Scramblases are a family of single-pass plasma membrane proteins, identified by their purported ability to scramble phospholipids across the two layers of plasma membrane isolated from platelets and red blood cells. However, their true in vivo role has yet to be elucidated. We report the generation and isolation of null mutants of two Scramblases identified in Drosophila melanogaster. We demonstrate that flies lacking either or both of these Scramblases are not compromised in vivo in processes requiring scrambling of phospholipids. Instead, we show that D. melanogaster lacking both Scramblases have more vesicles and display enhanced recruitment from a reserve pool of vesicles and increased neurotransmitter secretion at the larval neuromuscular synapses. These defects are corrected by the introduction of a genomic copy of the Scramb 1 gene. The lack of phenotypes related to failure of scrambling and the neurophysiological analysis lead us to propose that Scramblases play a modulatory role in the process of neurotransmission. 相似文献

Several indole derivatives, that were highly potent ligands of GluN2B-subunit-containing N-methyl-d-aspartate (NMDA) receptor, also demonstrated antioxidant properties in ABTS method. In particular, the 2-(4-benzylpiperidin-1-yl)-1-(5-hydroxy-1H-indol-3-yl)ethanone (1) proved to be a dual-effective neuroprotective agent. With the aim to increase the antioxidant properties we added a catechol moiety onto piperidine moiety. The designed hybrid derivative 3,4-dihydroxy-N-[1-[2-(5-hydroxy-1H-indol-3-yl)-2-oxoethyl]piperidin-4-yl]benzamide (10) was the most effective antioxidant agent (>94.1 ± 0.1% of inhibition at 17 μM) and showed GluN2B/NMDA receptor affinity at low micromolar concentration (IC50 0.66 μM). Entomopathogenic nematodes (EPNs) have been utilized in classical, conservation, and augmentative biological control programs. The vast majority of applied research has focused on their potential as inundatively applied augmentative biological control agents. Extensive research over the past three decades has demonstrated both their successes and failures for control of insect pests of crops, ornamental plants, trees and lawn and turf. In this paper we present highlights of their development for control of insect pests above and below ground. The target insects include those from foliar, soil surface, cryptic and subterranean habitats. Advances in mass-production and formulation technology of EPNs, the discovery of numerous efficacious isolates/strains, and the desirability of reducing pesticide usage have resulted in a surge of commercial use and development of EPNs. Commercially produced EPNs are currently in use for control of scarab larvae in lawns and turf, fungus gnats in mushroom production, invasive mole crickets in lawn and turf, black vine weevil in nursery plants, and Diaprepes root weevil in citrus in addition to other pest insects. However, demonstrated successful control of several other insects, often has not lead to capture of a significant share of the pesticide market for these pests. 相似文献

The internal vestibule of large-conductance Ca(2+) voltage-activated K(+) (BK) channels contains a ring of eight negative charges not present in K(+) channels of lower conductance (Glu386 and Glu389 in hSlo) that modulates channel conductance through an electrostatic mechanism (Brelidze, T.I., X. Niu, and K.L. Magleby. 2003. Proc. Natl. Acad. Sci. USA. 100:9017-9022). In BK channels there are also two acidic amino acid residues in an extracellular loop (Asp326 and Glu329 in hSlo). To determine the electrostatic influence of these charges on channel conductance, we expressed wild-type BK channels and mutants E386N/E389N, D326N, E329Q, and D326N/E329Q channels on Xenopus laevis oocytes, and measured the expressed currents under patch clamp. Contribution of E329 to the conductance is negligible and single channel conductance of D326N/E329Q channels measured at 0 mV in symmetrical 110 mM K(+) was 18% lower than the control. Current-voltage curves displayed weak outward rectification for D326N and the double mutant. The conductance differences between the mutants and wild-type BK were caused by an electrostatic effect since they were enhanced at low K(+) (30 mM) and vanished at high K(+) (1 M K(+)). We determine the electrostatic potential change, Deltaphi, caused by the charge neutralization using TEA(+) block for the extracellular charges and Ba(2+) for intracellular charges. We measured 13 +/- 2 mV for Deltaphi at the TEA(+) site when turning off the extracellular charges, and 17 +/- 2 mV for the Deltaphi at the Ba(2+) site when the intracellular charges were turned off. To understand the electrostatic effect of charge neutralizations, we determined Deltaphi using a BK channel molecular model embedded in a lipid bilayer and solving the Poisson-Boltzmann equation. The model explains the experimental results adequately and, in particular, gives an economical explanation to the differential effect on the conductance of the neutralization of charges D326 and E329. 相似文献

Paratuberculosis was diagnosed in a population of approximately 1,000 free-ranging fallow deer (Dama dama) sampled from 1997-98 in the Regional Hunting Reserve of El Sueve (Asturias, Spain). Five of eight animals observed with diarrhea were diagnosed as having paratuberculosis on the basis of gross lesions at postmortem examination and histopathology. In two deer, Mycobacterium avium subsp. paratuberculosis was cultured and identified by polymerase chain reaction. Indirect enzyme-linked immunosorbent assay and immunodiffusion tests were used to evaluate sera from 33 adult deer from this population. All fallow deer tested were seronegative. 相似文献

