The forelimb digital flexors of the horse display remarkable diversity in muscle architecture despite each muscle-tendon unit having a similar mechanical advantage across the fetlock joint. We focus on two distinct muscles of the digital flexor system: short compartment deep digital flexor (DDF(sc)) and the superficial digital flexor (SDF). The objectives were to investigate force-length behavior and work performance of these two muscles in vivo during locomotion, and to determine how muscle architecture contributes to in vivo function in this system. We directly recorded muscle force (via tendon strain gauges) and muscle fascicle length (via sonomicrometry crystals) as horses walked (1.7 m s(-1)), trotted (4.1 m s(-1)) and cantered (7.0 m s(-1)) on a motorized treadmill. Over the range of gaits and speeds, DDF(sc) fascicles shortened while producing relatively low force, generating modest positive net work. In contrast, SDF fascicles initially shortened, then lengthened while producing high force, resulting in substantial negative net work. These findings suggest the long fibered, unipennate DDF(sc) supplements mechanical work during running, whereas the short fibered, multipennate SDF is specialized for economical high force and enhanced elastic energy storage. Apparent in vivo functions match well with the distinct architectural features of each muscle.  相似文献

Background

The two sympatric species of Tunisian desert ants, Cataglyphis bicolor and C. mauritanica, do not exhibit any differences in their foraging ecology, e.g. in food preferences and in their spatial and temporal activity patterns. Here we show that instead the two species markedly differ in their life histories.

Results

We analysed mtDNA of specimens that were collected along a 250-km transect. C. bicolor exhibited a genetically unstructured population (with the genetic and geographic distances among colonies not being correlated). On the contrary the populations of the polygynous C. mauritanica were clearly structured, i.e. exhibited a strong correlation between genetic and geographic distances. This difference is in accordance with large queen dispersal distances due to far-reaching mating flights in C. bicolor and small queen dispersal distances due to colony foundation by budding in C. mauritanica. Furthermore, wherever we found populations of both species to coexist within the same habitat, the habitat was used agriculturally. Mapping nest positions over periods of several years showed that plowing dramatically decreased the nest densities of either species.

Conclusion

We conclude that owing to its greater queen dispersal potential C. bicolor might be more successful in quickly re-colonizing disturbed areas, while the slowly dispersing C. mauritanica could later out-compete C. bicolor by adopting its effective nest-budding strategy. According to this scenario the observed sympatry of the two species might be an intermediate stage in which faster colonization by one species and more powerful exploitation of space by the other species have somehow balanced each other out. In conclusion, C. bicolor and C. mauritanica represent an example where environmental disturbances in combination with different life histories might beget sympatry in congeneric species with overlapping niches.  相似文献

Background

Human solid tumors that are hard or firm on physical palpation are likely to be cancerous, a clinical maxim that has been successfully applied to cancer screening programs, such as breast self-examination. However, the biological relevance or prognostic significance of tumor hardness remains poorly understood. Here we present a fracture mechanics based in vivo approach for characterizing the fracture toughness of biological tissue of human thyroid gland tumors.

Methods

In a prospective study, 609 solid thyroid gland tumors were percutaneously probed using standard 25 gauge fine needles, their tissue toughness ranked on the basis of the nature and strength of the haptic force feedback cues, and subjected to standard fine needle biopsy. The tumors' toughness rankings and final cytological diagnoses were combined and analyzed. The interpreting cytopathologist was blinded to the tumors' toughness rankings.

Results

Our data showed that cancerous and noncancerous tumors displayed remarkable haptically distinguishable differences in their material toughness.

Conclusion

The qualitative method described here, though subject to some operator bias, identifies a previously unreported in vivo approach to classify fracture toughness of a solid tumor that can be correlated with malignancy, and paves the way for the development of a mechanical device that can accurately quantify the tissue toughness of a human tumor.  相似文献

A phylogeny of the arthropods was inferred from analyses of amino acid sequences derived from the nuclear genes encoding elongation factor-1 alpha and the largest subunit of RNA polymerase II using maximum- parsimony, neighbor-joining, and maximum-likelihood methods. Analyses of elongation factor-1 alpha from 17 arthropods and 4 outgroup taxa recovered many arthropod clades supported by previous morphological studies, including Diplopoda, Myriapoda, Insecta, Hexapoda, Branchiopoda (Crustacea), Araneae, Tetrapulmonata, Arachnida, Chelicerata, and Malacostraca (Crustacea). However, counter to previous studies, elongation factor-1 alpha placed Malacostraca as sister group to the other arthropods. Branchiopod crustaceans were found to be more closely related to hexapods and myriapods than to malacostracan crustaceans. Sequences for RNA polymerase II were obtained from 11 arthropod taxa and were analyzed separately and in combination with elongation factor-1 alpha. Results from these analyses were concordant with those derived from elongation factor-1 alpha alone and provided support for a Hexapoda/Branchiopoda clade, thus arguing against the monophyly of the traditionally defined Atelocerata (Hexapoda + Myriapoda).   相似文献

In O2 equilibrium studies of hemoglobin it was observed that the presence of urea increases f502 and decreases co-operativity. The results were interpreted on the basis of unfolding and swelling of the peptide chains. The results of the present kinetic investigation indicate that while the CO combination rate constants for αSH chains, hemoglobin H and myoglobin do not change with increasing concentrations of urea, the corresponding rate constants for deoxy hemoglobin A increase continuously with urea concentration. At urea concentrations of 4 m or more, the reaction time course becomes biphasic. The fast component of the reaction time course yields CO combination rate constants which are in close agreement with the rate constants of dimeric and monomeric hemoglobins. These results indicate that up to 4 m-urea the kinetic and equilibria parameters increase due to weakened constraints imposed by intersubunit contacts and bonds. At higher urea concentrations Hb4 is significantly dissociated into dimers and monomers, and hence the high ligand affinity and decreased co-operativity of the system. The implications of higher ligand combination rate constants of the deoxyhemoglobin tetramer in the presence of urea on the reaction mechanism are discussed.  相似文献

