Background
There are four widely used experimental strains of N. gonorrhoeae, one of which has been sequenced and used as the basis for the construction of a multi-strain, mutli-species pan-neisserial microarray. Although the N. gonorrhoeae population structure is thought to be less diverse than N. meningitidis, there are some recognized gene-complement differences between strains, including the 59 genes of the Gonococcal Genetic Island. Anoxic conditions should hamper the transport of sugar in the phloem, as this is an active process. The canopy is a carbohydrate source and the roots are carbohydrate sinks. By fumigating the shoot with N2 or flooding the rhizosphere, anoxic conditions in the source or sink, respectively, were induced. Volume flow, velocity, conducting area and stationary water of the phloem were assessed by non‐invasive magnetic resonance imaging (MRI) flowmetry. Carbohydrates and δ13C in leaves, roots and phloem saps were determined. Following flooding, volume flow and conducting area of the phloem declined and sugar concentrations in leaves and in phloem saps slightly increased. Oligosaccharides appeared in phloem saps and after 3 d, carbon transport was reduced to 77%. Additionally, the xylem flow declined and showed finally no daily rhythm. Anoxia of the shoot resulted within minutes in a reduction of volume flow, conductive area and sucrose in the phloem sap decreased. Sugar transport dropped to below 40% by the end of the N2 treatment. However, volume flow and phloem sap sugar tended to recover during the N2 treatment. Both anoxia treatments hampered sugar transport. The flow velocity remained about constant, although phloem sap sugar concentration changed during treatments. There are several differences associated with the behaviour of the four main experimental Neisseria gonorrhoeae strains, FA1090, FA19, MS11, and F62. Although there is data concerning the gene complements of these strains, the reasons for the behavioural differences are currently unknown. Phase variation is a mechanism that occurs commonly within the Neisseria spp. and leads to switching of genes ON and OFF. Background
Streptococcus agalactiae (Group B Streptococcus; GBS) is a major contributor to obstetric and neonatal bacterial sepsis. Serotype III strains cause the majority of late-onset sepsis and meningitis in babies, and thus appear to have an enhanced invasive capacity compared with the other serotypes that cause disease predominantly in immunocompromised pregnant women. We compared the serotype III and V whole genome sequences, strains NEM316 and 2603VR respectively, in an attempt to identify genetic attributes of strain NEM316 that might explain the propensity of strain NEM316 to cause late-onset disease in babies. Fourteen putative pathogenicity islands were described in the strain NEM316 whole genome sequence. Using PCR- and targeted microarray- strategies, the presence of these islands were assessed in a diverse strain collection including 18 colonizing isolates from healthy pregnant women, and 13 and 8 invasive isolates from infants with early- and late-onset sepsis, respectively. 相似文献67.

The aim of this work was to estimate the trypsin inhibitor activity (TIA) in seeds of cultivated pea varieties, wild varieties and selected crosses between varieties from the first group and wild pea varieties and to study the variation in genes coding trypsin inhibitors. Mean TIA in field pea varieties ranged from 3.12 TIU/mg of sample in field pea variety from Czech Republic to 12.90 TIU/mg of sample in field pea variety FP S4 of Serbian origin. Wild field pea varieties showed TIA between 0.98 TIU/mg of sample in Pisum elatius and 9.79 TIU/mg of sample in Pisum abyssinicum. Selected crosses between cultivated field pea varieties and Pisum elatius showed a decrease in TIA in comparison with a parent line that has higher TIA content. The PCR amplification resulted in variety-specific amplification. Varieties with low TIA activity showed amplification with At13/At5 primer pair, while varieties with higher TIA activity showed amplification with primer pairs At12/At5, At14/At5 and At14/At8. Thus, At13/At5 primer pair could be sufficient to distinguish most varieties. These markers can be applied during an early screening of the valuable materials for future breeding programs of pea cultivars with the low level of tripsin inhibitor. 相似文献

Purpose: Vegetation plays a very important role in industrial regions in terms of not only air purification, but also oxygen enrichment, air humidity, and city aesthetics. The paper presents the authors’ study of oleaster mildew during 2012–2013 in the cities of Pavlodar, Ekibastuz, and Aksu. Method: The species composition of mildew, patterns of its growth and reproduction, seasonal dynamics, and the level of trees infection according to a six-score scale were determined. Result: Three cultivars of mildew from three genera were registered. The species composition of mildew of oleaster is studied for the first time in large industrial cities of the Pavlodar region. The comparative analysis of the systematic structure of phytopathogenic fungi was followed by the study of the micro flora of green planting in megalopolises. The seasonal dynamics of growth, development, and reproduction was determined. The impact of pests on host-plants was studied. Conclusion: Timely diagnosis and localization of disease outbreak increases the likelihood of successful treatment and saving of plants. 相似文献

Manchego cheese is a high-fat pressed ewe's-milk cheese made in Castilla-La Mancha (Spain) and produced by enzymatic coagulation. The minimum ripening time before marketing required by the Regulatory Board of the Manchego Cheese Appellation of Origin is 60 days.
This paper describes the physicochemical, proteolysis, sensory and texture characteristics of Manchego cheese, and the degree of homogeneity of cheeses made under the Manchego Appellation of Origin. The data gathered in this study indicate that sensory and instrumental analysis are useful tools for detecting changes in Manchego cheese during ripening. These changes were first detected by the instrumental analysis (2 months). The panelists detected differences after 4 months' ripening in all the factories.
