Human lymphatic filariasis (HLF) is a neglected tropical disease which threatens nearly 1.4 billion people in 73 countries worldwide. Wuchereria bancrofti is the major causative agent of HLF and it closely resembles cattle filarial parasite Setaria digitata. Due to difficulties in procuring W. bancrofti parasite material, S. digitata cDNA library has been constructed to identify novel drug targets against HLF and many of the cDNA sequences are yet to be assigned structure and function. In this study, a 549 bp long cDNA (sdrbp) has been sequenced and characterized in silico. The shortest ORF of 249 bp from the isolated cDNA encodes a polypeptide of 82 amino acids and shows an amino acid identity of 54% with the RRM domain of human cleavage stimulation factor-64 kDa subunit (CstF-64). Structure of the protein (sdRBP) obtained by homology modelling using RRM of CstF-64 as template adopts classical RRM topology (β1α1β2β3α2β4). sdRBP model built was validated by superimposition tools and Ramachandran plot analysis. CstF-64 plays an important role in pre-mRNA polyadenylation by interacting with specific GU-rich downstream sequence element. Molecular docking studies of sdRBP with different RNA molecules revealed that sdRBP has greater binding affinity to GU-rich RNA and comparable results were obtained upon similar docking of RRM of CstF-64 with the same RNA molecules. Therefore, sdRBP is likely to perform homologous function in S. digitata. This study brings new dimensions to the functional analysis of RNA binding proteins of S. digitata and their evaluation as new drug targets against HLF.  相似文献

Abstract: Meiotic products in higher plants should undergo a determined number of mitotic cycles before differentiating gametes. This creates a unique meiosis-mitosis interface, traverse of which is an absolute requirement for gametophyte development. In the absence of cytokinesis during megasporogenesis - as seen in the bisporic and tetrasporic types - the haploid nuclei produced by meiosis are driven to undergo mitotic cycles within the same cell. Similarly, the last of the mitotic cycles leads to a unique type of cell wall formation resulting in cellularization of the coenocytic female gametophyte, creating a mitosis-cellularization interface. Cell cycle regulation in terms of the molecules that interface with these two key spatio-temporal developmental settings should be of interest to both cell and developmental biologists. High throughput techniques of functional genomics are required for both interpretation of female gametophyte evolution and success of the biotechnological initiatives of transferring apomixis-related genes to crop plants.  相似文献

Seven species of the montium subgroup of Drosophila found in South India were analysed with polyacrylamide disc electrophoresis. Eleven loci coding for enzymes were surveyed. The genetic distances between species have been measured. The dendrogram revealed three clusters. The first cluster includes D. nagarholensis, D. agumbensis, D. jambulina and D. kikkawai; the second group consists of D. truncata and D. anomelani; while D. mysorensis was the only species of the third group.We dedicate this paper to our Professor N. Sequencing genomes of different pathogenic fungi produced plethora of genetic information. This "omics" data might be of great interest to probe strain diversity, identify virulence factors and complementary genes in other fungal species, and importantly in predicting the role of proteins specific to pathogenesis in humans. We propose a component called "fungome" for those fungal proteins implicated in pathogenesis which, we believe, will allow researchers to improve the annotation of fungal proteins.  相似文献

The DNA from the two Drosophila nasuta races, D. n. nasuta and D. n. albomicana was investigated by CsCl density gradient centrifugation. D. n. nasuta has one major AT-rich satellite DNA sequence with a density of 1.664g/cm3, while D. n. albomicana has at least three satellites with densities of 1.674g/cm3, 1.665g/cm3 and 1.661 g/cm3. The isolated satellite sequences hybridize in situ to all heterochromatic regions of all metaphase chromosomes of both races. In polytene chromosomes the satellite sequences hybridize exclusively to the chromocenter. All chromosomal regions hybridizing with the satellites show also bright quinacrine fluorescence.  相似文献