A new expression for ion leakage from plant tissue, the tissue ionic conductance (gTi), is compared with electrical conductivity (EC) and a commonly used damage index (Id) to test the ability of each expression to correctly describe leakiness in two model systems representing examples of physiological processes with well-known effects on membrane permeability. In experiments in which drought-acclimated leaves were compared with nonacclimated leaves and senescing leaves were compared with nonsenescing leaves, Id contradicted our expectation that acclimated tissue would be less leaky than nonacclimated tissue, and gTi and EC confirmed this expectation. In a comparison of senescing and nonsenescing tissue, Id again contradicted our expectation that senescing tissue would be more leaky than nonsenescing, and EC and gTi were confirming. Using a diffusion analysis approach, we show that Id fails to account for variation in the concentration gradient between the tissue and the bathing solution and variation in the surface area through which efflux occurs. Furthermore, because Id is a parameter that relates treatment performance to control performance as a percentage value, it distorts the actual differences among treatments. The resulting artifacts lead to a presentation of membrane integrity which is probably incorrect. EC is a more direct measurement of net ion efflux and appears to be less vulnerable to artifact. However, because gTi is the only expression that explicitly includes chemical driving force and tissue surface area, it is the most reliable of the three expressions.  相似文献

Exposure of the outside of the isolated frog skin to a Ringer's solution, made hypertonic by the addition of mannitol, causes a rapid and sustained increase in transepithelial permeability through a structural distortion-a focal blistering-of the "tight" junctions of the outermost living cell layer. [(3)H]ouabain, used as an autoradiographic marker for the Na+-pump (Na+-K+-adenosine triphosphatase), is usually unable to penetrate the frog skin from the outside solution, but when added to a hypertonic mannitol- Ringer's solution in the outside bath it readily penetrates the epithelium, presumably through the opened shunt pathway. Radioautographic analysis of [(3)H]ouabain binding sites revealed that most of ouabain enters from the outside solution binds to the sites on the cell membranes of the stratum spinosum, as was the case when it was applied from the inside bath in an earlier study. The outer living cell layer, the first to be exposed to ouabain, does not appear to be the major site for the Na+-pump, and therefore, is not likely to be responsible for most of the active pumping of Na+. This result demonstrates that previous failure to show a high density of Na+-pump sites on the cells of the outermost layer, when [(3)H]ouabain was applied from the inside solution, was not due to the inability of the marker to reach these cells at a sufficient concentration to reveal all pump sites. These results provide further support for a model of Na+-transport across the frog skin which distributes the active pump step on the inward facing membranes of all living cells.  相似文献

Molecular adaptation of a leaf-eating bird: stomach lysozyme of the hoatzin   总被引:6,自引:1,他引:5  
This report describes a lysozyme expressed at high levels in the stomach of the hoatzin, the only known foregut-fermenting bird. Evolutionary comparison places it among the calcium-binding lysozymes rather than among the conventional types. Conventional lysozymes were recruited as digestive enzymes twice in the evolution of mammalian foregut fermenters, and these independently recruited lysozymes share convergent structural changes attributed to selective pressures in the stomach. Biochemical convergence and parallel amino acid replacements are observed in the hoatzin stomach lysozyme even though it has a different genetic origin from the mammalian examples and has undergone more than 300 million years of independent evolution.   相似文献

This study investigated the effects of a 16-h protocol of heavy intermittent exercise on the intrinsic activity and protein and isoform content of skeletal muscle Na(+)-K(+)-ATPase. The protocol consisted of 6 min of exercise performed once per hour at approximately 91% peak aerobic power (Vo(2 peak)) with tissue sampling from vastus lateralis before (B) and immediately after repetitions 1 (R1), 2 (R2), 9 (R9), and 16 (R16). Eleven untrained volunteers with a Vo(2 peak) of 44.3 +/- 2.3 ml x kg(-1) x min(-1) participated in the study. Maximal Na(+)-K(+)-ATPase activity (V(max), in nmol x mg protein(-1) x h(-1)) as measured by the 3-O-methylfluorescein K(+)-stimulated phosphatase assay was reduced (P < 0.05) by approximately 15% with exercise regardless of the number of repetitions performed. In addition, V(max) at R9 and R16 was lower (P < 0.05) than at R1 and R2. Vanadate-facilitated [(3)H]ouabain determination of Na(+)-K(+)-ATPase content (maximum binding capacity, pmol/g wet wt), although unaltered by exercise, increased (P < 0.05) 8.3% by R9 with no further increase observed at R16. Assessment of relative changes in isoform abundance measured at B as determined by quantitative immunoblotting showed a 26% increase (P < 0.05) in the alpha(2)-isoform by R2 and a 29% increase in alpha(3) by R9. At R16, beta(3) was lower (P < 0.05) than at R2 and R9. No changes were observed in alpha(1), beta(1), or beta(2). It is concluded that repeated sessions of heavy exercise, although resulting in increases in the alpha(2)- and alpha(3)-isoforms and decreases in beta(3)-isoform, also result in depression in maximal catalytic activity.  相似文献
