Effect of low density particles on the apparent liquid circulation velocity and overall gas holdup was studied in a modified reversed flow jet loop bioreactor. Experiments were conducted using polyurethane beads, polystyrene particles which are comparable to bioparticles found in biological applications and glass beads. Influence of gas and liquid flow rates, draft tube to reactor diameter ratio and solids loading on these hydrodynamic properties were studied. The liquid circulation velocity was found to increase with an increase in liquid flow rate but decrease with an increase in gas flow rate or solids loading. The overall gas holdup increased with an increase in gas or liquid flow rate but decreased with an increase in solids loading. The range of optimum draft tube to reactor diameter ratio was found to be 04–0.5. The results obtained with low density particles were comparatively better than those with glass beads. Correlations were proposed to evaluate liquid circulation velocity and overall gas holdup in terms of operational and geometrical variables.  相似文献

Mixing characteristics in the downcomer and the riser of a continuous down-flow jet loop bioreactor was studied with Newtonian and non-Newtonian fluids. The mixing parameters were determined through the curve fitting of the experimental impulse response data with the solution of one dimensional axial dispersion model. It was found that circulation number and axial dispersion coefficient increased with an increase in liquid flow rate and draft tube to column diameter ratio and the axial dispersion coefficient was comparatively higher in the riser. The circulation number increased with decrease in nozzle diameter. The model predicted the experimental data well within 8% deviation for both the systems (water and CMC). Correlations were obtained to predict axial dispersion coefficients in the riser and downcomer of the reactor.  相似文献

Proteins structurally related to eukaryotic actins have recently been identified in several prokaryotic organisms. These actin-like proteins (MreB and ParM) and the deviant Walker A ATPase (SopA) play a key role in DNA segregation and assemble into polymers in vitro and in vivo. MreB also plays a role in cellular morphogenesis. Whereas the dynamic properties of eukaryotic actins have been extensively characterized, those of bacterial actins are only beginning to emerge. We have established the fission yeast Schizosaccharomyces pombe as a cellular model for the functional analysis of the Escherichia coli actin-related protein MreB. We show that MreB organizes into linear bundles that grow in a symmetrically bidirectional manner at 0.46 +/- 0.03 microm/min, with new monomers and/or oligomers being added along the entire length of the bundle. Organization of linear arrays was dependent on the ATPase activity of MreB, and their alignment along the cellular long axis was achieved by sliding along the cortex of the cylindrical part of the cell. The cell ends appeared to provide a physical barrier for bundle elongation. These experiments provide new insights into the mechanism of assembly and organization of the bacterial actin cytoskeleton.  相似文献

A survey of the pressure resistance of aquatic animals in different stages of their life cycle shows that adults generally are more tolerant of pressure than the egg and nauplii, but older adults appear less pressure resistant than younger adults. Data on many species of aquatic animals of different size shows no correlation between size and pressure resistance. It is concluded that size is not a special determinant in the successful deep-sea colonization of shallow-water animals and this is consonant with the fact of occasional large deep-sea species whereas the average size is quite small in comparison with littoral species.  相似文献

Epidemiological studies have shown an inverse correlation between the incidence of lymphatic filariasis (LF) and the incidence of allergies and autoimmunity. However, the interrelationship between LF and type-2 diabetes is not known and hence, a cross sectional study to assess the baseline prevalence and the correlates of sero-positivity of LF among diabetic subjects was carried out (n = 1416) as part of the CURES study. There was a significant decrease in the prevalence of LF among diabetic subjects (both newly diagnosed [5.7%] and those under treatment [4.3%]) compared to pre-diabetic subjects [9.1%] (p = 0.0095) and non-diabetic subjects [10.4%] (p = 0.0463). A significant decrease in filarial antigen load (p = 0.04) was also seen among diabetic subjects. Serum cytokine levels of the pro-inflammatory cytokines—IL-6 and GM-CSF—were significantly lower in diabetic subjects who were LF positive, compared to those who were LF negative. There were, however, no significant differences in the levels of anti-inflammatory cytokines—IL-10, IL-13 and TGF-β—between the two groups. Although a direct causal link has yet to be shown, there appears to be a striking inverse relationship between the prevalence of LF and diabetes, which is reflected by a diminished pro-inflammatory cytokine response in Asian Indians with diabetes and concomitant LF.  相似文献

A laboratory scale Upflow Anaerobic Sludge Blanket (UASB) reactor study was done with spentwash (distillery effluent) as substrate. The start-up studies revealed that the recirculation of biogas, produced during the initial stages, under low organic loading rates helps in mixing as well as partial supplementation of carbon dioxide for excess hydrogen in the reactor. The gas composition before and after recirculation, the volatile acids profile and the solids variation along the reactor height before and after recirculation were studied. The process efficiency and methane content increases substantially after recirculation of biogas and the microscopic examination of the sludge during the experiment indicates that the granules mainly composed of long multicellular filaments of rod shaped organisms, presumably methanothrix soehngenii.  相似文献

