The aim of this study is focused on determining the Bacillus amyloliquefaciens QV15 priming fingerprint in two different plant species, Arabidopsis and blackberry as a crop of agronomic interest, associated with protection upon pathogen challenge. To achieve this goal, Arabidopsis thaliana plants were challenged with Pseudomonas syringae DC3000 under controlled conditions, and field-grown blackberries were challenged by a powdery Mildew outbreak, finding plant protection in plants treated with QV15, in both conditions. Changes in ROS scavenging enzymes’ activity, defense-related enzymes’ activity and gene expression were evaluated in both plant species, before and after pathogen challenge, revealing the ability of this strain to prime both. As a result of this analysis, the priming fingerprint induced by QV15 was defined by a decrease in ROS scavenging enzymes’ activity in pre- and post-challenged plants, an increase in glucanase and chitinase activity after pathogen challenge, significantly increasing the expression of PR1, indicating a salicylic acid (SA)-mediated pathway activation. These results suggest an excellent potential of B. amyloliquefaciens QV15 to protect different plant species against different pathogens in field conditions.  相似文献

In a study of the effect of short-term storage on the hatching rate of common carp Cyprinus carpio and tench Tinca tinca ova in vitro in various extenders at 21° C under aerobic conditions, the best extender for 30 min storage for common carp appeared to be Dettlaff 1. This gave the same hatching rate as controls without extender (55% v. 56%). For 60 min storage of ova, the best extenders were Dettlaff 2 (24% hatching rate) and Dettlaff 3 (30%), but hatching was significantly lower than in the control (58%). In carp ovarian artificial fluid (CAF) extender, the hatching rate of common carp ova was also high after 10 min, but decreased to 12% after 30 min. In tench, the hatching rate of ova increased after 10 min storage in Dettlaff 5 extender (44%) compared to the control (41%) without extender. However, it was significantly lower after storage in Dettlaff 1, 2, 3, 4, 5 and CAF extenders for 20, 30 and 60 min, compared to controls. Malformations (10–50%) were observed in the tench second control groups without extender after 10, 20 and 30 min storage of ova.  相似文献

Plant organogenesis is essentially a post-embryonic process that requires a strict balance between cell proliferation and differentiation. This is subject to a complex regulatory network which, in some cases, depends on the action of a variety of plant hormones. Of these, auxins and cytokinins are those best documented as impinging directly on cell cycle control. However, increasing evidence is accumulating to indicate that other hormones also have an impact on cell cycle control by influencing the availability of cell cycle regulators. In this article, we review the results that point to the variety of situations in which cell cycle progression is controlled by phytohormones.  相似文献

The chromosome complements of 45 species of spider mites (Tetranychidae) were studied, making the total number of species now examined in this family 57, approximately 10% of all species known. The chromosome numbers range fromn=2 ton=7. The modal number of the family is 3 (found in 44% of the species). It is argued that the ancestral number isn=2 (21% of the species).In the more primitive subfamily of theBryobiinae both thelytokous and arrhenotokous species occur, whereas the subfamily of theTetranychinae exclusively exhibits arrhenotoky. The karyotype evolution is discussed in connection with arrhenotoky. A combination of mutagenesis, calorimetry, kinetics, and small-angle X-ray scattering (SAXS) has been used to study the mechanism of ligand binding energy propagation through human cytochrome P450 reductase (CPR). Remarkably, the energetics of 2',5'-ADP binding to R597 at the FAD-binding domain are affected by mutations taking place at an interdomain loop located 60 A away. Either deletion of a 7 amino acid long segment (T236-G237-E238-E239-S240-S241-I242) or its replacement by poly-proline repeats (5 and 10 residues) results in a significant increase in 2',5'-ADP enthalpy of binding (DeltaHB). This is accompanied by a decrease in the number of thermodynamic microstates available for the ligand-CPR complex. Moreover, the estimated heat capacity change (DeltaCp) for this interaction changes from -220 cal mol-1 K-1 in the wild-type enzyme to -580 cal mol-1 K-1 in the deletion mutant. Pre-steady-state kinetics measurements reveal a 50-fold decrease in the microscopic rate for interdomain (FAD --> FMN) electron transfer in the deletion mutant (kobs = 0.4 s-1). Multiple turnover cytochome c reduction assays indicate that these mutations impair the ability of the FMN-binding domain to shuttle electrons from the FAD-binding domain to the cytochrome partner. Binding of 2',5'-ADP to wild-type CPR triggers a large-scale structural rearrangement resulting in the complex having a more compact domain organization, and the maximum molecular dimension (Dmax) decreases from 110 A in ligand-free enzyme to 100 A in the ligand-bound CPR. The SAXS experiments also demonstrate that what is affected by the mutations is indeed the relative diffusional motion of the domains. Furthemore, ab initio shape reconstruction and homology modeling would suggest that-in the deletion mutant-hindering of domain motion occurs concomitantly with dimerization. The results presented here show that the energetics of this highly localized interaction (2',5'-ADP binding) have a global character, and are highly sensitive to functional structural dynamics involving distal domains. These findings support early theoretical studies which postulate a single protein molecule to be a real, independent thermodynamic ensemble.  相似文献

