Changes in fatty acid, phospholipid and galactolipid contents during cellular and organ differentiation in Aegle marmelos have been described. Decrease in phosphatidylinositol content and presence of 3-trans-hexadecenoic acid in phosphatidylglycerol were related to greening and shoot buds differentiation. The galactolipids level, the monogalactosyl diglyceride/digalactosyl diglyceride ratio and the linolenic acid level (mainly in monogalactosyl diglyceride) increased with the degree of differentiation, indicating the possible biogenesis of functional chloroplasts.Abbreviations 2,4-D

This report demonstrates the elicitation effect on growth and stilbene accumulation in cell cultures of Cayratia trifolia (Vitaceae) by an extract of the angiosperm parasite Cuscuta reflexa and salicylic acid in combination with sucrose feeding. Cell cultures of C. trifolia, a tropical liana, were maintained in liquid Murashige and Skoog's basal medium containing 0.25 mg l−1 naphthalene acetic acid, 0.2 mg l−1 kinetin with 3% sucrose and 250 mg l−1 casein hydrolysate. The cells treated with Cuscuta elicitor showed increased polyphenol oxidase activity with increasing concentration of the elicitor, while total phenol content
remained almost unchanged. Enhanced yield of stilbenes (∼8-fold) was recorded in the cells treated with 200 mg l−1
Cuscuta elicitor for 7 d. Optimum accumulation of stilbenes with a non-significant decrease in cell growth as compared with control
was recorded with the addition of 3% sucrose on the seventh day of cell culture. Addition of 3% sucrose with salicylic acid
at 500 μM and Cuscuta extract at 200 mg l−1 on the seventh day enhanced total stilbene yield up to 50.1 mg l−1, which was ∼14-fold higher than in control cultures. Piceid content increased ∼200-fold in such cultures. 相似文献

Standardization of biomass production in different vessels and bioreactor using explants and media for growth, total phenolic content and antioxidant capacity of shoot culture of Bacopa monnieri is described. Maximum number of shoots per explant, higher explants response irrespective of the type of explants, and higher shoot length was obtained on MS medium containing BAP (2.5 mg l−1) and IAA (0.01 mg l−1) with 3 % sucrose. This medium was selected by varying BAP concentration and recorded optimal for shoot culture on gelled medium. The condition of 0.5 cm explant size and 20 explant/40 ml (1 explant/2 ml) was optimal for high explant response, number of shoots per explant regenerated and shoots length. Among the different vessels used, maximum growth index was achieved in Growtek bioreactor (10.0) followed by magenta box (9.16), industrial glass jar (7.7) and conical flask (7.2). The cultures grown in conical flask (100 ml) were used as control. The total phenolic content and antioxidant capacity of in vitro grown plants was higher to that recorded for in vivo material. Among in vitro regenerated plants, the activity was maximal in the tissues grown in 250 ml conical flask. The most critical function for vessels is to support the optimum profusion (growing area for maximum growth) of shoots and for B. monnieri, Growtek bioreactor supported 1980 shoots l−1 medium as compared to control (938 shoots l−1). Growtek bioreactor was considered effective system to produce B. monnieri biomass in culture without loss of antioxidant properties. 相似文献

Plant gum as an elicitor for guggulsterone production in cell cultures of Commiphora wightii is reported for the first time. Guggulsterone production increased 2.4 fold in the cell cultures by gum Arabic (100 mg l−1), while mesquite gum elicited 2 fold. The cells treated with gum Arabic at 7th and 9th day accumulated enhanced guggulsterones
within 24 h, which increased further up to 48 h and then declined. The cells treated at 9th day accumulated higher amount
(218 μg l−1) of guggulsterones after 48 h of elicitation as compared to cells treated at 7th day (164 μg l−1). The optimized elicitation conditions were used in vessels of varying capacity where maximum yield of 285 μg l−1 of guggulsterones was recorded in 3 l shake flasks. These experiments enabled highest guggulsterones yield in a short duration

