mating effort. Males that were experimentally forced to increase mating effort had reduced humoral immune responsiveness compared with control males. In addition, experimental activation of immune defence by vaccination with novel antigens reduced the expression of male ornament dimensions. To conclude, our results indicate that causality behind the trade-off between immune function and sexual advertisement may work in both directions: sexual activity suppresses immune function but immune challenge also reduces sexual advertisement. 相似文献

We compared four methods to enumerate three Bifidobacterium strains in fermented oat products during storage. Understanding the factors that underlie colonization success is crucial both for ecological theory and conservation practices. The most effective way to assess colonization ability is to introduce experimentally different sets of individuals in empty patches of suitable habitat and to monitor the outcome. We translocated mated female waterstriders, Aquarius najas, into 90 streams that were not currently inhabited by the species. We manipulated sizes of propagules (from 2 to 16 mated females) and numbers of origin populations (one or two). Three origin populations were genetically different from each other, but they were less than 150 km from the streams of translocation. The results demonstrate clearly that both the larger propagule size and the high number of source populations have positive effects on the probability of colonizing a new stream.