Guava (Psidium guajava) is an aggressive invasive plant in the Galapagos Islands. Determining its provenance and genetic diversity could explain its adaptability and spread, and how this relates to past human activities. With this purpose, we analyzed 11 SSR markers in guava individuals from Isabela, Santa Cruz, San Cristobal, and Floreana islands in the Galapagos, as well as from mainland Ecuador. The mainland guava population appeared genetically differentiated from the Galapagos populations, with higher genetic diversity levels found in the former. We consistently found that the Central Highlands region of mainland Ecuador is one of the most likely origins of the Galapagos populations. Moreover, the guavas from Isabela and Floreana show a potential genetic input from southern mainland Ecuador, while the population from San Cristobal would be linked to the coastal mainland regions. Interestingly, the proposed origins for the Galapagos guava coincide with the first human settlings of the archipelago. Through approximate Bayesian computation, we propose a model where San Cristobal was the first island to be colonized by guava from the mainland, and then, it would have spread to Floreana and finally to Santa Cruz; Isabela would have been seeded from Floreana. An independent trajectory could also have contributed to the invasion of Floreana and Isabela. The pathway shown in our model agrees with the human colonization history of the different islands in the Galapagos. Our model, in conjunction with the clustering patterns of the individuals (based on genetic distances), suggests that guava introduction history in the Galapagos archipelago was driven by either a single event or a series of introduction events in rapid succession. We thus show that genetic analyses supported by historical sources can be used to track the arrival and spread of invasive species in novel habitats and the potential role of human activities in such processes.  相似文献

In animal communication, signals are expected to evolve to be honest, so that receivers avoid being manipulated by signalers. One way that signals can evolve to be honest is for them to be costly, with only high‐quality individuals being able to bear the costs of signal expression. It has been proposed that parasites can introduce costs that affect the expression of sexually selected traits, and there is evidence to support the role of parasitism in modulating animal behavior. If host infection status or intensity is found to relate to differences in signal expression, it may indicate a fitness cost that mediates honesty of signals. Birdsong is a good model for testing this, and physically challenging songs representing complex motor patterns provide a good example of sexually selected traits indicating individual condition. We performed a field study to evaluate the relationship between song performance and avian malaria infection in a common songbird. Previous work on this subject has almost always evaluated avian malaria in terms of binary infection status; however, parasitemia—infection intensity—is rarely assessed, even though differences in parasite load may have profound physiological consequences. We estimated parasitemia levels by using real‐time PCR. We found that birds with higher parasitemia displayed lower vocal performance, providing evidence that this song trait is an honest signal of parasitic load of haemosporidian parasites. To our knowledge, this study links parasite load and the expression of a sexually selected trait in a way that has not been addressed in the past. Studies using song performance traits and parasitemia offer an important perspective for understanding evolution of characters via sexual selection.  相似文献
