A cDNA clone, pHGR81, encoding 358 amino-acid residues of the C-terminal region of human elongation factor 2 (EF-2), was isolated from a human ovarian granulosa cell cDNA library. The deduced amino-acid sequence of pHGR81, when compared with the known identical amino-acid sequences of hamster as well as rat EF-2 revealed a substitution of a glutamine by an alanine residue in the partially determined human sequence. The 15 amino-acid-residue sequence comprising the histidine-715, supposed to be of importance for the biological function of EF-2, is preserved in human EF-2. The coding region of the cDNA insert of pHGR81 displays a homology of 87% to hamster and of 88% to rat EF-2 cDNA. In Northern-transfer analysis, pHGR81 specifically hybridizes with an mRNA species of 3.1 kb.  相似文献

The effect of clearcutting on biomass production and nutrient consumption was examined during the two years in a clearcut and a mature Quercus ilex L. coppice. The comparison was limited to the leaf compartment and to the annual shoots, which were the only comparable compartments in the two situations. At the same time, the dynamics of the nutrient pool (N, P, K, Ca, Mg), monthly uptake and recycling from the foliage, were studied.The second year after cutting, the leaf production was roughly the same at the two sites. However, the photosynthates appear to be used differently: in the young stand the perennial wood production was greater than in the mature stand, where the maintenance costs are higher. This is corroborated by the patterns of nutrient use.  相似文献

In resting cells, c-Jun is phosphorylated on five sites. Three of these sites reside next to its DNA binding domain and negatively regulate DNA binding. In response to expression of oncogenic Ha-Ras, phosphorylation of these sites decreases, while phosphorylation of two other sites within c-Jun's activation domain is greatly enhanced. Phosphorylation of these residues, serines 63 and 73, stimulates the transactivation function of c-Jun and is required for oncogenic cooperation with Ha-Ras. We now show that the same changes in c-Jun phosphorylation are elicited by a variety of transforming oncoproteins with distinct biochemical activities. These oncoproteins, v-Sis, v-Src, Ha-Ras, and Raf-1, participate in a signal transduction pathway that leads to increased phosphorylation of serines 63 and 73 on c-Jun. While oncogenic Ha-Ras is a constitutive stimulator of c-Jun activity and phosphorylation, the normal c-Ha-Ras protein is a serum-dependent modulator of c-Jun's activity. c-Jun is therefore a downstream target for a phosphorylation cascade involved in cell proliferation and transformation.  相似文献

The rapids-dwelling suckermouth catfish genus Pseudolithoxus was previously only known from the Guiana-Shield-draining Orinoco and Casiquiare river systems of Colombia and Venezuela, but new records have expanded this range considerably further into the Amazon basin of Brazil, and include occurrences from rivers draining the northern Brazilian Shield. These highly disjunct records are now placed in an evolutionary and phylogeographic context using a dated species tree constructed from mitochondrial (Cytb) and nuclear (RAG1) gene sequence data. Due to mito-nuclear discordance, we also delimit the putative species using statistical coalescent models and a range of additional metrics. We infer that at least two species of Pseudolithoxus are present in the Amazon basin: P. nicoi, previously only recorded from the río Casiquiare, but now also reported from the upper rio Negro, and a new species, which we describe herein from south-draining Guiana Shield and north-draining Brazilian Shield. Our data reject a simple model of Miocene vicariance in the group following uplift of the Uaupés Arch separating the Orinoco and Amazon systems, and instead suggest more complex dispersal scenarios through palaeo-connections in the Pliocene and also via the contemporary rio Negro and rio Madeira in the late Pleistocene.