Seedlings of green gram (Vigna radiata cv. ADT-1 and CO-5) were exposed to daily showers of simulated acidic rain (H2SO4 : HNO3 : HCl, 4 : 2 : 1, v/v) for 10 d. The effects were analysed after 5 and 10 showers, respectively. Rain of pH 2.5 inhibited seedling growth and biomass accumulation, though in other acidic levels the effects were mostly inconsistent. Both cultivars had high degree of surface wettability indicated by high leaf surface contact angles and water-holding capacity. Treated leaves were thinner with smaller mesophyll cells. Stomatal index and trichome density were lower in contrast to epidermal cell density and stomatal frequency which increased with increasing acidity. Decreases in chlorophyll (Chl), carotenoid (Car), and starch contents in cv. ADT-1 at pH 2.5 were observed after 5 showers, while in cv. CO-5 decreases were noted only after 10 showers. In contrast to total sugar levels, the protein content of cv. CO-5 was augmented significantly after simulated acidic rain (SAR) treatment.  相似文献

Renal infiltration with mononuclear cells is associated with poor prognosis in systemic lupus erythematosus. A renal macrophage/dendritic cell signature is associated with the onset of nephritis in NZB/W mice, and immune-modulating therapies can reverse this signature and the associated renal damage despite ongoing immune complex deposition. In nephritic NZB/W mice, renal F4/80(hi)/CD11c(int) macrophages are located throughout the interstitium, whereas F4/80(lo)/CD11c(hi) dendritic cells accumulate in perivascular lymphoid aggregates. We show here that F4/80(hi)/CD11c(int) renal macrophages have a Gr1(lo)/Ly6C(lo)/VLA4(lo)/MHCII(hi)/CD43(lo)/CD62L(lo) phenotype different from that described for inflammatory macrophages. At nephritis onset, F4/80(hi)/CD11c(int) cells upregulate cell surface CD11b, acquire cathepsin and matrix metalloproteinase activity, and accumulate large numbers of autophagocytic vacuoles; these changes reverse after the induction of remission. Latex bead labeling of peripheral blood Gr1(lo) monocytes indicates that these are the source of F4/80(hi)/CD11c(int) macrophages. CD11c(hi)/MHCII(lo) dendritic cells are found in the kidneys only after proteinuria onset, turnover rapidly, and disappear rapidly after remission induction. Gene expression profiling of the F4/80(hi)/CD11c(int) population displays increased expression of proinflammatory, regulatory, and tissue repair/degradation-associated genes at nephritis onset that reverses with remission induction. Our findings suggest that mononuclear phagocytes with an aberrant activation profile contribute to tissue damage in lupus nephritis by mediating both local inflammation and excessive tissue remodeling.  相似文献

Eukaryotic F-actin is constructed from two protofilaments that gently wind around each other to form a helical polymer. Several bacterial actin-like proteins (Alps) are also known to form F-actin-like helical arrangements from two protofilaments, yet with varied helical geometries. Here, we report a unique filament architecture of Alp12 from Clostridium tetani that is constructed from four protofilaments. Through fitting of an Alp12 monomer homology model into the electron microscopy data, the filament was determined to be constructed from two antiparallel strands, each composed of two parallel protofilaments. These four protofilaments form an open helical cylinder separated by a wide cleft. The molecular interactions within single protofilaments are similar to F-actin, yet interactions between protofilaments differ from those in F-actin. The filament structure and assembly and disassembly kinetics suggest Alp12 to be a dynamically unstable force-generating motor involved in segregating the pE88 plasmid, which encodes the lethal tetanus toxin, and thus a potential target for drug design. Alp12 can be repeatedly cycled between states of polymerization and dissociation, making it a novel candidate for incorporation into fuel-propelled nanobiopolymer machines.  相似文献

Phenolic compound wastes from a large number of industries big and small which are highly toxic and pose a direct threat to human and aquatic life are generally let into the rivers and coastal waters. 2,4-dichlorophenol is used in the manufacture of industrial and agricultural products such as pesticides, germicides, soil sterilants, seed disinfectants and antiseptics. A modified Rotating Biological Contactor (RBC) was used for the treatability studies of synthetic 2,4-di-chlorophenolic (2,4 CP) wastewaters. The RBC used was a four stage laboratory model and the discs were modified by attaching porous netlon sheets to enhance biofilm area and volume. Synthetic wastewaters were prepared with influent concentrations from 40 to 200?mg/l of 2,4 CP. Four hydraulic loads were used in the range of 0.024 to 0.065 m3.m-2.d-1 and the organic loads used were in the range of 2 to 13?g 2,4 CP.m-2.d-1. The RBC was operated at a speed of 12?rpm. Effect of hydraulic loadings and influent 2,4-dichlorophenol concentration on 2,4-dichlorophenol removal were discussed and showed maximum organic removal at hydraulic loads of 0.024 and 0.046?m3.m-2.d-1. Also, a correlation plot between 2,4 CP applied and 2,4 CP removed was presented. A mathematical model was proposed using regression analysis.  相似文献
