Multiple Mitochondrial Dysfunctions Syndrome 1 (MMDS1) is a rare, autosomal recessive disorder caused by mutations in the NFU1 gene. NFU1 is responsible for delivery of iron-sulfur clusters (ISCs) to recipient proteins which require these metallic cofactors for their function. Pathogenic variants of NFU1 lead to dysfunction of its target proteins within mitochondria. To date, 20 NFU1 variants have been reported and the unique contributions of each variant to MMDS1 pathogenesis is unknown. Given that over half of MMDS1 individuals are compound heterozygous for different NFU1 variants, it is valuable to investigate individual variants in an isogenic background. In order to understand the shared and unique phenotypes of NFU1 variants, we used CRISPR/Cas9 gene editing to recreate exact patient variants of NFU1 in the orthologous gene, nfu-1 (formerly lpd-8), in C. elegans. Five mutant C. elegans alleles focused on the presumptive iron-sulfur cluster interaction domain were generated and analyzed for mitochondrial phenotypes including respiratory dysfunction and oxidative stress. Phenotypes were variable between the mutant nfu-1 alleles and generally presented as an allelic series indicating that not all variants have lost complete function. Furthermore, reactive iron within mitochondria was evident in some, but not all, nfu-1 mutants indicating that iron dyshomeostasis may contribute to disease pathogenesis in some MMDS1 individuals. 相似文献
Capsule: Playback with dual-sex calls increases the response rate of Manx Shearwaters Puffinus puffinus by 40–60% and reduces its daily variability, which would improve the precision of population estimates.
Aims: To test whether playing male and female calls would elicit more responses to playback than male-only calls, reduce daily variability and the length of response rate calibration trials, and give more precise estimates. We also measured response times to playback and assessed the reliability of visual signs of occupancy at burrow entrances.
Methods: Responses to four playback call-types (male-only and three variants of dual-sex calls) were compared in repeated trials at two colonies (Ramsey and Rum, UK) during May–June 2014.
Results: Dual-sex calls gave higher response rates with lower variance than male-only calls, because females frequently replied to female calls but rarely to male calls. In simulated 3–5-day calibration trials, response rates and correction factors were up to 50% more precise with dual-sex calls. Visual signs of burrow occupancy were unreliable.
Conclusion: Playback for Manx Shearwaters should use a 25 seconds recording of male and female calls intermixed, with 10 seconds listening time for delayed responses. Census-specific calibration trials are essential for accurate estimates of daily response rates. 相似文献
We studied the effect of ditch blocking on vegetation composition and water-table depths in a blanket peatland. Measurements were made for a period of four years (water tables) and five years (vegetation) in the inter-ditch areas of three experimental treatments: (i) open ditches, (ii) ditches blocked with closely-spaced dams and (iii) ditches partially infilled with peat and blocked with dams. It is often assumed that ditch blocking will lead to an increase in the abundance of Sphagnum and, potentially, a reduction in the abundance of sedges, particularly the cotton grasses. However, our data show no treatment effects on the abundance of either group. We did find an effect of time, with the abundance of both sedges and Sphagnum spp. varying significantly between some years. For the sedges there was no systematic change over time, while for the Sphagnum spp. abundance tended to increase through the study period. This systematic change was not related to a measure of the vigour of the sedges, although vigour was lower towards the end of the study compared to the beginning. Our vegetation data are consistent with our water-table data. As with plant type abundance, we did not find any statistically significant differences in water-table depths between treatments, both for annual averages and summer averages. We comment on why ditch blocking does not seem to have affected water tables and vegetation composition at our study site. 相似文献

Wilk‐Wo?niak ( 2017 , Diversity and Distributions, 23 , 113–117) published a letter in Diversity and Distributions debating our view that waterbirds are important vectors of alien species (C. Reynolds, N. A. F. Miranda & G. S. Cumming, 2015 Diversity and Distributions, 21 , 744–754; A. J. Green, 2016 Diversity and Distributions, 22 , 239–247) and question whether future research into the mechanisms under‐pinning this phenomenon can be advantageous for the practical management of alien species. Additionally, Solarz et al. suggest that human activities are the primary source of all alien species introductions and that waterbirds may only act as vectors of secondary dispersal. In this letter, we respond to several arguments raised by the authors surrounding the relevance of waterbird‐mediated dispersal in the introduction and spread of alien species. We emphasize the partly deterministic nature of waterbird dispersal and the significance of long‐distance dispersal events (and hence the potential for primary introductions of new alien species across political boundaries). Finally, we reaffirm the importance of further research into dispersal by birds to improve our capacity to foresee and manage invasions of those alien species with strong capacity to spread via avian vectors. 相似文献