A re‐evaluation of Dekeyseria brachyura and D. pulchra found them to be junior synonyms of D. picta, the oldest species among these congeners, based on continuous intraspecific variation in morphometrics and colour pattern. Examination of material deposited at Brazilian and Venezuelan collections, including the original type specimens of Ancistrus brachyurus and A. pictus, plus samples recently collected in the Rio Negro, Amazonas, Brazil, allowed re‐evaluation of the taxonomic status of D. picta and provides additional information on its distribution, habitats and reproductive strategies.  相似文献

We used x-ray and neutron diffraction to study the temperature- and pressure-dependent structure and phase behavior of the monoacylglycerides 1-monoelaidin (ME) and 1-monoolein (MO) in excess water. The monoacylglycerides were chosen for investigation of their phase behavior because they exhibit mesomorphic phases with one-, two-, and three-dimensional periodicity, such as lamellar, an inverted hexagonal and bicontinuous cubic phases, in a rather easily accessible temperature and pressure range. We studied the structure, stability, and transformations of the different phases over a wide temperature and pressure range, explored the epitaxial relations that exist between different phases, and established a relationship between the chemical structure of the lipid molecules and their phase behavior. For both systems, a temperature-pressure phase diagram has been determined in the temperature range from 0 to 100 degrees C at pressures from ambient up to 1400 bar, and drastic differences in phase behavior are found for the two systems. In MO-water dispersions, the cubic phase Pn3m extends over a large phase field in the T,p-plane. At temperatures above 95 degrees C, the inverted hexagonal phase is found. In the lower temperature region, a crystalline lamellar phase is induced at higher pressures. The phases found in ME-water include the lamellar crystalline Lc phase, the L beta gel phase, the L alpha liquid-crystalline phase, and two cubic phases belonging to the crystallographic space groups Im3m and Pn3m. In addition, the existence of metastable phases has been exploited. Between coexisting metastable cubic structures, a metric relationship has been found that is predicted theoretically on the basis of the curvature elastic energy approximation only.  相似文献

DNA sequence comparisons of two mitochondrial DNA genes were used to infer phylogenetic relationships among 17 Felidae species, notably 15 in the previously described pantherine lineage. The polymerase chain reaction (PCR) was used to generate sequences of 358 base pairs of the mitochondrial 12S RNA gene and 289 base pairs of the cytochrome b protein coding gene. DNA sequences were compared within and between 17 felid and five nonfelid carnivore species. Evolutionary trees were constructed using phenetic, cladistic, and maximum likelihood algorithms. The combined results suggested several phylogenetic relationships including (1) the recognition of a recently evolved monophyletic genus Panthera consisting of Panthera leo, P. pardus, P. onca, P. uncia, P. tigris, and Neofelis nebulosa; (2) the recent common ancestry of Acinonyx jubatus, the African cheetah, and Puma concolor, the American puma; and (3) two golden cat species, Profelis temmincki and Profelis aurata, are not sister species, and the latter is strongly associated with Caracal caracal. These data add to the growing database of vertebrate mtDNA sequences and, given the relatively recent divergence among the felids represented here (1-10 Myr), allow 12S and cytochrome b sequence evolution to be addressed over a time scale different from those addressed in most work on vertebrate mtDNA.   相似文献

Abstract. Seasonal variation in nutrient concentration in leaves and branches of Quercus pyrenaica was studied in natural Q. pyrenaica forest in the Sierra de Gata (Salamanca Province, Spain). Two permanent plots were established at the two extremes of a rainfall gradient in this area: annual mean precipitation from 720 mm at Fuenteguinaldo (granite bedrock) to 1580 mm at Navasfrias (schists and graywackes). Leaf and branch samples were collected every three weeks during the growing season from May to October, at three height levels of the tree canopy. Seasonal changes and internal nutrient dynamics were investigated for N, Ca, Mg, K, Na, Mn, Fe and P during a two-year period. The concentrations of all nutrients varied among the seasons; these variations were related to nutrient mobility and the annual physiological cycle. Nutrient concentrations decrease in the case of K and P, while the sparse mobile nutrients Ca, Mg, Mn and Fe gradually accumulated during each growing season. In Navasfrias a considerable resorption of P from senescing leaves was detected. Different patterns were found for the other nutrients studied (Na and N).  相似文献