AbstractAbiotic stress imposed by many factors such as: extreme water regimes, adverse temperatures, salinity, and heavy metal contamination result in severe crop yield losses worldwide. Plants must be able to quickly respond to these stresses in order to adapt to their growing conditions and minimize metabolic losses. In this context, transporter proteins play a vital role in regulating stress response mechanisms by facilitating movement of a variety of molecules and ions across the plasma membrane in order to maintain fundamental cellular processes such as ion homeostasis, osmotic adjustment, signal transduction, and detoxification. Aquaporins play a crucial role in alleviating abiotic stress by transporting water and other small molecules to maintain cellular homeostasis. Similarly, other transporter families such as CDF, ZIP, ABC, NHX, HKT, SWEETs, TMTs, and ion channels also contribute to abiotic stress tolerance. Hormones and other signaling molecules are necessary to coordinate responses across different tissues and to precisely regulate molecular trafficking. The present review highlights the current understanding of how membrane transporters orchestrate stress responses in plants. It also provides insights about the importance of these sensing and adaptive mechanisms for ensuring improved sustainable crop production during unfavorable conditions. Finally, this review discusses future prospects for the use of computational tools in constructing signaling networks to improve our understanding of the behavior of transporters under abiotic stress. 相似文献

Guggulsterone, a hypolipidemic natural agent, is produced in resin canals of the plant Commiphora wightii. In this study, the efficacy of different plant growth regulators was evaluated for optimizing its production. Morphactin
was found to be effective in enhancing the accumulation of guggulsterones in callus cultures. Maximum callus growth was recorded
on medium containing morphactin (0.1 mg l−1) and 2iP (2.5 mg l−1), whereas maximum guggulsterone production occurred when the calluses were cultured on medium containing 0.1 mg l−1 morphactin and 1.0 mg l−1 2iP. Morphactin and 2iP interacted significantly to enhance the callus growth and guggulsterone production by about 8-folds
in one-year-old cultures. However, the effect of morphactin on callus growth and guggulsterone production was not uniform
over the levels of 2iP tested. Such an effect of morphactin has never been reported on the production of secondary metabolites. 相似文献

Isoflavonoid production in cell cultures of Pueraria tuberosa as influenced by an angiospermic parasite, Cuscuta reflexa, was studied. During the time course, maximum isoflavonoid content was recorded when Cuscuta elicitor was added on day 15 of culture. Among various concentrations of elicitor tried, 1 g l−1 of Cuscuta elicitor was found to be the most effective. The optimized elicitation conditions were used in vessels of varying capacity
where maximum yield of ~91 mg l−1 of isoflavonoid was recorded in a 2-l bioreactor which was about 19% higher than the control cultures. In this case, puerarin
content increased up to 11 mg l−1 which was 580% higher that the value recorded in the control cultures. In the bioreactor, 8 days of elicitation was optimal
for the high accumulation of isoflavonoid, giving productivity of ~4 mg l−1 day−1. The study showed persistent high isoflavonoid yield even during scale-up. Use of a preparation of Cuscuta reflexa as an elicitor is reported for the first time. The increase in isoflavonoid content was elicitor dose-dependent and can be
explored to trigger high yields of isoflavonoid/secondary metabolites in production. 相似文献

Cell cultures of Commiphora wightii (Arnott.) Bhandari were grown in shake flasks and a bioreactor and an increase in guggulsterone accumulation up to 18 μg
l−1 was recorded in cells grown in the production medium containing a combination of sucrose:glucose (4% total), precursors (phenylalanine,
pyruvic acid, xylose, and sodium acetate), morphactin, and 2iP. A yield of 10 g l−1 biomass and ∼200 μg l−1 guggulsterone was recorded in a 3-l flask and in a 2-l stirred tank bioreactor compared with 6.6 g biomass and 67 μg l−1 guggulsterone in 250-ml flasks. Increased vessel size was correlated with increased biomass and guggulsterone accumulation.
2iP alone was not effective for biomass and guggulsterone accumulation in cell cultures of C. wightii. 相似文献