Pulmonate land snails often are able to estivate to survive dry hot seasons were water and food are scarce. The aperture of the shell is closed with an epiphragm, and metabolism is depressed to approximately one fourth of basal metabolism. We investigated a molecular aspect of estivation focussing on the heat shock protein 70 (Hsp70) stress response during estivation in the Mediterranean Grunt Snail Cantareus apertus. Sequences of a new inducible hsp70 and of actin are presented and expression of the hsp70 gene as well as Hsp70 protein content was measured in estivating animals. Both Hsp70 protein and mRNA do not show a significant change from the control, although there is a trend that hsp70 mRNA is less abundant in estivating specimens. After heat shock, the expression of hsp70 increased and a higher Hsp70 protein content was detected. Water relations were also investigated. After a period of 6 months in the dormant state, the snails contained 14% less water than active ones, implying a constricted protection against desiccation, compared to the desert snail Sphincterochila zonata, and a Mediterranean-type water economy. 相似文献
The malaria agent Plasmodium falciparum is predicted to export a "secretome" of several hundred proteins to remodel the host erythrocyte. Prediction of protein export is based on the presence of an ER-type signal sequence and a downstream Host-Targeting (HT) motif (which is similar to, but distinct from, the closely related Plasmodium Export Element [PEXEL]). Previous attempts to determine the entire secretome, using either the HT-motif or the PEXEL, have yielded large sets of proteins, which have not been comprehensively tested. We present here an expanded secretome that is optimized for both P. falciparum signal sequences and the HT-motif. From the most conservative of these three secretome predictions, we identify 11 proteins that are preserved across human- and rodent-infecting Plasmodium species. The conservation of these proteins likely indicates that they perform important functions in the interaction with and remodeling of the host erythrocyte important for all Plasmodium parasites. Using the piggyBac transposition system, we validate their export and find a positive prediction rate of approximately 70%. Even for proteins identified by all secretomes, the positive prediction rate is not likely to exceed approximately 75%. Attempted deletions of the genes encoding the conserved exported proteins were not successful, but additional functional analyses revealed the first conserved secretome function. This gave new insight into mechanisms for the assembly of the parasite-induced tubovesicular network needed for import of nutrients into the infected erythrocyte. Thus, genomic screens combined with functional assays provide unexpected and fundamental insights into host remodeling by this major human pathogen. 相似文献
We have previously shown that targeting human CD34(+) hematopoietic stem cells (HSC) with a bispecific antibody (BiAb) directed against myosin light chain (MLC) increases delivery of cells to the injured hearts and improves cardiac performance in the nude rat. In this study, we have sought to validate our previous observations and to perform more detailed determination of ventricular function in immunocompetent mice with myocardial infarction (MI) that were treated with armed CD34(+) HSC. We examined whether armed CD34(+) HSC would target the injured heart following MI and restore ventricular function in vitro. MI was created by ligation of the left anterior descending artery. After 48 h, adult ICR mice received either 0.5 x 10(6) human CD34(+) HSC armed with anti-CD45 x anti-MLC BiAb or an equal volume of medium through a single tail vein injection. Two weeks after stem cell administration, ventricular function of hearts from mice receiving armed CD34(+) HSC was significantly greater compared with the same parameters from control mice. Immunohistochemistry confirmed the accumulation of CD34(+) HSC in MI hearts infused with stem cells. Angiogenesis was significantly enhanced in CD34(+) HSC-treated heart as determined by vascular density per area. Furthermore, histopathological examination revealed that the retained cardiac function observed in CD34(+) HSC-treated mice was associated with decreased ventricular fibrosis. These results suggest that peripheral administration of armed CD34(+) HSC results in localization of CD34(+) HSC to injured myocardium and restores myocardial function. 相似文献
Reactive oxygen species (ROS) are natural products inevitably generated along cellular metabolism. Due to their highly reactive nature, which can damage DNA, proteins and lipids, cells utilize antioxidative or defense systems to balance these toxic products to keep the cells in a state of redox homeostasis. However, under the situation of imbalance in redox status, depending on the magnitude of ROS encountered, high levels of ROS can induce apoptosis, whereas chronic low levels of ROS promote vascular diseases such as arteriosclerosis. Although ROS seem to be catastrophic to life, accumulating evidence points to the beneficial roles of ROS by virtue of the ability as chemotherapeutic agents to cure human diseases. Many anti-cancer drugs have been developed in this way which can generate ROS and cause oxidative stress-induced apoptosis in cancer cells. The effects of ROS are paradoxical because they can act as both disease culprits and chemotherapeutic agents. In this review, the current knowledge of ROS and the potential applications of ROS in cancer therapeutic will be discussed. 相似文献
