The acute nephrotoxicity caused by N-(3,5-dichlorophenyl) succinimide (NDPS) has been shown to be due to a metabolite(s) of the parent compound. This study examined the toxicity of NDPS, its known metabolites N-(3,5-dichlorophenyl)-2-hydroxysuccinimide (NDHS), N-(3,5-dichlorophenyl)-2-hydroxysuccinamic acid (NDHSA), N-(3,5-dichlorophenyl)malonamic acid (DMA), N-(3,5-dichlorophenyl)succinamic acid (NDPSA), and two postulated metabolites N-(3,5-dichlorophenyl)maleamic acid (NDPMA) and N-(3,5-dichlorophenyl)maleimide (NDPM) to suspensions of renal proximal tubules (RPT) prepared from male Fischer 344 rats. Tubule viability and mitochondrial function were not adversely affected by exposure of RPT to either 1 mM NDPS, NDHS, NDHSA, DMA, NDPSA, or NDPMA for 4 h. However, NDPM caused a concentration-(25-100 microM) and time-dependent (0.25-4 h) loss in basal and nystatin stimulated oxygen consumption and tubule viability. Investigations using isolated renal cortical mitochondria (RCM) showed that NDPM was a potent inhibitor of mitochondrial function. Isolated RCM respiring on pyruvate/malate and exposed to NDPM exhibited a concentration (25-100 microM) dependent decrease in state 3 and state 4 respiration. Inhibition of mitochondrial state 3 respiration by NDPM was mediated through site 1 of the respiratory chain. NDPM did not inhibit cytochrome c-cytochrome oxidase or the electron transport chain. These results indicated that NDPS, its known metabolites, and NDPMA were not directly toxic to rat RPT. However, the postulated metabolite NDPM, was a potent tubule cytotoxicant that inhibited mitochondrial function in isolated RCM and RPT and may produce cell death through this mechanism.  相似文献

Trap-constructing organisms provide a unique opportunity for the study of resource allocation, because an observer can unambiguously determine the allocation to foraging. In species that synthesize a trap from physiologically important compounds, there is the further advantage that there may be direct trade-offs between allocation of resources to foraging and physiological functions. We examined the ability of the spider Nephila clavipes (L.; Araneae: Tetragnathidae) to synthesize resources that are known to be used for both web synthesis and non-foraging physiological functions. We found that choline, required for both web function and physiological function, is an essential nutrient: it is not synthesized by this spider. Under laboratory conditions with a diet of fruit flies, choline is limiting, and the spiders make allocation trade-offs between investing choline in foraging (the web) or in their body.  相似文献

Mouse epididymal protein (MEP) 10 has recently been characterized in our laboratory. Amino acid sequence analysis of the N-terminal of MEP 10 revealed an 86% similarity in sequence with rat proteins B and C, characterized by Brooks and Higgins [J Reprod Fertil 1980; 59:363-375]. Proteins B and C, have been recently shown to possess retinoic acid-binding ability [Newcomer ME, Ong DE. J Biol Chem 1990; 265:12876-12879; Ong DE, Chytil F. Arch Biochem Biophys 1988; 267:474-478]. Therefore, it was of interest to determine whether MEP 10 possessed the same ability to bind retinoic acid. Mouse caudal fluid was trace-labeled with 3H-retinoic acid and applied to a DEAE ion-exchange column. Analysis of the fractions for both the presence of radioactivity by scintillation counting and MEP 10 by ELISA revealed that the peak of radioactivity corresponded to the peak of MEP 10 immunoreactivity. These peak fractions were pooled and used for subsequent binding analysis and SDS-PAGE and Western blot analysis. SDS-PAGE and Western blot analysis revealed that the peak fractions were enriched for a protein of 18 kDa and that this protein was MEP 10. Competitive binding assays revealed that all-trans-retinoic acid was effective in inhibiting binding of labeled retinoic acid, but that the 13-cis isomer of retinoic acid was only moderately effective in inhibiting binding of the labeled ligand. All-trans-retinol was ineffective in the binding inhibition assay. Activation of human PBL T cells with solid phase anti-CD3 mAb or during the course of an MLR response gives rise to the association of CD4 or CD8 molecules with the protein tyrosine phosphatase, CD45, on the cell surface. This paired association of cell-surface molecules occurs late in the activation cycle and appears to be dependent upon Ti-CD3-mediated signaling because mitogen-driven activation does not induce formation of the complex. Maximal association occurred 72 to 96 h after exposure to anti-CD3 mAb on both CD4+ and CD8+ T cells. In contrast, association between CD8 and CD45 during an MLR response did not occur until day 6 of a MLR whereas CD4-CD45 association was detected by 72 h of culture. The kinetics of association between CD4 or CD8 and CD45 was measured by fluorescence resonance energy transfer and confirmed by immunoprecipitation of dithiobis succinimidylpropionate or disuccinimidyl suberate cross-linked 125I-labeled resting or activated T cells. The molecules that co-precipitated with either CD4 or CD8 and had an apparent kDa of 180 to 205 could be immunodepleted with anti-CD45 mAb. Furthermore, CD4 or CD8 immunoprecipitates from 96-h activated T cells contained significant levels of protein tyrosine phosphatase activity whereas corresponding immunoprecipitates from resting or recently activated T cells showed little protein tyrosine phosphatase activity. This association may allow CD45 to engage and dephosphorylate lck or another CD4- or CD8-associated substrate in order to reset the receptor complex to receive a new set of stimuli. Our observations suggest that synergistic signaling provided as a consequence of CD4 or CD8 association with the TCR after antigenic stimulation may develop on a different temporal scale than that observed after soluble anti-CD4+ anti-CD3 heteroconjugate antibody cross-linking.  相似文献

Ever since J.B.S. Haldane proposed the idea, evolutionary biologists are aware that individual level adaptations do not necessarily lead to optimal population performance. A few deeply mathematical models, drawing from a diverse range of systems, even predict that individual selection can lead to the extinction of the whole population, a phenomenon which has become known as evolutionary suicide. Due to the complexity of both following adaptation and determining the exact cause of an extinction, evolutionary suicide has remained untested empirically. However, three recent empirical studies suggest that it may occur, and that suicide should be taken seriously as a potentially important evolutionary phenomenon. Here we ask whether or not evolutionary suicide can occur, briefly reviewing the theoretical and empirical evidence. We further highlight systems which may be used to test whether or not individual level selection can cause extinction.  相似文献