We characterized a gene encoding an YchF-related protein, TcYchF, potentially associated with the protein translation machinery of Trypanosoma cruzi. YchF belongs to the translation factor-related (TRAFAC) class of P-loop NTPases. The coding region of the gene is 1185 bp long and encodes a 44.3 kDa protein. BlastX searches showed TcYchF to be very similar (45-86%) to putative GTP-binding proteins from eukaryotes, including some species of trypanosomatids (Leishmania major and Trypanosoma brucei). A lower but significant level of similarity (38-43%) was also found between the predicted sequences of TcYchF and bacterial YyaF/YchF GTPases of the Spo0B-associated GTP-binding protein (Obg) family. Some of the most important features of the G domain of this family of GTPases are conserved in TcYchF. However, we found that TcYchF preferentially hydrolyzed ATP rather than GTP. The function of YyaF/YchF is unknown, but other members of the Obg family are known to be associated with ribosomal subunits. Immunoblots of the polysome fraction from sucrose gradients showed that TcYchF was associated with ribosomal subunits and polysomes. Immunoprecipitation assays showed that TcYchF was also associated with the proteasome of T. cruzi. Furthermore, inactivation of the T. brucei homolog of TcYchF by RNA interference inhibited the growth of procyclic forms of the parasite. These data suggest that this protein plays an important role in the translation machinery of trypanosomes. 相似文献

This study was carried out to evaluate maize plants of different recurrent selection cycles of the variety (Zea mays L.—Saracura-BRS-4154) regarded to genetic gains, morphophysiology, and grain yield, achieved over the selection cycles under
intermittent flooding of the soil. This variety has the capacity to survive and produce in temporarily flooded soils and was
developed by the National Maize and Sorghum Research Center (EMBRAPA). The experiment was conducted in greenhouse by using
ten alternating selection cycles (Cycle 1–18) and BR 107 a variety known for its susceptibility to flooding. The flooding
initiated at six-leaf stage by applying 50 mm of water three times a week. At flowering, the following parameters were evaluated:
rate of leaf photosynthesis, stomatal conductance, intercellular CO2 concentration, transpiration rate, photosynthetically active leaf area, root porosity, relative chlorophyll content, grain
yield, harvest index, ear length, and interval between male and female flowering. Yield as a function of root porosity and
photosynthesis were also evaluated. An index was created in this study, in order to help the discussion of the characteristics
evaluated, it was called “Relative Tolerance Value—RTV”, only gaseous exchange measurements was not included in this index.
By the way, it was observed throughout the selection cycles an increase in all gaseous exchange parameters, being the cycle
18, the one which presented the greatest averages. RTV for leaf area showed the greatest values for cycles 7 and 18, whereas
root porosity, chlorophyll relative content, and harvest index, the greater RTV values were found in cycles 17 and 18. The
largest grain yield RTV was observed in cycle 7, followed by cycles 13, 15, and 18. Flooding resulted in longer Anthesis-Silking
Interval, especially for the first cycles. At flooding condition, grain yield was strongly related to root porosity (R
2 = 0.66). These results showed that the selection cycles of “Saracura” maize improved some morphophysiologic characters, which
favor their survival in flooded environments, also resulting in higher productivity. 相似文献

Papaya (Carica papaya) is a very important crop in many tropical countries but it is highly susceptible to parasitic diseases, physiological disorders,
mechanical damage and fruit overripening. Here we report a study on ACC oxidase cosuppression and its effects on papaya fruit
ripening. Papaya ACC oxidase was isolated using PCR and embriogenic cells transformed by biolistic using the CaMV 35S promoter
to drive the expression of the PCR fragment in sense orientation. Fifty transgenic lines were recovered and 20 of those were
grown under field conditions. Southern analysis showed incorporation of the transgene in different copy numbers in the papaya
genome. Fruits were evaluated in terms of texture (firmness), colour development, respiration and ethylene production. A sharp
reduction in ethylene and CO2 production was detected, whereas softening and colour development of the peel were also altered.
Overall, transgenic fruits showed a delay in ripening rate. A reduction in mRNA level for ACC oxidase in transgenic fruit
was clearly detectable by northern blot. More studies are necessary before this technology can be used to extend the shelf

Carbon isotope discrimination (Δ13C) in charred grains from archaeological sites provides reliable information about water availability of ancient crops. However,
as cereals are cultivated plants, they may reflect not only climatic fluctuations, but also the effect on water status of
certain agronomic practices, such as sowing in naturally wet soils or irrigation. In this work, we propose a methodological
approach to combine Δ13C data from different plant species, in order to discriminate between climate-derived and anthropogenic effects on ancient
crops. We updated previous models for estimating water inputs from Δ13C of cereal grains of Hordeum vulgare and Triticum aestivum/durum, and we applied them to published data from several archaeological sites, including samples from the Neolithic to the present
day in northeast and southeast Spain, as well as from the Neolithic site of Tell Halula (northwest Syria). We found an important
decrease in water availability from the Neolithic to the present time in the three areas of study, especially clear for the
two driest areas (southeast Spain and northwest Syria). Potential differences in water management practices between wheat
and barley, as well as between cereal and legume crops (Vicia faba and Lens culinaris), are also discussed on the basis of the comparison of Δ13C values across several archaeological sites. 相似文献
